Since 2014,"Bring Your Own Data"workshops(BYODs)have been organised to inform people about the process and benefits of making resources Findable,Accessible,Interoperable,and Reusable(FAIR,and the FAIRification process).The BYOD workshops'content and format differ depending on their goal,context,and the background and needs of ***-focused BYODs educate domain experts on how to make their data FAIR to find new answers to research ***-focused BYODs promote the benefits of making data FAIR and instruct project managers and policy-makers on the characteristics of FAIRification ***-focused BYODs gather software developers and experts on FAIR to implement or improve software resources that are used to support ***,these BYODs intend to foster collaboration between different types of stakeholders involved in data management,curation,and reuse(*** experts,trainers,developers,data owners,data analysts,FAIR experts).The BYODs also serve as an opportunity to learn what kind of support for FAIRification is needed from different communities and to develop teaching materials based on practical examples and *** this paper,we detail the three different structures of the BYODs and describe examples of early BYODs related to plant breeding data,and rare disease registries and biobanks,which have shaped the structure of the *** discuss the latest insights into making BYODs more productive by leveraging our almost ten years of training experience in these workshops,including successes and encountered ***,we examine how the participants'feedback has motivated the research on FAIR,including the development of workflows and software.

Bacteriological Profile of Infections Encountered in a Pneumology Service in a Moderate-Income country [Pneumology Department of cHU cocody (côte d’Ivoire)]

Introduction: Infections represent a real public health problem aggravating the morbidity and mortality of hospitalized patients. Methods: This was a retrospective study with descriptive purposes over a period of 05 years, in the Pneumology Department of the University Hospital of cocody. Results: The average age in our population was 42 years. We observed a male predo- minance of 64.5%, with a sex ratio of 1.8. Medical history was dominated by HIV infection (23.5%), followed by tuberculosis (15.6%). concerning lifestyle, smoking was found in 38% of cases. Symptoms progressed chronically in 80% of cases. An infectious syndrome was found in 75% of cases. Microbial culture was positive in 42% of cases. Pseudomonas aeruginosa and Klebsiella pneumoniae were encountered in 26.4% of cases each, followed by Echerichia coli in 10.4% of cases. Klebsiella pneumoniae in the first three years topped the list, but gradually. Pseudomonas aeruginosa maintained its leadership over the last three years. Pseudomonas aeruginosa isolates expressed a resistance rate of 9.8% to ceftazidime and 8.1% to imipenem;to aztreonam (36%), ticarcillin (33.3%) and levofloxacin. These strains were susceptible to fosfomycin (100%), mero- penem (96.6%) and amikacin (96%). For isolated strains of Entero- bacteria- ceae, resistance was observed about ticarcillin (83.3%) and amoxicillin clavula- nic acid (71.2%). Streptococcaceae showed resistance to tetracycline (69.2%) and erythromycin (50%). Over the years there has been an increase in re- sistance to amoxicillin-clavulanic acid ceftriaxone. The death rate was 14%. conclusion: The bacterial profile of infections is dominated by germs respon- sible for nosocomial infection with significant mortality.

Extract of codiaeum luzonicum Merr. overcomes multidrug resistance in human colon cancer cells by modulating P-glycoprotein

Objective:To investigate anti-multidrug resistance(MDR)activity and safety of the bioactive fraction(cL11)from codiaeum luzonicum crude leaf ***:cytotoxic activity of cL11 against MDR and nonresistant colon cancer cells was assessed using MTT *** of cell death was investigated by annexin V-propidium iodide staining,TUNEL,and Jc-1 *** examine mechanism of action,the effect on the expression and function of the MDR-implicated protein P-glycoprotein was tested using Western blotting and calcein assay,***:cL11 had an Ec50 of 0.18,1.03 and 38.52µg/mL against HcT-15,HcT-15/Dox and HcT116,*** was mediated by inhibition of P-glycoprotein function and *** mode of cell death involved mitochondrial membrane depolarization and was mostly non-apoptotic at Ec50 concentrations against HcT-15 and HcT-15/***:Fraction cL11 of codiaeum luzonicum induces nonapoptotic cell death in MDR cancer cells by overcoming MDR through inhibition of P-glycoprotein expression and function.

The B7/cD28 families of immune checkpoints play vital roles in negatively or positively regulating immune cells in homeostasis and various *** basic and clinical studies have revealed novel biology of the B7/cD28 families and new therapeutics for cancer *** this review,we discuss the newly discovered KIR3DL3/TMIGD2/HHLA2 pathways,PD-1/PD-L1 and B7-H3 as metabolic regulators,the glycobiology of PD-1/PD-L1,B7x(B7-H4)and B7-H3,and the recently characterized PD-L1/B7-1 *** also cover the tumor-intrinsic and-extrinsic resistance mechanisms to current anti-PD-1/PD-L1 and anti-cTLA-4 immunotherapies in clinical ***,we review new immunotherapies targeting B7-H3,B7x,PD-1/PD-L1,and cTLA-4 in current clinical trials.

