Insights into glycan biosynthesis in chemically-induced hepatocellular carcinoma in rats: A glycomic analysis

AIM： To evaluate the qualitative and quantitativechanges in N-linked glycosylation, which occurredin association with diethyl nitrosamine-inducedhepatocellular carcinoma （HCC） in ***： Liver tissues of （1） normal （non-tumorbearing）rats; and （2） tumor-bearing rats; were collectedand were used for histological and GlycanMap？ ***, GlycanMap？ analysis is a high-throughputassay that provides a structural and quantitativereadout of protein-associated glycans using a unique,automated 96-well assay technology coupled tomatrix-assisted laser desorption/ionization time-offlightmass spectrometry and custom *** studies were carried out to ensure thedevelopment of HCC in the tested ***： The N-glycomic analysis revealed 5glycans; Glc1Man9GlcNAc2, Gal2Man3GlcNac4Fuc1Neu1,Man4G l c N a c 2, G a l 2Man3G l c N a c 4Neu3OAc 3, andMan3GlcNac5Fuc1, which showed significant changesin rat HCC tissues when compared with normal livertissues. Four glycans were increased （P 〈 0.05） andGlc1Man9GlcNAc2 was decreased （5.89 ± 0.45 vs 3.54± 0.21, P 〈 0.01） in HCC tissues compared to normal liver tissues. An increase （66.5 ± 1.05 vs 62.7 ± 1.1,P 〈 0.05） in high-mannose structures in HCC rats wasobserved compared to normal rats. Importantly, HCCrats showed an increase （P 〈 0.05） in both tumorassociatedcarbohydrates and in branched glycans. Thechanges in glycans correlated well with glycan flowchanges reported in the glycan biosynthetic pathway,which indicates the importance of enzyme activitiesinvolved in glycan synthesis at different ***： The reported HCC-associated changesin glycan flow and subcellular localization explain theincrease in high mannose glycans and siayl Lewisglycans common in HCC liver tissues.

最近我们实验室发展了激光诱导击穿光谱技术(LIBS)用于原位、实时诊断磁约束核聚变装置的第一壁。在模拟EAST托卡马克运行的环境下,研究了第一壁材料钨、钼的LIBS特性。为进一步压制背景噪声,增强LIBS光谱的灵敏度、探测限及分辨率,我们采用偏振分辨激光诱导击穿光谱技术(PRLIBS)来抑制由等离子产生的连续光谱背景。发现PRLIBS技术可较好抑制连续背景提高信号背景比,其原因是因为激光等离子体的连续背景具有很强的偏振特性。同时,我们还研究了PRLIBS发射光谱强度和环境气压与入射激光的偏振方向的关系,发现PRLIBS发射光谱强度不依赖与入射激光的偏振方向。实验中,我们还进一步研究了PRLIBS信号强度和检偏器角度的关系,发现PRLIBS信号随检偏器变化符合马吕斯定律。最后,我们对比研究了纳秒PRLIBS和飞秒PRLIBS对激光等离子体连续背景的抑制作用,发现fs-PRLIBS技术对连续背景的抑制作用更明显,其原因是因为飞秒激光的功率密度高,激发出的等离子体各向异性性更强,致使激光等离子体连续背景的极化率更高,此外,fs-PRLIBS的弱基体效应,也是一个重要原因。

Contamination of Heavy Metals (Lead, Zinc, Magnesium and Manganese) Concentrations in Human Eyes

The aim of our study is to measure the concentration of Lead, Zinc, Magnesium and manganese, toxic/transition metals in human lens. The analyzed samples were categorized according to their personal habits (smoking, food, drinking and medical history). Demographic and laboratory characteristics of patients, the patients were twenty six males and twenty females. The comparison of trace elements between patients and control revealed significant increase of lead and manganese levels in patients and significant decrease of magnesium while there was few significant difference in zinc concentration among them.

