Chemical Characterization and in Vitro Antitumor Activity of the Essential Oils from the Leaves and Flowers of Callistemon viminalis

the objective of this study was to characterize and verify the in vitro antitumor activity of essential oils (EOs) extracted from the leaves and flowers of Callistemon viminalis. the EOs were extracted by hydrodistillation using a modified Clevenger apparatus. the identification and quantification of constituents were performed on a gas chromatograph coupled to a mass spectrometer and a gas chromatograph with a flame ionization detector. the antitumoral activity was evaluated by a colorimetric assay (MtS) using different cell lines derived from human tumors (breast, lung, glioblastoma, and melanoma). the major constituents of the EOs of leaves and flowers were similar, only quantitative differences being observed. the compounds 1,8-cineole, α-pinene and α-terpineol were found in concentrations of 50.4%, 25.8% and 8.7% in the EOs obtained from the leaves and 48.8%, 24.5% and 3.9% in the EOs obtained from the flowers, respectively. the cytotoxic activity of the EOs was observed only in melanoma cultures (Ht144). Cultures treated for 48 h with EOs from leaves and flowers (200 μg·mL-1) reduced the viability by 40% and 25%, respectively. thus, the antiproliferative activity of the EO from leaves was more pronounced than the EO from flowers in cells derived from melanoma.

the use of medicinal plants as therapeutic and economic resources has been on the rise in recent years. In Brazil, however, increasing doubt is being cast on the quality of these products, owing to the prevalence of adulteration and fraud. Solar radiation can cause serious damage to human skin, as a result, mostly, of ultraviolet light, which is a cause of skin cancer. Photoprotective substances are capable of absorbing, reflecting, or refracting ultraviolet radiation and thus protecting skin from exposure to sunlight. the present study aimed to characterize samples and examine the phytochemical profile and photoprotective potential of bark and leaves of Erythrina velutina Willd. the samples underwent five extraction methods using 80% ethanol. the phenolic content was measured using spectrophotometry. Antioxidant activity was examined using the DPPH and the photoprotective properties of the plant extracts were assessed using the method developed by Mansur. there was a quantitative difference in some groups of metabolites, with higher levels of tannin in the bark and of flavonoids in the leaves. the latter showed greater DPPH free radical scavenging capacity than the bark, although higher levels of SPF were obtained from the bark, with no statistically significant differences between methods. the results indicate that Erythrina velutina Willd. has potential as a photoprotector.

Subcutaneous Model for the Study of Dengue Virus Infection in Immune Competent Mice

Various mouse models to study dengue have been described by different authors, some of them using immunodeficient or some using humanized mice. Our group reported previously a deadly murine model, which used the intracranial inoculum of highly virulent Dengue virus (DENV) in immune competent mouse. Here we present a model of immune competent mouse (C57BL/6), infected subcutaneously by the same highly virulent DENV (DENV3 genotype I). In this immunocompetent systemic mice model, the cytokine levels and hematological parameters such as total and differential leukocyte and platelets counts, together with weight loss, were considered important monitoring parameters, allowing a better understanding of the systemic human disease. Mice were inoculated subcutaneously and evaluated by the percentage weight variation as well as the clinical signs. Hematological parameters and cytokines levels were measured and viral titration in brain tissue or serum neutralization was performed to confirm mice infection. the subcutaneously DENV inoculated mice showed weight loss after infection, but they did not show any other clinical signs. the leukocytes and platelets decreased after subcutaneous inoculation. the cytokines tNF alpha and IFN gamma increased after infection in mice. the subcutaneous model provided scope for improved understanding of the dengue pathogenesis, as well as possible mechanism for protection to subsequent mouse infected by intracranial route in mice. this model could be used to study the vertebrate immune response and evaluation of drugs or vaccine against dengue virus.

the upland rice productivity in the state of Rond?nia is still low, in view of the potential of culture. the use of cultivars adapted to different regions and more responsive to fertilizer employed is an essential practice which can change that. the aim of this study was to evaluate the agronomic characteristics and productivity of two upland rice cultivars with nitrogen, phosphorus and potassium (N-P-K) doses in two municipalities in the southern state of Rondonia region. the experimental design was a randomized block design with four replications in a factorial 2 × 2 × 5, with the first factor composed of two cultivars (hybrid Ecco and conventional farming AN Cambará), the second factor, the environment of the two municipalities, Cerejeiras and Vilhena and the third factor of five doses of N-P-K (0-0-0, 30-40-30, 60-60-60, 90-90-90 and 120-100-120 kg·ha-1). the characteristics evaluated were: tillering, number of integers and sterile grains per panicle, weight of 1000 grains, grain yield (kg·ha-1) and whole grain yield. there was no triple interaction between the three factors for any of the traits. the hybrid Ecco has higher tillering ability than AN Cambara and presents fewer sterile grains per panicle, heavier 1000 grains and hence greater productivity. For both cultivars, the highest yields are obtained with a dose of 120-100-120 kg·ha-1 N-P-K. For the Ecco, the productivity is achieved with this dose and the dose of 90-90-90 kg·ha-1 is statistically similar. there is no difference in productivity between the municipalities when the dose of N-P-K is less than 60-60-60 kg·ha-1. the highest yield of whole grains in function of N-P-K fertilization is obtained in Cerejeiras.

