During sepsis,neutrophil activation induces endothelial cell(EC)dysfunction partly through neutrophil extracellular trap(NET)release.The triggering receptor expressed on myeloid cell-1(TREM-1)is an orphan immune receptor that amplifies the inflammatory response mediated by Toll-like receptor-4(TLR4)engagement.Although the key role of TLR4 signaling in NETosis is known,the role of TREM-1 in this process has not yet been investigated.Here,we report that TREM-1 potentiates NET release by human and murine neutrophils and is a component of the NET structure.In contrast,pharmacologic inhibition or genetic ablation of TREM-1 decreased NETosis in vitro and during experimental septic shock in vivo.Moreover,isolated NETs were able to activate ECs and impair vascular reactivity,and these deleterious effects were dampened by TREM-1 inhibition.TREM-1 may,therefore,constitute a new therapeutic target to prevent NETosis and associated endothelial dysfunction.

The triggering receptor expressed on myeloid cells-1(TREM-1)is a receptor expressed on innate immune cells.By promoting the amplification of inflammatory signals that are initially triggered by Toll-like receptors(TLRs),TREM-1 has been characterized as a major player in the pathophysiology of acute and chronic inflammatory diseases,such as septic shock,myocardial infarction,atherosclerosis,and inflammatory bowel diseases.However,the molecular events leading to the activation of TREM-1 in innate immune cells remain unknown.Here,we show that TREM-1 is activated by multimerization and that the levels of intracellular Ca 2+release,reactive oxygen species,and cytokine production correlate with the degree of TREM-1 aggregation.TREM-1 activation on primary human monocytes by LPS required a two-step process consisting of upregulation followed by clustering of TREM-1 at the cell surface,in contrast to primary human neutrophils,where LPS induced a rapid cell membrane reorganization of TREM-1,which confirmed that TREM-1 is regulated differently in primary human neutrophils and monocytes.In addition,we show that the ectodomain of TREM-1 is able to homooligomerize in a concentration-dependent manner,which suggests that the clustering of TREM-1 on the membrane promotes its oligomerization.We further show that the adapter protein DAP12 stabilizes TREM-1 surface expression and multimerization.TREM-1 multimerization at the cell surface is also mediated by its endogenous ligand,a conclusion supported by the ability of the TREM-1 inhibitor LR12 to limit TREM-1 multimerization.These results provide evidence for ligand-induced,receptor-mediated dimerization of TREM-1.Collectively,our findings uncover the mechanisms necessary for TREM-1 activation in monocytes and neutrophils.

Recently,machine learning-based technologies have been developed to automate the classification of wafer map defect patterns during semiconductormanufacturing.The existing approaches used in the wafer map pattern classification include directly learning the image through a convolution neural network and applying the ensemble method after extracting image features.This study aims to classify wafer map defects more effectively and derive robust algorithms even for datasets with insufficient defect patterns.First,the number of defects during the actual process may be limited.Therefore,insufficient data are generated using convolutional auto-encoder(CAE),and the expanded data are verified using the evaluation technique of structural similarity index measure(SSIM).After extracting handcrafted features,a boosted stacking ensemble model that integrates the four base-level classifiers with the extreme gradient boosting classifier as a meta-level classifier is designed and built for training the model based on the expanded data for final prediction.Since the proposed algorithm shows better performance than those of existing ensemble classifiers even for insufficient defect patterns,the results of this study will contribute to improving the product quality and yield of the actual semiconductor manufacturing process.

The high demand for lung transplants cannot be matched by an adequate number of lungs from donors. Since fully ex-novo lungs are far from being feasible, tissue engineering is actively considering implantation of engineered lungs where the devitalized structure of a donor is used as scaffold to be repopulated by stem cells of the receiving patient. A decellularized donated lung is treated inside a bioreactor where transport through the tracheobronchial tree (TBT) will allow for both deposition of stem cells and nourishment for their subsequent growth, thus developing new lung tissue. The key concern is to set optimally the boundary conditions to utilize in the bioreactor. We propose a predictive model of slow liquid ventilation, which combines a one-dimensional (1-D) mathematical model of the TBT and a solute deposition model strongly dependent on fluid velocity across the tree. With it, we were able to track and drive the concentration of a generic solute across the airways, looking for its optimal distribution. This was given by properly adjusting the pumps’ regime serving the bioreactor. A feedback system, created by coupling the two models, allowed us to derive the optimal pattern. The TBT model can be easily invertible, thus yielding a straightforward flow/pressure law at the inlet to optimize the efficiency of the bioreactor.

