Most plant cells contain a large central vacuole that is essential to maintain cellular turgor. We report a new mutant allele of VTI11 that implicates the SNARE protein VTI11 in homotypic fusion of protein storage and lytic vacuoles. Fusion of the multiple vacuoles present in vtill mutants could be induced by treatment with Wortmannin and LY294002, which are inhibitors of Phosphatidylinositol 3-Kinase （PI3K）. We provide evidence that Phosphatidylinositol 3-Phosphate （Ptdlns（3）P） regulates vacuole fusion in vtill mutants, and that fusion of these vacuoles requires intact microtubules and actin filaments. Finally, we show that Wortmannin also induced the fusion of guard cell vacuoles in fava beans, where vacuoles are naturally fragmented after ABA-induced stomata closure. These results suggest a ubiquitous role of phosphoinositides in vacuole fusion, both during the development of the large central vacuole and during the dynamic vacuole remodeling that occurs as part of stomata movements.

AIM To investigate the contribution of polymorphisms in the CYP1A1, CYP2E1 and EPHX1 genes on sporadic colorectal cancer(SCRC) risk. METHODS Six hundred forty-one individuals(227 patients with SCRC and 400 controls) were enrolled in the study. The variables analyzed were age, gender, tobacco and alcohol consumption, and clinical and histopathological tumor parameters. The CYP1A1 *2A, CYP1A1 *2C CYP2E1 *5B and CYP2E1 *6 polymorphisms were analyzed by polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP). The EPHX1 Tyr113 His, EPHX1 His139 Arg and CYP1A1 *2C polymorphisms were detected by real-time PCR. Chisquared test and binary logistic regression were used in the statistical analysis. Haplotype analysis was conducted using the Haploview program, version *** Age over 6 2 years was a risk factor for SCRC development(OR = 7.54, 95%CI: 4.94-11.50, P < 0.01). Male individuals were less susceptible to SCRC(OR = 0.55, 95%CI: 0.35-0.85, P < 0.01). The CYP2E1*5B polymorphism was associated with SCRC in the codominant(heterozygous genotype: OR = 2.66, 95%CI: 1.64-4.32, P < 0.01), dominant(OR = 2.82, 95%CI: 1.74-4.55, P < 0.01), overdominant(OR = 2.58, 95%CI: 1.59-4.19, P < 0.01), and log-additive models(OR = 2.84, 95%CI: 1.78-4.52, P < 0.01). The CYP2E1*6 polymorphism was associated with an increased SCRC risk in codominant(heterozygous genotype: OR = 2.81, 95%CI: 1.84-4.28, P < 0.01; homozygous polymorphic : OR = 7. 3 2, 9 5 % C I : 1.85-28.96, P < 0.01), dominant(OR = 2.97, 95%CI: 1.97-4.50, P < 0.01), recessive(OR = 5.26, 95%CI: 1.35-20.50, P = 0.016), overdominant(OR = 2.64, 95%CI: 1.74-4.01, P < 0.01), and log-additive models(OR = 2.78, 95%CI: 1.91-4.06, P < 0.01). An essential oil is the volatile lipophilic component extracted from plants. Microencapsulation systems protect the essential oil from degradation and evaporation, and at the same time allow a sustained release. This work analyzed and characterized the rosemary essential oil microcapsules prepared by co-extrusion technique using alginate as wall material and calcium chloride as cross linker. Several instrumental techniques were used: optical microscopy, coulter counter, Fourier transform infrared spectroscopy (FTIR), differential scanning calorimetry (DSC), termogravimetric analysis (TGA), spectrophotometry, antimicrobial test and chromatography. Results show that rosemary oil has pesticidal properties, and its microencapsulation allows knowing that these properties remain inside the microcapsules.

Introduction: Obesity has been considered a global epidemic and a serious public health problem closely related to other diseases. Moreover, obese individuals have limitations in physical capacity and pulmonary function. Weight loss induced by bariatric surgery appears to help reverse this situation. However, questions about physical capacity and lung function, and its relationship with body composition remain to be clarified. Objective: To evaluate the physical capacity, body composition and lung function in women undergoing bariatric surgery. Methods: Twenty five women who underwent bariatric surgery between 6 and 24 months ago, and 10 non-obese women were evaluated for exercise capacity by the six minutes walk test (6 MWT), body composition by bio-electrical impedance analysis and lung function by spirometry and measurement of maximal static respiratory pressures. Results: Subjects with BMI 2 were found to have better physical performance, demonstrated by increased 6 MWT distance, not differing from non-obese women. When compared with non-obese women, there were no significant differences in body composition and lung function. Higher BMI was correlated with shorter distances in 6 MWT. Conclusion: 6 to 24 months after surgery, when the weight loss of patients who underwent bariatric surgery reaches levels considered non-obese, it provides improvement in physical performance to a level similar to that of non-obese patients.

