Immunotherapy remains more effective for hematologic tumors than for solid *** of the main challenges to immunotherapy of solid tumors is the immunosuppressive microenvironment these tumors generate,which limits the cytotoxic capabilities of immune effector cells(e.g.,cytotoxic T and natural killer cells).This microenvironment is characterized by hypoxia,nutrient starvation,accumulated waste products,and acidic ***-hijacked cells,such as fibroblasts,macrophages,and T regulatory cells,also contribute to this inhospitable microenvironment for immune cells by secreting immunosuppressive cytokines that suppress the antitumor immune response and lead to immune ***,there is a strong interest in developing new drugs and cell formulations that modulate the tumor microenvironment and reduce tumor cell immune *** systems(mPSs)are versatile tools that may accelerate the development and evaluation of these therapies,although specific examples showcasing the potential of mPSs remain *** in microtechnologies have led to the development of sophisticated microfluidic devices used to recapitulate tumor *** resulting models,also known as microphysiological systems(mPSs),are versatile tools with which to decipher the molecular mechanisms driving immune cell antitumor cytotoxicity,immune cell exhaustion,and immune cell exclusion and to evaluate new targeted ***,we review existing microphysiological platforms to study immuno-oncological applications and discuss challenges and opportunities in the field.

Effects of a 3-month vigorous physical activity intervention on eating behaviors and body composition in overweight and obese boys and girls

Purpose: This study aims to analyze the effects of a 3-month vigorous physical activity(VPA) intervention on eating behavior and body composition in overweight and obese children and ***: Forty-seven participants(7–16 years) took part in the study: 28 were assigned to the intervention group(IG)(10 boys and 18 girls) and19 in a control group(cG)(8 boys and 11 girls). Body composition(dual-energy X-ray absorptiometry), anthropometrics(body mass, height, and body mass index(BmI)), and eating behavior traits(Three-Factor Eating Questionnaire-R21 c) were determined before and after the VPA ***: A decrease in the percentage of body fat and BmI(-2.8% and-1.8%, respectively), and an increase in most lean mass variables were found in the IG(all p ≤ 0.05). In relation to the eating behavior traits, IG subjects showed a 14% reduction in the Emotional Eating score(p = 0.04),while cognitive Restraint score did not change after the VPA intervention. The baseline factors of the questionnaire predicted changes in body mass and fat mass variables only in the ***: A 3-month VPA intervention influenced eating behaviors of overweight or obese young, especially the Emotional Eating factor, in the presence of favorable body composition changes.

Simple statistical models for relating river discharge with precipitation and air temperature——case study of River Vouga （Portugal）

Simple statistical models were developed to relate available meteorological data with daily river discharge （RD） for rivers not influenced by melting of ice and snow. In a case study of the Vouga River （Portugal）, the RD could be determined by a linear combination of the recent （PR） and non-recent （PNR） atmospheric precipitation history. It was found that a simple linear model including only PR and PnR cannot account for low RD. The model was improved by including non-linear terms of precipitation that accounted for the water loss. Additional improvement of the models was possible by including average monthly air temperature （7）. The best model was robust when up to 60% of the original data were randomly removed. The advantage is the simplicity of the models, which take into account only PR, PNR and T. These models can provide a useful tool for RD estimation from current meteorological data.

