Modification of Periodate Oxidation Method to Produce HRP-IgG conjugate and Test its Stability Overtime

Goat anti-Rabbit Horseradish Peroxidase (HRP) secondary conjugate was produced using a modified periodate oxidation method. The obtained conjugate was tested in the quality control techniques of therapeutic proteins. To determine the working dilution, titration of the prepared conjugate was performed in Indirect Enzyme-Linked Immunosorbent Assay (ELISA) and found to be 1:5000. This dilution was further tested in Western Blot analysis. The secondary conjugate was kept at 4°c for one month and its stability was verified by Western Blot and Indirect ELISA techniques.

Understanding the holistic relationship between refinery production scheduling(RPS) and the cyber-physical production environment with smart scheduling is a new question posed in the study of process systems engineering. Here, we discuss state-of-the-art RSPs in the crude-oil refining field and present examples that illustrate how smart scheduling can impact operations in the high-performing chemical process industry. We conclude that, more than any traditional off-the-shelf RPS solution available today, flexible and integrative specialized modeling platforms will be increasingly necessary to perform decentralized and collaborative optimizations,since they are the technological alternatives closer to the advanced manufacturing philosophy.

AIM： To investigate the relationship between mucin 6 （MUc6） VNTR length and Hpylori infection. METHODS： Blood samples were collected from patients visiting the can Tho General Hospital for upper gastrointestinal endoscopy. DNA was isolated from whole blood, the repeated section was cut out using a restriction enzyme （Pvu 11） and the length of the allele fragments was determined by Southern blotting. Hpylori infection was diagnosed by ^14c urea breath test. For analysis, MUc6 allele fragment length was dichotomized as being either long （〉 13.5 kbp） or short （≤ 13.5 kbp） and patients were classified according to genotype [long- long （LL）, long-short （LS）, short-short （SS）]. RESULTS： 160 patients were studied （mean age 43 years, 36% were males, 58% H pylori positive）. MUc6 PvuⅡ-restricted allele fragment lengths ranged from 7 to 19 kbp. Of the patients with the LL, LS, SS MUc6 genotype, 43% （24/56）, 57% （25/58） and 76% （11/46） were infected with Hpylori, respectively （P = 0.003）. cONcLUSION： Short MUc6 alleles are associated with H pylori infection.

Technological development, particularly the development of signal processing, has contributed to the improvement of the quality of results in the analysis of the characteristics of hydrocarbon reservoirs. This scientific advance is especially marked by the introduction of seismic attributes in the characterization of reservoirs. This paper is aimed at evaluating the contribution of seismic attributes in the lithological characterization of hydrocarbon accumulation zones. Thus, it has been found that instantaneous attributes make it possible to highlight the different properties of reservoir rocks. Techniques for extracting three-dimensional volume attributes have been applied to a 3D seismic data volume of the F3 block in the Dutch sector of the North Sea. The purpose of using these volume attributes is to extract signal characteristics from lithofacies in hydrocarbon zones. The results provide a remarkable contribution to instantaneous seismic attributes in reservoir location, hydrocarbon detection, and reservoir lithological prediction. The instantaneous amplitude, the derivative of amplitude 1, the factor Q, the energy and the Hilbert transform are attributes which highlight the presence of hydrocarbons through the bright spots. Thus, curvature, similarity, dip, and coherence are useful in the determination of traps such as faults and anticlines. The crossing of the instantaneous amplitude and the instantaneous frequency makes it possible to analyze the lithofacies. In the same way, the analysis of the crossed diagrams between the porosity and the instantaneous amplitude made it possible to define the different layers of the reservoir.

The present work is a study on the influence of temperature and the velocity of air on the drying kinetics of pineapple slices. The main objective is to define from this drying kinetics rules to help a designer choose an adequate principle or drying configuration and optimize the design of dryers adapted to the use of it. The experiments were made in an electric dryer using a parallel flow system. The air was used at three different temperatures （40, 50 and 60 ~c） and at four different velocities （0.27, 0.5, 1 and 1.8 m/s） during the experiment. The results show that an increase of 10 ℃ of the drying temperature reduced the drying time by 45% and increased the drying velocity by 30%. On the set of air velocities tested, it was noticed that the influence of air velocity on drying was more striking at the beginning than at the end of the drying process （variation of less than 20%）. Whatever the variation of parameters during the drying process, the best performance was observed when the products water content during the drying process was between 50% and 80%.

Background:Inbreeding depression can adversely affect traits related to fitness,reproduction and productive *** current research suggests that inbreeding levels are generally low in most goat breeds,the impact of inbreeding depression on phenotypes of economic interest has only been investigated in a few studies based on genealogical ***:We genotyped 1040 goats with the Goat SNP50 *** information was used to estimate different molecular inbreeding coefficients and characterise runs of homozygosity and homozygosity *** detected38 genomic regions with increased homozygosity as well as 8 ROH hotspots mapping to chromosomes 1,2,4,6,14,16 and *** hundred seventeen goats with available records for dairy traits were analysed to evaluate the potential consequences of inbreeding depression on milk *** regions on chromosomes 8 and 25were significantly associated with inbreeding depression for the natural logarithm of the somatic cell ***,these regions contain several genes related with immunity,such as SYK,IL27,ccL19 and ***,one region on chromosome 2 was significantly associated with inbreeding depression for milk ***:Although genomic inbreeding levels are low in Murciano-Granadina goats,significant evidence of inbreeding depression for the logarithm of the somatic cell count,a phenotype closely associated with udder health and milk yield,have been detected in this *** inbreeding would be expected to augment economic gain by increasing milk yield and reducing the incidence of mastitis,which is one of the main causes of dairy goat culling.