The accurate simulation of disperse two-phase flows,where a discrete particulate condensed phase is transported by a carrier gas,is crucial for many applications;Eulerian approaches are well suited for high performance computations of such flows.However when the particles from the disperse phase have a significant inertia compared to the time scales of the flow,particle trajectory crossing(PTC)occurs i.***.the particle velocity distribution at a given location can become multi-valued.To properly account for such a phenomenon many Eulerian moment methods have been recently proposed in the literature.The resulting models hardly comply with a full set of desired criteria involving:1-ability to reproduce the physics of PTC,at least for a given range of particle inertia,2-well-posedness of the resulting set of PDEs on the chosen moments as well as guaranteed realizability,3-capability of the model to be associated with a high order realizable numerical scheme for the accurate resolution of particle segregation in turbulent flows.The purpose of the present contribution is to introduce a multi-variate Anisotropic Gaussian closure for such particulate flows,in the spirit of the closure that has been suggested for out-of-equilibrium gas dynamics and which satisfies the three criteria.The novelty of the contribution is three-fold.First we derive the related moment system of conservation laws with source terms,and justify the use of such a model in the context of high Knudsen numbers,where collision operators play no role.We exhibit the main features and advantages in terms of mathematical structure and realizability.Then a second order accurate and realizable MUSCL/HLL scheme is proposed and validated.Finally the behavior of the method for the description of PTC is thoroughly investigated and its ability to account accurately for inertial particulate flow dynamics in typical configurations is assessed.

Mapping mountain areas: learning from Global, European and Norwegian perspectives

Defining the spatial extent of mountain areas has long been a challenge. In the present century, the availability of digital elevation models(DEMs) incorporated into geographic information systems(GIS) has allowed the definition of mountain areas based on topographic and other criteria. This paper presents the various delineations of mountains that have been prepared at three scales – global, regional(Europe), and national – and explores the reasons and processes leading to these delineations, and how they have been used. A detailed case study is then presented for Norway. Overall, two types of approaches to mapping mountains have been taken: first, considering mountains per se, based on elevation and/or topography; second, considering them among other categories, e.g., landforms or biogeographical, environmental or landscape zones. All attempts to map mountain areas derive essentially from the objectives of those commissioning and/or undertaking the work; a unitary definition remains unlikely.

Remote photoplethysmography （rPPG） allows remote measurement of the heart rate using low-cost RGB imaging equipment. In this study, we review the development of the field of rPPG since its emergence in 2008. We also classify existing rPPG approaches and derive a framework that provides an overview of modular steps. Based on this framework, practitioners can use our classification to design algorithms for an rPPG approach that suits their specific needs. Researchers can use the reviewed and classified algorithms as a starting point to improve particular features of an rPPG algorithm.

Tracing the Origin of Metasedimentary Rocks in the Tortiya Diamond Region (Northern Côte d’Ivoire)

Metasedimentary rocks from the Tortiya diamondiferous region (northern Côte d’Ivoire), located in the Ivorian Paleoproterozoic domain, were analyzed with the aim of identifying their different petrographic facies, constraining their provenance and tectonic environment, and assessing the effects of sedimentary processes and weathering on the signature of their source rocks. These analyses were carried out with a view to helping trace the origin of Tortiya diamonds, which are generally hosted in these metasediments. Major and trace element geochemical data reveal that these samples are characterized by negative Eu anomaly values, low to moderate weathering index values and a high compositional variation index (ICV). These results led to the definition of these metasediments as immature sediments potentially originating from rapid erosion and sediment deposition from local sources. Chemical alteration index (CIA) values and Al2O3-(CaO* + Na2O)-K2O composition space suggest low to medium degrees of meteoric alteration of the parent rock. These rocks therefore show poorly differentiated compositions from the parent rock and poor sediment sorting. Geochemical data and ratios of immobile elements, e.g. Al2O3/TiO2, Cr/Th, Eu/Eu* and (La/Yb)n, indicate that the clastic materials derive mainly from felsic to intermediate sources. Sedimentary tectonic discrimination diagrams demonstrate that most of the samples from the Tortiya diamond-bearing region were deposited on an active continental margin or in a continental arc-island environment. Although this region has a long history of diamond mining, its petrogenesis remains unresolved, and this petrogeochemical work is a step towards initiating scientific research in the area.
