Dynamic localization of hepatocellular transporters in health and disease

Vesicle-based traffi cking of hepatocellular transporters involves delivery of the newly-synthesized carriers from the rough endoplasmic reticulum to either the plasma membrane domain or to an endosomal,submembrane compartment,followed by exocytic targeting to the plasma membrane. Once delivered to the plasma membrane,the transporters usually undergo recycling between the plasma membrane and the endosomal compartment,which usually serves as a reservoir of pre-existing transporters available on demand. The balance between exocytic targeting and endocytic internalization from/to this recycling compartment is therefore a chief determinant of the overall capability of the liver epithelium to secrete bile and to detoxify endo and xenobiotics. Hence,it is a highly regulated process. Impaired regulation of this balance may lead to abnormal localization of these transporters,which results in bile secretory failure due to endocytic internalization of key transporters involved in bile formation. This occurs in several experimental models of hepatocellular cholestasis,and in most human cholestatic liver diseases. This review describes the molecular bases involved in the biology of the dynamic localization of hepatocellular transporters and its regulation,with a focus on the involvement of signaling pathways in this process. Their alterations in different experimental models of cholestasis and in human cholestatic liver disease are reviewed. In addition,the causes explaining the pathological condition (e.g. disorganization of actin or actin-transporter linkers) and the mediators involved (e.g. activation of cholestatic signaling transduction pathways) are also discussed. Finally,several experimental therapeutic approaches based upon the administration of compounds known to stimulate exocytic insertion of canalicular transporters (e.g. cAMP,tauroursodeoxycholate) are described.

Effect of Poly(ε-caprolactone-b-tetrahydrofuran)Triblock Copolymer Concentration on Morphological,Thermal and Mechanical Properties of Immiscible PLA/PCL Blends

In this study a low molecular weight triblock copolymer derived fromε-caprolactone and tetrahydrofuran was used as a non-reactive compatibilizer of immiscible PLA/PCL *** blends with 0,1.5 wt%,3 wt%and 5 wt% copolymer and about 75 wt%PLA were prepared by single screw extrusion and characterized by scanning electron microscopy(SEM),differential scanning calorimetry(DSC),dynamic mechanical analysis(DMA),tensile and Izod impact *** micrographs showed that the size of the dispersed PCL domains was practically constant regardless of copolymer *** result can be explained by the low shear rate employed during processing step and a decrease of PCL viscosity by presence of the triblock ***,when the copolymer concentration increased,strain at break of PLA/PCL blends also ***/PCL blend with 0 wt% copolymer presented 2%strain at break,whereas PLA/PCL blend with 5 wt%copolymer exhibited 90%.

Glacial reconstruction and periglacial dynamics at the end of Late Pleistocene on the surface of Cofre de Perote volcano,México:a climatological retrospective

Despite being within the intertropical region of the planet,the Mexican territory still has glacier-covered *** recent decades,important advances have been made in studies on glaciology and periglacial environment in Mexico both for current and past ***,in spite of Cofre de Perote volcano(4200 m a.s.l.)being a strategically located mountain,it has not yet been studied in regards to the glacial and periglacial processes;in fact,those dynamics have modified the mountain massifs in the *** complement the series of studies on glacial history in the high mountain environment of México,this study reconstructs the glacial cover and the periglacial environment of the volcano surface during the final stage of the Late Pleistocene based on climatic retrospective and through the identification of geomorphological *** findings indicate the existence of a large glacier(ice cap)that covered the northern,western,and southern slopes of the mountain;while in the eastern sector there were two small glaciers,one being of cirque type,and the other of valley *** current temperature conditions prevent the occurrence of permanent ice bodies;at the same time,it was found that the periglacial blockfields of the slopes is a legacy of the climatic conditions that prevailed at the end of Late Pleistocene.

Settling,consolidation and shear strength behaviour of coal tailings slurry

The volume change and shear strength behaviour of tailings slurry with the changes in gravimetric moisture content is important to effectively utilise the storage volume and analysis of dam failure *** testing of tailings from a slurry-like to soil-like state is a critical task,and conventional consolidation apparatus does not have the capability of doing such testing,as the tailings slurries contain high water *** tests conducted on slurries having a 25%solids concentration showed very low efficiency;final sediment was still *** intrinsic point was identified based on pore water pressure dissipation during consolidation tests in a slurry consolidometer that can define two states of the tailings ***-like and *** the slurry-like state,the volume change was greater for Slurry 2 than Slurry 1,while the inverse was observed in the soil-like *** evolution of peak vane shear strength with the changes in moisture content was almost similar for both samples and obeyed the power *** models have been developed and validated to predict the shear strength of materials at any gravimetric moisture content or void ratio.