Colombian ecosystems maintain key ecological processes that support thousands of species, including human beings. With the expansion of the country’s population, and the implementation of a government’s development plan based on an economy centred on extraction patterns, the conservation of these ecosystems is at serious risk. It is a priority to implement effective strategies that ensure the protection of the country’s biological diversity as well as the mitigation and prevention of threats and to contribute to its proper use. Colombia’s development strategies as well as its peoples’ wellbeing depend on the suitable condition of its natural assets. The identification of surrogates of conservation, the formulation of conservation goals, the prioritization of key areas and the formulation of conservation strategies based on the preservation, restoration and sustainable use of the territory and its biodiversity are proposed for 60% of the emerged land (~ 700,000 Km2). This research aims at giving proper guidelines to manage the territory and finding common points between development and biodiversity conservation, as well as to use this input for the development and implementation of a National Decision-making Support System (DSS) that will potentially have an impact on Colombia’s environmental policies and territorial planning schemes.

DNA vaccines are the third generation vaccines based on purified plasmid preparations containing transgenes that encode antigenic/therapeutic proteins or peptides capable of triggering an immune response against a wide range of diseases. This vaccine platform presents several attributes that confer distinct advantages over other vaccine technologies in terms of safety, ease of fabrication and stability. Many aspects, such as antigen expression and especially vector design, are under study because of their great influence on immunogenicity and efficacy of DNA vaccines. In this regard, with the attempt of improving the efficiency of DNA vaccines, co-expression of stimulatory sequences and diverse vector delivery systems are being optimized. With this in mind, this review aims to giving a conceptual approach of DNA vaccines, explaining their mechanisms of action and listing the already licensed veterinary DNA vaccines presented in the market.

GTD (Gestational Trophoblastic Disease) is a pathology that encompasses benign and malignant clinical forms, affects women of childbearing age, has a variable incidence and is more frequent in developing or underdeveloped countries, colliding with the economic barrier. The frequent absence of clear protocols and guidelines for the correct diagnosis of the pathology results in inadequate classification, imprecise treatment and failed post-therapeutic observation, increasing the risk of relapses, morbidity and mortality. The present study aims to point out updated national and international practice protocols of diagnosis of GTD, through an integrative review. Seven articles were selected and it was observed that the main international reference centers are agreed with the management suggested by the IFGO (International Federation of Gynecology and Obstetrics), being the conduct in the Hydatidiform Mole (HM): evacuation by suction and curettage under ultrasound guidance, followed by hCG monitoring every 1 - 2 weeks until normalized (usually one month for Partial Hydatidiform Mole six months for Complete Hydatidiform Mole and one year for Gestational Trophoblastic Neoplasia). Unfortunately, regarding the diagnosis of MH, the guidelines of some countries show the absence or difficulty of access to the karyotype test and ploid p57 or pelvic ultrasound accompanying the uterine curettage, contrary to what is proposed by the IFGO guideline. Establishing and complying with consistent guidelines can improve patient care, with early diagnosis of the pathology and its complications, reducing the rate of recurrence, morbidity and mortality, especially in less developed countries.

The present study aims to compare the different national and international guidelines in the management of GTD, through an integrative review. Nine articles were selected and it was observed that the main international reference centers are agreed with the management suggested by the IFGO (International Federation of Gynecology and Obstetrics), being the conduct in the Hydatidiform Mole (HM): evacuation by suction and curettage under ultrasound guidance, followed by hCG monitoring every 1 - 2 weeks until normalized;in low-risk GTN (Gestational Trophoblastic Neoplasm): chemotherapy with methotrexate or actinomycin D, in high-risk: EMA/CO protocol, in ultra-high-risk EMA/PE, methotrexate with radiotherapy for brain metastases. All medical societies recommend the registration of these patients in GTD screening centers, endorse the use of the IFGO scoring system (2000) and recommend the surgical management of placental site trophoblastic or epithelioid tumors, as chemotherapy is less effective in these cases. The controversies are in the proper follow-up after the treatment of HM, use of ultrasound to evacuate the uterus, administration of anti-D immunoglobulin, time of oxytocin infusion and rescue regimens that can be used in cases of resistant or recurrent GTN.
