Evolutionary fingerprint, phylogenetic and forest structure of tropical montane Atlantic cloud forests along an elevation gradient

Environmental conditions can change markedly over geographical distances along elevation gradients,making them natural laboratories to study the processes that structure *** work aimed to assess the influences of elevation on Tropical Montane Cloud Forest plant communities in the Brazilian Atlantic Forest,a historically neglected *** evaluated the phylogenetic structure,forest structure(tree basal area and tree density)and species richness along an elevation gradient,as well as the evolutionary fingerprints of elevation-success on phylogenetic lineages from the tree *** do so,we assessed nine communities along an elevation gradient from 1210 to 2310 m *** large elevation *** relationships between elevation and phylogenetic structure,forest structure and species richness were investigated through Linear *** occurrence of evolutionary fingerprint on phylogenetic lineages was investigated by quantifying the extent of phylogenetic signal of elevation-success using a genus-level molecular *** results showed decreased species richness at higher elevations and independence between forest structure,phylogenetic structure and *** also verified that there is a phylogenetic signal associated with elevation-success by *** concluded that the elevation is associated with species richness and the occurrence of phylogenetic lineages in the tree communities evaluated in Mantiqueira *** the other hand,elevation is not associated with forest structure or phylogenetic ***,closely related taxa tend to have their higher ecological success in similar ***,we highlight the fragility of the tropical montane cloud forests in the Mantiqueira Range in face of environmental changes(*** warming)due to the occurrence of exclusive phylogenetic lineages evolutionarily adapted to environmental conditions(*** temperature)associated with each elevation range.

Aims Climate and soil are among the most important factors determin-ing variation in tree communities,but their effects have not been thoroughly elucidated to date for many vegetation *** this study,we evaluate how climate and soil gradients affect gradients of vegetation composition,species diversity and dominance,structure and functional traits(seed mass and wood density)using over 327000 trees in 158 sites distributed along environmental gradients in the transitions among the Atlantic forest,Cerrado and Caatinga in Minas Gerais State(MG),Brazil(nearly 600000 km2).Methods Gradients in species,genus and family abundance in addition to basal area,stem density,species diversity(Fisher’s alpha),domin-ance percentage,seed mass and wood density were correlated using multiple regressions with environmental variables,as summarized in four principal component analysis axes(two climatic-precipi-tation seasonality and temperature range-and two edaphic-soil fertility and soil moisture).Additionally,ordinary kriging maps were used to better illustrate the *** Findings Multiple regression models indicate that all variables but domin-ance percentage were affected by one or more of the environmental gradients,but the average R2 was low(26.25%).Kriging maps rein-forced the patterns observed in the regression *** seasonality and soil moisture gradients were the most important gra-dients affecting vegetation *** finding suggests that water availability is an important determinant of vegetation features in these vegetation transitions.

The Conservation Units (CU) were created to protect natural environments from growing degradation and to impede the expansion of urbanization and agricultural crops. The Serra da Canastra National Park, established to protect the headwaters of the S?o Francisco River and other places of scenic and ecological interest, is extensively visited due to its many tourist attractions, such as waterfalls, fauna and flora. An analysis of the park’s geography is needed to assess the risk involved in and its suitability for public use due to its territorial extension and environmental complexities. Thus, this study aimed to investigate the background of issues of interest to CU management. Additionally, we used high-resolution RapidEye imagery, altimetry and database of park infrastructure to build geospatial database and estimate classes of suitability for and risk in public use through GIS tools. The resulting cartographic data can support the planning of policies concerning the landscape and park’s territorial management.

Distribution and origin of polycyclic aromatic hydrocarbons in surface sediments from an urban river basin at the Metropolitan Region of Curitiba,Brazil

The concentrations of 16 polycyclic aromatic hydrocarbons（PAHs） were determined in surface sediment samples from nine sites located at the Iguau River Basin in the Metropolitan Region of Curitiba,Brazil to evaluate their distribution and *** total concentration of the PAHs was greater for sediments from highly urbanized areas,while the sediments from the Iraí Environmental Protection Area（Iraí EPA） showed significantly low *** sediments from the Iguau and Barigui rivers were classified as highly contaminated,while those from the Cercado and Curralinho rivers were classified as moderately *** predominance of PAHs containing two to four aromatic rings in most of the samples suggested the direct input of raw sewage into the water resources ***[g,h,i]perylene,dibenzo[a,h]anthracene and indeno[1,2,3-cd]pyrene were predominant in sediments from the areas under the greatest urban and industrial *** correlation between thermodynamic stability and the kinetics of evolution presented by the isomeric pairs indicated that combustion is the predominant source of PAHs in the sediments because the combustion of fossil fuels affected most of the points evaluated,followed by combustion of biomass and eventually combustion of oil product *** general,the results showed that areas under strong urban influence,as well as the Iraí EPA,receive contributions of PAHs from similar sources.