An important factor for the sustainability of soils highly susceptible to degradation is the use of monitoring tools that promptly and realistically reflect changes imposed on soil by different cropping systems. To select soil quality indicator variables in sugarcane (Saccharum officinarum L.) production areas that fulfill the criteria of sensitivity to management practices and between-season consistency in the management discrimination, ten composite soil samples (0–10 cm) were collected in July 2005 (rainy season) and again in march 2006 (dry season) from areas under cultivation of organic sugarcane (OS), green sugarcane (GS), burned sugarcane (BS) and from an adjacent native forest (NF) area at Usina Triunfo, Boca da mata, Alagoas, Brazil. microbial biomass-c (mBc), total organic c (TOc), soil enzyme activity expressed as the rate of fluorescein diacetate (FDA) hydrolysis, mean weight diameter of water-stable soil aggregates (mWD), and percentage of water-stable macroaggregates (PWSA) were analyzed. Although mBc and TOc were higher in NF than in the cultivated areas, no differences were observed in these c pools between the three sugarcane systems. The response of FDA to the site management was dependent on the sampling time. In the rainy period, the activity followed the order: NF > OS > GS > BS, whereas in the dry season, only NF differed from the other treatments. Irrespective of the sampling time, mWD and PWSA decreased in the order NF > OS = GS > BS. The variables mWD and PWSA are quite sensitive for discriminating between site management histories regardless the sampling season.

Root proliferation in native perennial grasses of arid Patagonia,Argentina

Pappophorum vaginatum is the most abundant c4 perennial grass desirable to livestock in rangelands of northeastern Patagonia, Argentina. We hypothesized that （1） defoliation reduce net primary productivity, and root length density and weight in the native species, and （2） root net primary productivity, and root length density and weight, are greater in P. vaginatum than in the other, less desirable, native species （i.e., Aristida spegazzinfi, A. subulata and Sporobolus cryptandrus）. Plants of all species were either exposed or not to a severe defoliation twice a year during two growing seasons. Root proliferation was measured using the cylinder method. cylindrical, iron structures, wrapped up using nylon mesh, were buried diagonally from the periphery to the center on individual plants. These structures, initially filled with soil without any organic residue, were dug up from the soil on 25 April 2008, after two successive defoliations in mid-spring 2007. During the second growing season （2008-2009）, cylinders were destructively harvested on 4 April 2009, after one or two defoliations in mid- and/or late-spring, respectively. Roots grown into the cylinders were obtained after washing the soil manually. Defoliation during two successive years did reduce the study variables only after plants of all species were defoliated twice, which supported the first hypothesis. The greater root net primary productivity, root length den- sity and weight in P. vaginatum than in the other native species, in support of the second hypothesis, could help to explain its greater abundance in rangelands of Argentina.

Doped graphene/carbon black hybrid catalyst giving enhanced oxygen reduction reaction activity with high resistance to corrosion in proton exchange membrane fuel cells

Nitrogen doping of the carbon is an important method to improve the performance and durability of catalysts for proton exchange membrane fuel cells by platinum–nitrogen and carbon–nitrogen bonds. This study shows that p-phenyl groups and graphitic N acting bridges linking platinum and the graphene/carbon black(the ratio graphene/carbon black = 2/3) hybrid support materials achieved the average size of platinum nanoparticles with(4.88 ± 1.79) nm. It improved the performance of the lower-temperature hydrogen fuel cell up to 0.934 W cm^(-2) at 0.60 V, which is 1.55 times greater than that of commercial Pt/c. Doping also enhanced the interaction between Pt and the support materials, and the resistance to corrosion, thus improving the durability of the low-temperature hydrogen fuel cell with a much lower decay of 10 mV at 0.80 A cm^(-2) after 30 k cycles of an in-situ accelerated stress test of catalyst degradation than that of 92 mV in Pt/c, which achieves the target of Department of Energy(mV). meanwhile,Pt/Nr EGO_(2)-cB_(3) remains 78% of initial power density at 1.5 A cm^(-2) after 5 k cycles of in-situ accelerated stress test of carbon corrosion, which is more stable than the power density of commercial Pt/c, keeping only 54% after accelerated stress test.