Introduction: Child obesity has important consequences on the child’s health and is related to the risk of developing chronic diseases. that brings up the need for research assessing the impact of obesity-and overweight-reduction programs with close attention to methodology aspects and precise indicators of the impact of such programs. Goal: Assessing the impact of a physical activity program on the plasma concentrations of adiponectin, leptin, and ghrelin in overweight and obese schoolchildren. Methods: Randomized controlled trial, double-blind, with 6-to 9-year-old overweight and obese school-children. A sample of 79 schoolchildren randomly distributed into control and intervention groups was subjected to a physical activity program. the levels of adiponectin, leptin, ghrelin, body mass index, body fat percentage, anthropometry, lipid profile and glycemia were evaluated. the program’s impact was assessed through multivariate linear regression models. Result: When comparing the groups, a significant difference was found for adiponectin, which shows an increase in the proportion of subjects with higher adiponectin concentrations for the intervention group and a reduction in body mass index and body fat percentage. Conclusion: the impact of the intervention program was positive in increasing adiponectin levels associated with a reduction in body mass index and body fat percentage.

Characterization of Virulence Factors in Enteroaggregative and Atypical Enteropathogenic Escherichia coli Strains Isolated from Children with Diarrhea

Enteroaggregative (EAEC) and atypical enteropathogenic (EPEC) Escherichia coli are important bacterial etiologic agents causing diarrhea among children. the aim of the present study was to examine the impact of virulence factors predisposes to diarrhea. In this study some virulence properties were examined on 11 EAEC and 8 EPEC strains identified by Polymerase Chain Reaction (PCR), isolated from stool samples of children were analyzed genotypically and phenoltypically for the prevalence of virulence factors. the most frequently detected factor was resistance to serum (94%), followed by curli fimbriae (78%), biofilm production (73%), and gene coding for Extended-Spectrum Beta-Lactamase (ESBL) (68%). EPEC isolates showed at least three of the evaluated properties, while EAEC isolates showed at least two. the prevalence of these virulence factors between the two strains showed no statistical difference. this study showed the heterogeneity of the virulence profile of the isolates of EAEC and atypical EPEC strains and suggests that this diversity may influence in the disease severity.

Stimulation of the P6 Acupuncture Point for Prophylaxis of Nausea and Vomiting in Pregnant Women Submitted to Cesarean Section: A Blinded Clinical trial

In pregnant women subjected to spinal anesthesia for a Cesarean section, episodes of nausea and vomiting are common both during and following surgery. Acupuncture for the prophylaxis and treatment of these complications has been gaining in popularity due to its low cost, simplicity, absence of side effects and confirmed efficacy. this study investigated the efficacy of stimulating the P6 acupoint in conjunction with the use of dexamethasone as prophylaxis for nausea and vomiting in pregnant women submitted to spinal anesthesia for a Cesarean section. the patients (n = 100) were randomly distributed into two groups. In the first group (n = 50), a site located one centimeter laterally from P6 was stimulated. this is not a true acupuncture point (sham acupuncture). In the second group (n = 50), P6 was stimulated. In both groups, 4 mg of dexamethasone were administered intravenously. A questionnaire was used to obtain information on the occurrence of nausea and vomiting during surgery and in the first 12 hours postpartum. the chi-square test and Fisher’s exact test were used to assess differences between the groups. Age and physical status were similar in both groups. the incidence of nausea during surgery was 32% (n = 16) in the control group and 22% (n = 11) in the P6 group (p > 0.05). In the first 12 hours following surgery, nausea occurred in 16% of the women in the control group (n = 6) and in 4% in the P6 group (n = 4) (p = 0.045). the incidence of vomiting in the control group was 12% (n = 6) during surgery and 10% (n = 5) in the postoperative period compared to 8% (n = 4) and 4% (n = 2), respectively, in the P6 group (p > 0.05). Although these differences were not statistically significant with the exception of the incidence of nausea in the first 12 hours postpartum, a reduction occurred in the incidence of all the outcomes evaluated in the P6 group.

Morbid obesity is regarded as a disease due to excess body weight, causing a silence of life as a whole and entailing the most varied disabilities for the person, such as: physical, social, psychological, affective, etc. It represents cases of “public health”, thereby involving competent bodies in the development of solutions that encompass various medical specialties and other health fields, in addition to influencing the mind of these people, causing depression that, due to metabolic involvement, can evolve to the death of the individual. the participation of multidisciplinary health focuses on weight loss, freely and spontaneously, or on the indication of bariatric surgery. We know how difficult it is to lose weight. In order to achieve successful procedures, we recommend the “Obese Workshop” or pre- and post-surgical follow-ups close to the patients, with a view to avoiding recurrences or the “accordion effect” (very common), which can interfere with the Body Mass Index (BMI). Everyone, males and females, complained of tiredness and the impossibility of any physical exercise, even the lightest and simplest to be performed, in addition to the fact that they cannot attend gyms and are unable to open a simple door handle.

Amyloidosis is the result of fibrous and insoluble amyloid protein deposition in extracellular spaces of tissues and organs, including the skin. through the analysis of medical records, clinical condition and laboratory tests, this article proposes a rare case report of an adult patient with symptoms and diagnosis of polymyositis, histopathology of nodular amyloidosis with skin lesions suggestive of lupus. the patient is still being followed at the dermatology clinic of Valença Medical School, in order to improve the prognosis and prevent the progression of existing symptoms.