The effective osteointegration of orthopedic implants is a key factor for the success of orthopedic ***,local metabolic imbalance around implants under osteoporosis condition could jeopardize the fixation *** by the bone structure and the composition around implants under osteoporosis condition,alendronate(A)was grafted onto methacryloyl hyaluronic acid(H)by activating the carboxyl group of methacryloyl hyaluronic acid to be bonded to inorganic calcium phosphate on trabecular bone,which is then integrated with aminated bioactive glass(AB)modified by oxidized dextran(O)for further adhesion to organic collagen on the trabecular *** hybrid hydrogel could be solidified on cancellous bone in situ under UV irradiation and exhibits dual adhesion to organic collagen and inorganic apatite,promoting osteointegration of orthopedic implants,resulting in firm stabilization of the implants in cancellous bone *** vitro,the hydrogel was evidenced to promote osteogenic differentiation of embryonic mouse osteoblast precursor cells(MC3T3-E1)as well as inhibit the receptor activator of nuclear factor-κB ligand(RANKL)-induced osteoclast differentiation of macrophages,leading to the upregulation of osteogenic-related gene and protein *** a rat osteoporosis model,the bone-implant contact(BIC)of the hybrid hydrogel group increased by 2.77,which is directly linked to improved mechanical stability of the orthopedic ***,this organic-inorganic,dual-adhesive hydrogel could be a promising candidate for enhancing the stability of orthopedic implants under osteoporotic conditions.

Quantitative alpha-defensin testing: Is synovial fluid dilution important?

BACKGROUND Alpha-defensin has been widely studied for the diagnosis of periprosthetic joint infection(PJI).However,there is a lack of detailed information regarding the proper laboratory technique of the enzyme-linked immunosorbent assay(ELISA)method,such as sample *** To assess the influence of dilution in the synovial fluid during ELISA for the diagnosis of knee PJI;and determine which dilution presents a better *** Forty samples of synovial fluid from arthroplasty knees were included,17 in the infected group and 23 in the aseptic group,according to Musculoskeletal Infection Society ***,five synovial fluid samples from each group were assessed for quantitative analysis of alpha-defensin using *** dilution ratios(1:10,1:100,1:500,1:1000 and 1:5000)were tested based on the predetermined cutoff value of 5.2 mg/*** dilutions that performed better were used to compare the results of all *** For infected cases,a gradual increase in the dilution of synovial fluid samples led to an equivalent increase in alpha-defensin *** same was not observed in the aseptic *** 1:1000 and 1:5000 dilutions presented satisfactory results to differentiate infected and aseptic *** analyses were performed using 1:1000 and 1:5000 for all 40 *** 1:1000 dilution resulted in a sensitivity of 88.2%(95%CI,66%-98%)and specificity of 95.7%(95%CI,79%-99%),whereas the 1:5000 dilution presented a sensitivity of 94.1%(95%CI,73%-99%)and a specificity of 100%(95%CI,86%-100%).CONCLUSION The synovial fluid dilution had an important influence on the alpha-defensin ELISA *** of 1:5000 showed the best performance for the diagnosis of knee *** results of this study set the basis for a more reliable and reproducible alpha-defensin ELISA during the investigation of PJI,contributing to the expansion of this technique in different treatment centers worldwide.

Performance of alpha-defensin lateral flow test after synovial fluid centrifugation for diagnosis of periprosthetic knee infection

BACKGROUND The quantitative alpha-defensin enzyme-linked immunosorbent assay(ELISA)demands a prior synovial fluid centrifugation,whereas this processing is not routinely required prior to the alpha-defensin lateral flow *** To evaluate whether a prior synovial fluid centrifugation could lead the lateral flow performance to achieve comparable results to ELISA during periprosthetic joint infection(PJI)*** Fifty-three cases were included in this study:22 classified as PJI and 31 classified as aseptic cases,according to Musculoskeletal Infection Society 2013 *** fluid samples were submitted to centrifugation,and the supernatant was evaluated by ELISA and lateral flow *** sensitivity(SE),specificity(SP)and accuracy of each method were calculated as well as the agreement between those two *** In all of the 31 samples from aseptic patients,alpha-defensin ELISA and lateral flow tests showed negative results for *** the 22 infected patients,the lateral flow test was positive in 19 cases(86.4%)and the ELISA was positive in 21(95.5%).Sensibility,SP and accuracy were,respectively,86.4%(95%CI:65.1%-97.1%),100%(95%CI:88.8%-100%)and 93.2%(95%CI:82.8%-98.3%)for the lateral flow test and 95.5%(95%CI:77.2%-99.9%),100%(95%CI:88.8%-100%)and 98.1%(95%CI:89.9%-100%)for *** agreement of 96.2%between those methods were *** statistical difference was found between them(P=0.48).CONCLUSION Alpha-defensin lateral flow test showed high SE,SP and accuracy after a prior synovial fluid centrifugation,achieving comparable results to *** the lower complexity of the lateral flow and its equivalent performance obtained in this condition,a prior centrifugation might be added as a valuable step to enhance the PJI diagnosis.

Development and implementation of an automated system to aid laboratory diagnosis using image processing

The objective of this work is to provide an automatic system to count white blood cells in a blood smear. To do so an experiment was assembled, composed by a standard microscope with two step motors coupled to its knobs in order to move the microscope in x and y directions and a web cam which was mounted in the top of the microscope responsible for to acquire images from the smear. The step motors and the web cam are controlled by a microcomputer PC standard via software developed inDelphi. The motors use the parallel port to communicate with the PC and the camera use the USB port. The main idea is to set an initial point into the smear and the automated system will carry over the smear acquiring images (frames with 640 × 480 pixels) and counting the white blood cells encountered. The double histogram threshold technique is implemented to initially exclude the red cells from the image leaving only the white ones. Preliminaries results are obtained and show that the system is quite fast and has a good capacity of selection, even when different kinds of smear are used.

Are the cerrado and the seasonal forest woody floras assembled by different processes despite their spatial proximity?