Nucleic acids-based therapies have recently developed as next-generation agents for treating and preventing viral infection, cancer, and genetic disorders, but their use is still limited due to its relatively poor delivery into targeted cells. We designed and synthesized new amphiphilic amino acid derivatives (cysteine-based) of low molecular weight, formed by the same pentapeptide (AG2: WWcOO) N-acylated, with different hydrophobic chains containing from 12 to 18 carbons, named AG2-cn (N), which dimerize by oxidation in the presence of pLenti-cmV-GFP Puro plasmid (P) in the respective gemini. We determined transfection efficiency, critical micelle concentration, particle size, ζ-potential and cytotoxicity for the derivatives obtained. We found that all the synthesized compounds were active for DNA delivery and had greater ability to transfect cHO-K1 cells. In particular, AG2-c18 is a promising carrier for gene delivery because it showed no cytotoxicity and its activity was greater than or equal to the commercial actives currently used.

Thin hybrid films of ZnO/eosin-Y were prepared by electrodeposition at-0.8 and-0.9 V in aqueous and non-aqueous baths at temperatures ranging from 40 to 90 ℃ with dye concentrations of 100 and 400 μmol·*** films were characterized by X-ray diffraction (XRD),scanning electron microscopy (SEm),energy-dispersive X-ray analysis (EDX),and absorption *** films prepared in a non-aqueous bath were non-porous and did not adsorb dye molecules on their ***,the films grown in aqueous media were porous in nature and adsorbed dye during the deposition of *** growth of the film along the (002) face was observed,and the highest crystallinity was achieved when the film was deposited at 60 ℃.The maximum absorption was achieved for the films grown at 60 to 70 ℃,a deposition potential of-0.9 V,and a dye concentration of 100 μmol·L-1.

Loss of mgat5a-mediated N-glycosylation stimulates regeneration in zebrafish

Background:We are using genetics to identify genes specifically involved in hearing *** a large-scale genetic screening,we identified mgat5a,a gene in the N-glycosylation biosynthesis pathway whose activity negatively impacts hair cell ***:We used a combination of mutant analysis in zebrafish and a hair cell regeneration assay to phenotype the loss of mgat5a activity in *** used pharmacological inhibition of N-glycosylation by *** also used over-expression analysis by mRNA injections to demonstrate how changes in N-glycosylation can alter cell ***:We found that mgat5a was expressed in multiple tissues during zebrafish embryo development,particularly enriched in neural tissues including the brain,retina,and lateral line *** mgat5a insertional mutation and a cRISPR/cas9-generated truncation mutation both caused an enhancement of hair cell regeneration which could be phenocopied by pharmacological inhibition with *** addition to hair cell regeneration,inhibition of the N-glycosylation pathway also enhanced the regeneration of lateral line axon and caudal *** analysis showed that N-glycosylation altered the responsiveness of TGF-beta ***:The findings from this study provide experimental evidence for the involvement of N-glycosylation in tissue regeneration and cell signaling.

Electron confinement Effect of Laser Dyes within Dendritic Structures

Dendrimers are a novel class of nanometric-size macromolecules with a regular tree-dimensional like array of branch units.1,2 Their synthetic availability in a wide range of sizes combined with their peculiar architecture makes them versatile building blocks for a wide range of potential applications.3 Some years ago, meijer and co-workers reported that the modification of terminal amine functionalities of a fifth generation poly(propyleneimine) dendrimer (DAB-dendr-(NH2)64) with bulky substituents, (typically N-t-BOc protected phenylalanine), results in the formation of the so-called “dendritic box” (DAB-dendr-(NH-t-BOc-L-Phe)64).4 Within this macromolecular structure it is possible to encapsulate a variety of guest molecules due to the existence of internal cavities in the core. The photophysical properties of the guests can be modulated by the innovative electron confinement effect. In this respect, we wish to report that the emission frequency of organic dyes can be easily modulated by encapsulation in a dendritic box. The emission bands of dye molecules incorporated into a dendrimer can effectively be red shifted with respect to their emission in solution and contrary to other confined spaces of considerable hardness, the magnitude of this shifting can be regulated under appropriate experimental conditions. This peculiar effect could have unprecedented applications in the development of supramolecular devices relating to the frequency tuning of organic laser dyes.
