Overproduction of Alkaloid Phytoalexins in California Poppy Cells Is Associated with the Co-Expression of Biosynthetic and Stress-Protective Enzymes

Many plant cells respond to pathogens by the induction of phytoalexin biosynthesis, but the underlying changes of gene expression are often obscured by their close linkage to the complex rearrangements during pathogen defense, especially the hypersensitive cell death. In root-derived cell cultures of Eschscholzia californica, the overproduction of cytotoxic benzophenanthridine alkaloids can be triggered by a minimum of pathogen pressure that does not evoke hypersensitive reactions. Such conditions activate a signal chain that is initiated by a short contact to low concentrations of yeast glycoprotein elicitor and includes a transient acidification of the cytoplasm. In contrast, high elicitor concentrations signal via an increase of jasmonate and trigger hypersensitive cell death, preceded by a drastic decay of translatable mRNAs. The main changes in protein and mRNA patterns caused by either signal path were compared by 2D proteomic separation, MS/MS sequencing and mRNA-in vitro translation. The four proteins showing the highest overexpression were identical between cells that received low or high-elicitor treatment and overlapped with the three proteins most upregulated by artificial pH shifts. They comprised one biosynthetic enzyme （norcoclaurine：SAM 4＇ O-methyl-transferase） plus a unique combination of stress-protective proteins： a heat shock protein （hsp 70）; a peptidyl-prolyl-cis/trans isomerase （cyclophilin）; and a glyceraldehyde-3-phosphate dehydrogenase. It appears that overproduction of the benzophenanthri- dine phytoalexins requires the up-regulation of a rate-limiting biosynthetic enzyme plus the coordinated expression of a specific set of protective enzymes and thus is managed like an oxidative stress.

A self-reported long-term follow-up of patients operated with either shortening techniques or a TachoSil grafting procedure

The aim of this article is to analyse the long-term results of different surgical techniques for correction of penile deviations in Peyronie＇s disease. Patients who underwent surgery for a penile deviation in Peyronie＇s disease between 1997 and 2007 were included into this study. Cases were retrospectively analysed by chart review. The current situation was evaluated by a 16-item standardized questionnaire addressing penile straightness, sensation, length, sexual function and satisfaction. Ninety patients were contacted with a return rate of 75 （83%） evaluabie questionnaires. Thirty-two patients were operated by shortening techniques （STs） with either Schroeder-Essed （n= 16） or Nesbit （n= 16）. Forty-three were operated by a plaque in-lexcision and defect covering by TachoSil method （TM）. Both groups were similar regarding age （ST 56 years, TM 57 years）, comorbidities, mean preoperative erection hardness score （EHS; 3.1 ST, 3.4 TM） and time of follow-up （total mean, 63 months）. Preoperatively ST patients had significantly less plaques （P〈0.05） and a lower deviation angle in comparison to TM patients （ST 56° versus TM 74°; P〈 0.001）. Still TM patients reported slightly better straightening results, but a significantly lower mean EHS （ST 3.3 versus TM 2.6; P〈0.001） during the follow-up. Satisfaction rates were similar in both groups. In conclusion, both techniques revealed to be safe and sustainable successful in penile straightening with better functional outcome of patients operated by STs.

INTRODUCTION Congenital hearing loss affects approximately two infants per 1,000 live births in the United States(USA),making it one of the nation’s most common developmental disabilities(1).When left undetected,permanent congenital hearing loss can negatively impact children through delays in their speech,language,social,and emotional development(2).Children who are identified as deaf or hard of hearing(D/HH)should begin receiving intervention services as early as possible to help mitigate the potential adverse effects(3).

Living in a High Mountain Border Region:the Case of the'Bhotiyas'of the Indo-Chinese Border Region

This article introduces one of South Asia's most important border regions into academic discourse, namely, the Central Himalayan mountain rim separating India and the Tibetan Autonomous Region (People's Republic of China). What makes this border region so interesting is a tangled interplay of changing environmental, cultural, and political forms to which the local populations constantly have to adapt in order to make a living there. We focused on the so-called 'Bhotiyas' of Uttarakhand, former trans-Himalayan traders whose ethnicity and livelihood was traditionally associated with the Indo-Chinese border that was sealed as a result of the India-China war in 1962. Drawing on the work of borderland scholarship, we identified the key processes and developments that changed the perspective of this area. Competing political aspirations as well as the 'Bhotiyas' countervailing strategies were considered equally important for understanding local livelihoods and identities within the dynamics of a 'high mountain border region'. Through an exemplary analysis of historical differences of power in one 'Bhotiya' valley, we further explored the ways in which shifting socio-spatial constellations are creatively re-interpreted by the borderlanders.

Ternary deep eutectic solvents catalyzed D-glucosamine self-condensation to deoxyfructosazine: NMR study

Ternary deep eutectic solvents(TDESs) comprising choline chloride(Ch Cl), glycerol and L-arginine were synthesized as catalysts and solvents for the conversion of D-glucosamine(GlcNH_(2)) into deoxyfructosazine(DOF). The interactions between these three components in the prepared TDESs were studied by ^(1)H-,^(35)Cl-NMR spectra and ^(1)H diffusion-ordered spectroscopy(DOSY) measurements. The chemical shift changes of active hydrogen in the ^(1)H-NMR spectra of TDES system and widening of signals in the^(35)Cl-NMR spectra confirmed the hydrogen bonding interaction between the components, which was further supported by the decrease of diffusion coefficients(D) of the TDES components according to ^(1)H DOSY measurements. The influences of reaction temperature and L-arginine content in the TDESs on the yield of DOF were also studied. The experimental results have shown that when the molar ratio of Ch Cl, glycerol, and L-arginine was 1:2:0.1, DOF was the major product with a yield of 22.6% at 90℃ for 120 min. The chemical shift titration indicated that the carboxyl group of L-arginine in the TDES is the catalytical active site, so the mechanism of the catalytic reaction between Glc NH_(2) and the TDES was proposed. Moreover, a reaction intermediate, dihydrofructosazine, was identified in the self-condensation reaction of Glc NH_(2) by an in situ ^(1)H NMR technique.

Optimal physicochemical properties of dried litchis for Thai consumers

The litchi is a fruit essential for the economies of several Southeast Asian countries,but markets regularly reject it,mainly due to *** extends the shelf-life of litchis,but in Thailand the optimal characteristics of the dried product have not yet been *** purpose of this study was to determine the optimum physicochemical properties of dried litchis-those suitable for Thai *** dried fruits were submitted to physicochemical measurement and consumer evaluation,with datasets subsequently integrated using circular ideal-point regression *** surface methodology was then used to predict the optimum physicochemical properties of the *** was found that Thai consumer preferences with regard to dried litchis are for the fruits to be of golden-yellow color(L*ranging around 54;H ranging around 79°),to have a soft flesh(SMF ranging between 13 and 14 kN/100 g)and to have a sweet taste(TSS:TA ranging between 25 and 28).The results may be used in the future to prescribe pretreatments and drying conditions.