The computational simulation of the manufacturing process of lithium-ion battery composite electrodes based on mechanistic models allows capturing the influence of manufacturing parameters on electrode ***,ensuring that these properties match with experimental data is typically computationally *** this work,we tackled this costly procedure by proposing a functional data-driven framework,aiming first to retrieve the early numerical values calculated from a molecular dynamics simulation to predict if the observable being calculated is prone to match with our range of experimental values,and in a second step,recover additional values of the ongoing simulation to predict its final *** demonstrated this approach in the context of the calculation of electrode slurries *** report that for various electrode chemistries,the expected mechanistic simulation results can be obtained 11 times faster with respect to the complete simulations,while being accurate with a R^(2)_(score) equals to 0.96.

New insights into the control of symbiotic nitrogen fixation by carbon supply

The interaction between legumes and rhizobium-type bacteria allows symbiotic nitrogen fixation(SNF).Symbiosis takes place in a new root organ,the nodule,in which the plant host fuels the bacteria differentiated into bacteroids with dicarboxylic acids,enabling their active carbon and energy metabolisms(Figure 1).In addition,the plant maintains the bacteroids in microoxic conditions allowing the nitrogenase to reduce atmospheric nitrogen(N_(2))to ammonia(NH_(3)).Ammonia is not assimilated in the bacteroids but is subsequently transferred into the surrounding plant cells,where it combines with carbon(c)skeletons through the glutamine synthetase/glutamate synthase *** amino acids and/or ureides are finally distributed from the root to the to the whole plant.

The deacetylase SIRT2 contributes to autoimmune disease pathogenesis by modulating IL-17A and IL-2 transcription

Aberrant IL-17A expression together with reduced IL-2 production by effector cD4^(+)T cells contributes to the pathogenesis of systemic lupus erythematosus(SLE).Here,we report that Sirtuin 2(SIRT2),a member of the family of NAD^(+)-dependent histone deacetylases,suppresses IL-2 production by cD4^(+)T cells while promoting their differentiation into Th17 ***,we show that SIRT2 is responsible for the deacetylation of p70S6K,activation of the mTORc1/HIF-1α/RORγt pathway and induction of Th17-cell ***,SIRT2 was shown to be responsible for the deacetylation of c-Jun and histones at the Il-2 gene,resulting in decreased IL-2 *** found that the transcription factor inducible cAMP early repressor(IcER),which is overexpressed in T cells from people with SLE and lupus-prone mice,bound directly to the Sirt2 promoter and promoted its ***-7,a SIRT2 inhibitor,limited the ability of adoptively transferred antigen-specific cD4^(+)T cells to cause autoimmune encephalomyelitis in mice and limited disease in lupus-prone MRL/lpr ***,cD4^(+)T cells from SLE patients exhibited increased expression of SIRT2,and pharmacological inhibition of SIRT2 in primary cD4^(+)T cells from patients with SLE attenuated the ability of these cells to differentiate into Th17 cells and promoted the generation of IL-2–producing T ***,these results suggest that SIRT2-mediated deacetylation is essential in the aberrant expression of IL-17A and IL-2 and that SIRT2 may be a promising molecular target for new SLE therapies.

Supramolecular self-assembly in water based on non-covalent bonding is attracting major attention due to the potential of hydrogels and aqueous polymers in biomedical *** supramolecular polymerization in organic solvents is well established,the key design features,the assembly mechanisms in water and achieving control over the aggregate structures remain ***,we present the assembly and disassembly of geometrical isomers of a stiff-stilbene bis-urea amphiphile(SA)in pure water.A remarkable feature of this system is that the(E)-isomer forms supramolecular polymers in both pure water and organic *** advantage of this unique property,the hydrophobic effect was studied by comparing the supramolecular assembly in both *** assembly process inwater follows an enthalpy-driven nucleation-elongation(cooperative)supramolecular polymerization mechanism with a standard Gibbs free energy(ΔG°=−53 kJ mol^(−1))double the value of the one found in *** attributed this distinctive feature to the hydrophobic effect in ***,we discovered an isomer-dependent assembly process,which can be used to control aggregation in aqueous *** to the substantial geometric difference between(E)-SA and(Z)-SA,we compared their assembly in water to study the influence of different driving forces involved in the *** supramolecular polymerization of(E)-SA was cooperatively influenced by hydrogen bonding,π-stacking,and hydrophobic effects,whereas the assembly of(Z)-SAwasmainly driven by hydrophobic *** a result,the fiber length of(E)-SA in water is much longer than that of(Z)-SA,presenting opportunities for geometrical control of aggregation in aqueousmedia.
