Plasma Levels of Angiotensin-Converting Enzymes 1 and 2 and AGTR2 (T1247G and A5235G) Gene Polymorphisms Are Associated to Breast Cancer Progression

Background: Breast cancer is the most common type of cancer among women. Diagnosed and treated timely, patients may have good prognostics. in Brazil, in 2012, the estimate of new cases was 52,680 and the number of registered deaths in 2012 was 12,852. The Renin-Angiotensin System (RAS) is known for its role in arterial hypertension and in other cardiovascular diseases. Angiotensin-Converting Enzyme 2 (ACE2) is the key to Ang-(1-7) formation, and counterbalances the ACE1/Angii/AGTR1 axis actions. RAS components have complex interactions with different tissues and their actions are not restricted to the cardiovascular system. Recently, the RAS has been associated with different types of cancers and in particular with gynecological cancers. Objectives: Our aim is to investigate possible associations between allelic distribution of two genetic polymorphisms in the AGTR2 receptor with ACEs 1 and 2 plasma levels among women with breast cancer. Patients and Methods: Patients with breast cancer were genotyped for two polymorphisms of the AGTR2 (T1247G and A5235G). Genotyping assays (TaqMan) were performed with genomic DNA extracted from blood cells. ACEs plasma level measurements were conducted in women from the breast-cancer group (N = 53). ACEs were measured in the plasma of these patients using ELiSA kits. Results: SNPs genotype distribution is correlated with ACEs plasma levels. ACEs plasma levels are also correlated with clinical variables and ACE2 high levels are associated with better prognostics. Conclusions: Changes in circulating levels of ECA1/Angii ECA2/ Ang-(1-7) determine the magnitude of the inflammatory response that an individual can trigger and the variation in ACE 1 and 2 plasma level measurements in the blood of breast cancer patients suggests an association with the process of mammary carcinogenesis. Thus, the RAS may be associated with the process of mammary carcinogenesis by both genotypic variations of RAS components and by circulating levels of ACEs

Nutritional Characteristics of Brachiaria brizanthaCultivars Subjected to Different intensities Cutting

This study evaluated the nutritional characteristics of Brachiaria brizantha cultivars subjected to three cutting intensities in the course of one year. The experiment was conducted at the University of Rio Verde. The experiment was performed as a randomized 3 × 3 factorial in complete block design with three replications and repeated measures in the time. Three Brachiaria brizantha cultivars (Marandu palisadegrass, Xaraes palisadegrass, and Piata palisadegrass), were tested at three cutting intensities (10, 20, and 30 cm sward height). The evaluations were conducted on the same plots throughout one year and during all four seasons (autumn, winter, spring, and summer). The results showed that the Piata palisadegrass had the best chemical composition compared to the Marandu palisadegrass and Xaraes palisadegrass. The management of Brachiaria brizantha cultivars at the lowest residual height (10 cm) affected the nutritional value of these feed grasses. Seasonality also influenced the nutritional value of these forages.

Low Genetic Polymorphism at the Cytochrome C Oxidase i in Silkworm Strains of the Brazilian Germplasm Bank

Nucleotide sequences have been used to distinguish species and specimens for many years. More recently, the use of a partial sequence of 650 bp of the cytochrome c oxidase i, COi mitochondrial gene, has been proposed for species identification, known as DNA barcodes. in this work, a short sequence of the DNA barcode is described—approximately 250 bp, named as “DNA mini-barcode”—to molecularly identify different silkworm strains maintained at the unique public Germplasm Bank of Bombyx mori, at the Universidade Estadual de Maringá, UEM, Brazil. Analysis revealed no significant differences among the silkworm strains. The phylogenetic tree obtained by the neighbor-joining method and K2P distance, in which specimens of B. mandarina were used as outgroup, clustered all the specimens of B. mori in a unique clade. Genetic variability detect within B. mori was low or nonexistent. in conclusion, the partial region of 250 bp of the mitochondrial gene COi herein analyzed may not be efficient to discriminate silkworm strains from the UEM Germplasm Bank of Bombyx mori.

Liver biopsy for visceral leishmaniasis diagnosis in pregnancy: report of 2 cases

Visceral leishmaniasis(VL) or kala-azar is a zoonosis caused by intracellular protozoa of the Leishmania genus and is transmitted to humans by the bite of phlebotomine sandflies. it particularly affects cells in the phagocytic mononuclear system, accompanied by disturbances of cellular and humoral immunity. VL is potentially fatal and is characterized by fever, hepatosplenomegaly, diarrhea, epistaxis, jaundice, anemia, leucopenia, thrombocytopenia, hypoalbuminemia and hyperglobulinemia. Diagnostic suspicion is based on epidemiological, clinical and laboratory data and isconfirmed by detecting the parasite in infected tissue. Splenic aspiration is the most sensitive method, followed by bone marrow aspiration(BMA) by sternal puncture, liver biopsy and lymph node aspiration; but, due to safety concerns, BMA is the most recommended method. VL is included as a target disease by players in drug research and development. Severe liver dysfunction associated with VL is uncommon. We report two VL cases in pregnant women from Bauru, Sao Paulo state, Brazil, considered an endemic area. The first of them developed hepatic failure due to fulminant hepatitis. in both cases, BMA was unable to find the protozoan; thus, liver biopsy was the only means of making the diagnosis.

Essential Oils from Lippia origanoidesKunth. and Mentha spicataL.: Chemical Composition, insecticidal and Antioxidant Activities

This work describes the chemical composition of the essential oils extracted from fresh leaves of Lippia origanoides Kunth. and Mentha spicata L. and their antioxidant and insecticidal activities. The essential oils were extracted by steam distillation using a modified Clevenger apparatus and subsequently analyzed by gas chromatography-flame ionization detector and gas chromatography-mass spectrometry. The antioxidant activities were determined by the β-carotene-linoleic acid method and by sequestration of the 2,2-diphenyl-1-picryl-hidrazila radical. The concentrations of the essential oils and the synthetic standard, butylated hydroxyltoluene, were 25, 50, 100, 150, 200, 250, 300 and 500 μg mL-1. insecticidal activity was analyzed by non-preference with choice and no choice against the aphid Myzus persicae Sulzer. Gas chromatography analysis of the essential oil from Lippia origanoides Kunth. revealed carvacrol (41.51%), p-cymene (18.36%), γ-terpinene (17.03%) and thymol (4.86%) as major constituents, and the essential oil from Mentha spicata L. contained piperitona (81.18%), piperitenone (14.57%) and limonene (1.47%) as the principal components. The essential oils and the standard exhibited dose-dependent antioxidant activities at the concentrations tested. The essential oils were shown to be potential agents in the integrated management of the aphid Myzus persicae Sulzer.