The project consists in the implementation of a biocomposite based on tannin resin and natural rubber matrices with the bast fibres of Triumfetta cordifolia ***.“Okong” from the equatorial region of cameroon as reinforcement. A study of this still little known fibre is necessary. This paper evaluates the physico-chemical and mechanical characteristics of the fibers. The fibers are extracted by us. A series of experiments is conducted for this purpose: morphological observation with a scanning electron microscope (SEM);density evaluation with a helium pycnometer;absorption rate evaluation according to the protocol available in the literature, Fourier Transform Infrared Spectrometry (FT-IR), chemical composition evaluation according to ASTM 1972 and ASTM 1977 standards, thermogravimetric analysis (TGA) and tensile tests on fiber bundles according to NF T25-501-3. The results show that the fiber is made up of several elementary fibers with oval cross-sections. A density of 1.477g/cm3 close to that of hemp. These fibers have a water absorption rate of 342.5%, which correlates with the presence of free hydroxyl functional groups obtained from the spectrometry study (FT-IR). chemical analysis reveals that the fiber is made up of celluloses (44.4%), hemicelluloses (30.8%), lignins (18.9%), pectins (3.3%), waxes (0.5%) and minerals (2.1%). In addition, we learn that the fibers studied dehydrate at 11.49%, showinga notable thermal stability around 235°c with a peak thermal decomposition of cellulose located at 420°c. In terms of mechanical behaviour, the results reveal that the fibers offer a Young’s modulus in traction of 12.4 ± 6.9 GPa, a tensile strength of 526 ± 128 MPa and an elongation at break of 2.25%. The information thus obtained makes it possible to place these fibers in the same fiber group as flax and jute. They could therefore be used for the same types of applications. AIM： Although increased insulin-like growth factor-I receptor （IGF-IR） gene expression has been reported in hepatocellular carcinoma, studies assessing IGF-IR in chronic hepatitis c （cHc） and cirrhosis are scarce. We therefore aimed to evaluate IGF-IR and IGF-I rnRNA expression in liver from patient with cHc. METHODS： IGF-IR and IGF-I rnRNA content were determined by semi-quantitative RT-PcR and IGF-IR protein expression was determined by immunohistochemistry in hepatic tissue obtained from patients with cHc before （34 patients） and after （10 patients） therapy with interferon-α and ribavirin. RESULTS： An increase of IGF-IR rnRNA content was observed in hepatic tissue obtained from all cHc patients as well as from 6 cadaveric liver donors following orthopic transplantation （an attempt to evaluate normal livers） in comparison to normal liver, while no relevant modifications were detected in IGF-I mRNA content. The irnrnunohistochemical results showed that the raise in IGF-IR rnRNA content was related both to ductular reaction and to increased IGF-IR expression in hepatocytes. A decrease in IGF-IR rnRNA content was observed in patients who achieved sustained virological response after therapy, suggesting an improvement in hepatic damage. cONcLUSION： The up-regulation of IGF-IR expression in hepatocytes of patients with cHc could constitute an attempt to stimulate hepatocyte regeneration. considering that liver is the organ with the highest levels of IGF-I, our finding of increased IGF-IR expression after both acute and chronic hepatic damage highlights the need for additional studies to elucidate the role of IGF-I in liver regeneration.

Multispectral microscopy enables information enhancement in the study of specimens because of the large spectral band used in this technique. A low cost multimode multispectral microscope using a camera and a set of quasi-monochromatic Light Emitting Diodes (LEDs) ranging from ultraviolet to near-infrared wavelengths as illumination sources was constructed. But the use of a large spectral band provided by non-monochromatic sources induces variation of focal plan of the imager due to chromatic aberration which rises up the diffraction effects and blurs the images causing shadow around them. It results in discrepancies between standard spectra and extracted spectra with microscope. So we need to calibrate that instrument to be a standard one. We proceed with two types of images comparison to choose the reference wavelength for image acquisition where diffraction effect is more reduced. At each wavelength chosen as a reference, one image is well contrasted. First, we compare the thirteen well contrasted images to identify that presenting more reduced shadow. In second time, we determine the mean of the shadow size over the images from each set. The correction of the discrepancies required measurements on filters using a standard spectrometer and the microscope in transmission mode and reflection mode. To evaluate the capacity of our device to transmit information in frequency domain, its modulation transfer function is evaluated. Multivariate analysis is used to test its capacity to recognize properties of well-known sample. The wavelength 700 nm was chosen to be the reference for the image acquisition, because at this wavelength the images are well contrasted. The measurement made on the filters suggested correction coefficients in transmission mode and reflection mode. The experimental instrument recognized the microsphere’s properties and led to the extraction of the standard transmittance and reflectance spectra.