Computer-aided diagnosis(CAD)models exploit artificial intelligence(AI)for chest X-ray(CXR)examination to identify the presence of tuberculosis(TB)and can improve the feasibility and performance of CXR for TB screening and *** the same time,CXR interpretation is a time-consuming and subjective ***,high resemblance among the radiological patterns of TB and other lung diseases can result in ***,computer-aided diagnosis(CAD)models using machine learning(ML)and deep learning(DL)can be designed for screening TB *** this motivation,this article develops a Water Strider Optimization with Deep Transfer Learning Enabled Tuberculosis Classification(WSODTL-TBC)model on Chest X-rays(CXR).The presented WSODTL-TBC model aims to detect and classify TB on CXR ***,the WSODTL-TBC model undergoes image filtering techniques to discard the noise content and U-Net-based image ***,a pre-trained residual network with a two-dimensional convolutional neural network(2D-CNN)model is applied to extract feature *** addition,the WSO algorithm with long short-term memory(LSTM)model was employed for identifying and classifying TB,where the WSO algorithm is applied as a hyperparameter optimizer of the LSTM methodology,showing the novelty of the *** performance validation of the presented WSODTL-TBC model is carried out on the benchmark dataset,and the outcomes were investigated in many *** experimental development pointed out the betterment of the WSODTL-TBC model over existing algorithms.

Incidence and survival of stomach cancer in a high-risk population of Chile

AIM:To study the incidence and survival rate of stomach cancer(SC)and its associated factors in a high risk population in Chile. METHODS:The population-based cancer registry of Valdivia,included in the International Agency for Research on Cancer system,covers 356 396 residents of Valdivia Province,Southern *** studied all SC cases entered in this Registry during 1998-2002 (529 cases).Population data came from the Chilean census(2002).Standardized incidence rates per 100 000 inhabitants(SIR)using the world population, cumulative risk of developing cancer before age 75, and rate ratios by sex,age,ethnicity and social factors were *** survival(EdererⅡmethod) and age-standardized estimates(Brenner method) were *** survival rates(Kaplan-Meier) were measured at 3 and 5 years and survival curves were analyzed with the Logrank and Breslow tests. Survival was studied in relation to demographics, clinical presentation,laboratory results and medical management of the *** variables significantly associated with survival were later included in a Cox multivariate model. RESULTS:Between 1998 and 2002,529 primary gastric cancers occurred in Valdivia(crude incidence rate 29.2 per 100000 inhabitants).Most cases were male(69.0%), residents of urban areas(57.5%)and Hispanic(83.2%), with a low education level(84.5%<8 school years). SC SIR was higher in men than women(40.8 and 14.8 respectively,P<0.001),risk factors were low education RR 4.4(95%CI:2.9-6.8)and 1.6,(95%CI:1.1-2.1) for women and men respectively and Mapuche ethnicity only significant for women(RR 2.2,95%CI:1.2-3.7).Of all cases,76.4%were histologically confirmed,11.5% had a death certificate only(DCO),56.1%were TNM stageⅣ;445 cases(84.1%)were eligible for survival analysis,all completed five years follow-up;42 remained alive,392 died of SC and 11 died from other causes. Specific 5-year survival,excluding cases with DCO,was 10.6%(95%CI:7.7-13.5);5-year relative survival rate was 12

EDT Method for Multiple Labelled Objects Subject to Tied Distances

The success of new scientific areas can be assessed by their potential for contributing to new theoretical approaches aligned with real-world *** Euclidean distance transform(EDT)has fared well in both cases,providing a sound theoretical basis for a number of applications,such as median axis transform,fractal analysis,skeletonization,and Voronoi *** its wide applicability,the discrete form of the EDT includes interesting properties that have not yet been fully exploited in the *** this paper,we are particularly interested in the properties of 1)working with multiple objects/labels;and 2)identifying and counting equidistant pixels/voxels from certain points of *** some domains(such as dataset classification,texture,and complexity analysis),the result of applying the EDT transform with different objects,and their respective tied distances,may compromise the *** this sense,we propose an efficient modification in the method presented in[1],which leads to a novel approach for computing the distance transform in a space with multiple objects,and for counting equidistant pixels/voxels.

Enzymatic Hydrolysis of Sugarcane Biomass and Heat Integration as Enhancers of Ethanol Production

The aim of this study is to assess the possibility of increasing ethanol production by introducing the bagasse hydrolysis process into conventional *** were performed for mass and energy balances using Aspen Plus?*** was assumed that sugarcane trash and lignin cake—hydrolysis process residues—are available as supplementary *** cases were evaluated,including:(a)conventional ethanol distillery,(b)conventional plant combined with a hydrolysis process without heat integration,with different solid contents in the hydrolysis reactor,and(c)conventional plant combined with the hydrolysis process applying heat integration by pinch *** highest ethanol yield was achieved in the case of heat integration and concentration of cellulose hydrolysate by the membrane system with a solid content of 5%in the hydrolysis *** represents an increase of 22%over conventional distilleries currently found in the industry.
