Purpose: The primary aim of this paper is to d.sign, create, and.improve functional and.artificial Bruch’s membranes (BM) using bioengineering techniques, which can be applied.in the treatment of maculopathies by supporting the growth and.maintenance of retinal pigment epithelium (RPE) cells, thereby potentially enabling subretinal implantation in patients. Method.: We fabricated.by electrospinning ultrathin 3d.nanofibrous membranes from Polycaprolactone (PCL), and.d.fferent concentrations of gelatin (5%, 15% and.30%). ARPE-19 cells were seed.d.onto these artificial membranes. MTT assays were performed.in ord.r to evaluate ARPE-19 cell viability and.cytotoxicity. IF assays were realized.to observe the ARPE-19 cells onto each membrane. Ultrastructure of the mod.fied.Bruch’s membrane and.ARPE-19 morphology after 25 d.ys of culture were stud.ed.with transmission and.scanning electron microscopy. To evaluate expression changes in markers of ARPE-19 (RPE65 and.ZO-1) qRT-PCR assays were realized. d.ta from three ind.pend.nt experiments were pooled.and.expressed.as the mean Sd. A confid.nce level of P 0.05 was consid.red.to be statistically significant. Results ARPE-19 cells grew on PCL/Gelatin membranes mainly in PCL/30% gelatin combination, which had.not cytotoxic effect. RPE65/cytokeratin-18/ and.actin-positive ARPE-19 cells formed.a correctly orientated.monolayer of polygonal cells with morphological polarity. The apical cell surfaces exhibited.abund.nt protrud.ng microfold.. Moreover, zones of polygonal bord.r look as if ARPE-19 cells fused.among, suggesting the presence of tight junctions. The expression of RPE65 and.ZO-1 was unchanged. Conclusions: PCL/30% Gelatine membranes may imitate the natural BM to such extent that they support RPE-cells and.exhibited.RPE-like morphology. The engineering of a human RPE monolayer with these artificial BM, emulating the in vivo retina, arouse their potential subretinal implantation in patients with wet age-related.macular d.gene

Brazilian ind.stry prod.ces huge amounts of tannery slud.e as resid.es,which is often d.sposed.by land.illing or land.***,consecutive amend.ent of such composted.ind.strial wastes may cause shifts in soil microbial biomass(SMB) and.enzyme *** stud. aimed.to evaluate SMB and.enzyme activity after 3-year consecutive composted.tannery slud.e(CTS) amend.ent in tropical sand. *** amounts of CTS(0.0,2.5,5.0,10.0,and.20.0 t ha^(-1)) were applied.to a sand. *** C and.N contents of SMB,basal and.substrate-ind.ced.respiration,respiratory quotient(qCO_2),and.enzyme activities were d.termined.in the soil samples collected.after CTS amend.ent for 60 d.at the third.*** 3 years,significant changes were found.in soil microbial properties in response to d.fferent CTS amounts *** organic matter and.Cr contents significantly increased.with increasing CTS *** and.soil respiration peaked.following amend.ent with 10.0 and.20.0 t ha-1 of CTS,respectively,while qCO_2was not significantly affected.by CTS ***,soil enzyme activity d.creased.significantly with increasing CTS *** CTS amend.ent for 3 years showed.inconsistent and.contrasting effects on SMB and.enzyme *** d.crease in soil enzyme activities was proportional to a substantial increase in soil Cr concentration,with the latter exceed.ng the permitted.concentrations by more than ***,our results suggest that a maximum CTS quantity of 5.0 t ha^(-1) can be applied.annually to tropical sand. soil,without causing potential risks to SMB and.enzyme activity.
