IL-6 prevents Th2 cell polarization by promoting SOCS3-dependent suppression of IL-2 signaling

Defective interleukin-6 (IL-6) signaling has been associated with Th2 bias and elevated IgE levels. However, the underlying mechanism by which IL-6 prevents the development of Th2-driven diseases remains unknown. Using a model of house dust mite (HDm)-induced Th2 cell differentiation and allergic airway inflammation, we showed that IL-6 signaling in allergen-specific T cells was required to prevent Th2 cell differentiation and the subsequent IgE response and allergic inflammation. Th2 cell lineage commitment required strong sustained IL-2 signaling. We found that IL-6 turned off IL-2 signaling during early T-cell activation and thus inhibited Th2 priming. mechanistically, IL-6-driven inhibition of IL-2 signaling in responding T cells was mediated by upregulation of Suppressor Of Cytokine Signaling 3 (SOCS3). This mechanism could be mimicked by pharmacological Janus Kinase-1 (JAK1) inhibition. Collectively, our results identify an unrecognized mechanism that prevents the development of unwanted Th2 cell responses and associated diseases and outline potential preventive interventions.

The ancestral cultures have described many therapeutic properties of garlic,therefore,it is of central interest to elucidate the molecular basis explaining this millenary empirical ***,it has been demonstrated a neuroprotective effect of allicin–a phytochemical present in garlic-linked to oxidative-inflammatory *** improved neuronal injury by heat shock protein 70(Hsp70)and inducible nitric oxide synthase(iNOS)***,allicin exerts renal protection involving a possible angiotensin type 1 receptor(AT1)*** connection,AT1 overexpression has been recognized as a central deleterious factor in many brain ***,there are no studies that evaluate AT1-Hsp70-iNOS interaction as a mechanism linked to ***,our central aim is to evaluate if the allicin protective effect is associated with an AT1-Hsp70-iNOS counterbalance *** this study,a murine microglial cell line(BV-2)was injured with lipopolysaccharides and treated or not with ***,it was evaluated cell viability,proinflammatory cytokine levels,cellular oxidative stress,iNOS,Hsp70,and AT1 protein expression(cellular and mitochondrial fractions),nitrite levels,and protein-protein *** results demonstrated that allicin could prevent neuronal injury due to a reduction in oxidative stress and inflammatory status mediated by an AT1-Hsp70-iNOS counterbalance axis linked to direct protein-protein interaction.

Silicone rubber films were modified by the consecutive grafting of 2-(diethylamino)ethyl methacrylate (DEAEmA) and N-vinylcaprolactam (NVCL) using direct method on two steps with gamma-rays. The effect of absorbed dose and monomer concentration on grafting degree was determined. The grafted samples were verified by FTIR-ATR spectroscopy and swelling;thermal properties were analyzed by DSC and TGA. The stimuli-responsive behavior was studied by swelling and/or DSC. Thermo- and pH-sensitive films of (PP-g-DEAEmA)-g-NVCL presented a pH critical at 3.2 and LCST around 63.5℃.

The poor gas selectivity problem has been a long-standing issue for miniaturized chemical-resistor gas *** electronic nose(e-nose)was proposed in the 1980s to tackle the selectivity issue,but it required top-down chemical functionalization processes to deposit multiple functional ***,we report a novel gas-sensing scheme using a single graphene field-effect transistor(GFET)and machine learning to realize gas selectivity under particular conditions by combining the unique properties of the GFET and e-nose *** of using multiple functional materials,the gas-sensing conductivity profiles of a GFET are recorded and decoupled into four distinctive physical properties and projected onto a feature space as 4D output vectors and classified to differentiated target gases by using machine-learning *** single-GFET approach coupled with trained pattern recognition algorithms was able to classify water,methanol,and ethanol vapors with high accuracy quantitatively when they were tested ***,the gas-sensing patterns of methanol were qualitatively distinguished from those of water vapor in a binary mixture condition,suggesting that the proposed scheme is capable of differentiating a gas from the realistic scenario of an ambient environment with background *** such,this work offers a new class of gas-sensing schemes using a single GFET without multiple functional materials toward miniaturized e-noses.

Carbon nanomaterials offer excellent prospects as therapeutic agents,and among them,graphene quantum dots(GQDs)have gained considerable interest thanks to their aqueous solubility and intrinsic fluorescence,which enable their possible use in theranostic approaches,if their biocompatibility and favorable pharmacokinetic are *** prepared ultra-small GQDs using an alternative,reproducible,top-down synthesis starting from graphene oxide with a nearly 100%*** materials were tested to assess their safety,demonstrating good biocompatibility and ability in passing the ultrafiltration barrier using an in vitro *** leads to renal excretion without affecting the ***,we studied the GQDs in vivo biodistribution confirming their efficient renal clearance,and we demonstrated that the internalization mechanism into podocytes is ***,considering the reported characteristics,it appears possible to vehiculate compounds to kidneys by means of GQDs,overcoming problems related to lysosomal degradation.

AIm: To study anti-Epstein-Barr virus(EBV) Ig G antibodies in Crohn′s disease in relation to treatment, immune cells, and prior tonsillectomy/***: This study included 36 CD patients and 36 healthy individuals(controls), and evaluated different clinical scenarios(new patient, remission and active disease), previous mucosa-associated lymphoid tissue removal(tonsillectomy and appendectomy) and therapeutic regimens(5-aminosalicylic acid, azathioprine, anti-tumor necrosis factor, antibiotics, and corticosteroids). T and B cells subsets in peripheral blood were analyzed by flow cytometry(markers included: CD45, CD4, CD8, CD3, CD19, CD56, CD2, CD3, TCRαβ and TCRγδ) to relate with the levels of anti-EBV Ig G antibodies, determined by enzyme-linked immunosorbent ***: The lowest anti-EBV Ig G levels were observed in the group of patients that were not in a specific treatment(95.4 ± 53.9 U/m L vs 131.5 ± 46.2 U/m L, P = 0.038). The patients that were treated with 5-aminosalicylic acid showed the highest anti-EBV Ig G values(144.3 U/m L vs 102.6 U/m L, P = 0.045). CD19+ cells had the largest decrease in the group of CD patients that received treatment(138.6 vs 223.9; P = 0.022). The analysis of anti-EBV Ig G with respect to the presence or absence of tonsillectomy showed the highest values in the tonsillectomy group of CD patients(169.2 ± 20.7 U/m L vs 106.1 ± 50.3 U/m L, P = 0.002). However, in the group of healthy controls, no differences were seen between those who had been tonsillectomized and subjects who had not been operated on(134.0 ± 52.5 U/m L vs 127.7 ± 48.1 U/m L, P = 0.523).CONCLUSION: High anti-EBV Ig G levels in CD are associated with 5-aminosalicylic acid treatment, tonsillectomy, and decrease of CD19+ cells.