Cadmium Toxicity in Plants and Role of Mineral Nutrients in Its Alleviation

Cadmium (Cd) is a toxic heavy metal that enters the environment through various anthropogenic sources, and inhibits plant growth and development. Cadmium toxicity may result from disturbance in plant metabolism as a consequence of disturbance in the uptake and translocation of mineral nutrients. Plant nutrients and Cd compete for the same transporters and, therefore, presence of Cd results in mineral nutrients deficiency. The optimization of mineral nutrients under Cd stress could reduce Cd toxicity by greater availability at the transport site resulting in reduced accumulation of Cd, and could also alleviate Cd-induced toxic effects by enhancing biochemical reactions and physiological processes in plants. In the present review the role of plant macro, micro and beneficial elements in alleviating Cd stress in crop plants is discussed.

Here,we use multi-type feature fusion and selection to predict COVID-19 infections on chest computed tomography(CT)*** scheme operates in four ***,we prepared a database containing COVID-19 pneumonia and normal CT *** images were retrieved from the Radiopaedia COVID-19 *** images were divided into training and test sets in a ratio of 70:***,multiple features were extracted from the training *** used canonical correlation analysis to fuse the features into single vectors;this enhanced the predictive *** next implemented a genetic algorithm(GA)in which an Extreme Learning Machine(ELM)served to assess GA *** on the ELM losses,the most discriminatory features were selected and saved as an ELM *** images were sent to the model,and the best-selected features compared to those of the trained model to allow nal *** employed the collected chest CT *** best predictive accuracy of the ELM classier was 93.9%;the scheme was effective.

Weed Control Practices and Varying Sowing Dates Effects on Seed Production of Pearl Millet (<i>Pennisetum americanum</i>L.) under Semi-Arid Environment

Peral millet being drought tolerant has substantial potential to contribute in food security ensuring the food, fodder and nutritional value in different Asian and African countries. Susceptibility to abiotic and biotic factors and low productivity are the main reasons for decreasing productivity and area of millets. In this context, evaluation of the effect of weed control practices and varying sowing dates on grain yield of kharif season grown pearl millet (Pennisetum americanum L.) was demonstrated at post graduate agriculture research station, University of Agriculture, Faisalabad during 2015. Forage pearl millet was sown at three different sowing dates i.e. mid-June, end of June and mid-July and four weed control practices viz. weedy check (no weeding), twice hoeing at 15 and 30 days after sowing (DAS), weed control using herbicides i.e. application of Atrazine (Awax 38 SC) @330 g a.i. ha-1 at 15 DAS, and twice foliar applications of 10% Sorghum water extract (Sorgaab) (at 15 and 30 DAS). The experiment was laid out in randomized complete block design (RCBD) under split plot arrangement, comprising of three replications. The treatments with varying sowing dates were randomized in main plots and weed control practices were in subplots. Results showed that the highest plant height (279.51 cm), leaf area (2777.80 cm2), fresh weight of leaves per plant (155.57 g), maximum number of grains per head (3162.0) and grain yield (3419.7 kg·ha-1) were obtained in the treatment combination of 30th June sowing × twice weed hoeing (at 15 and 30 DAS) while, maximum 1000-grain weight (8.45 g) was observed in treatments where weeds were controlled by hoeing (at 15 and 30 DAS). Moreover, cultural weed control practices reduce significantly weed density, fresh and dry Wight of weeds. In sum, it is concluded that to reduce the weed-crop competition and to gain higher productivity of pearl millet, field should be weed free 20 - 45 days after sowing.

Evaluation of Two Highly Porous Microcrystalline Biphasic Calcium Phosphate-Based Bone Grafts for Bone Regeneration: An Experimental Study in Rabbits

A biphasic bone grafting biomaterial based on a mixture of calcium phosphates and beta-tricalcium phosphate (β-TCP) phases with high nanoporosity was synthesized. The synthesis route was based on calcium phosphate composition and the incorporation of glycolic acid as a pore former, giving a material composed of 97% β-TCP and 3% calcium orthophosphates (CaPO4). An in vitro study of the purity, microstructure, crystalline domain, and pores size for the material obtained was performed by SEM analysis as well as full structural characterization. The region of interest related to the surface was determined by the specific surface area measured with the BET method. In vivo evaluation of bone response was performed by implanting the new low-cost biphasic manufacturing material synthesized in this work, which was compared with a biphasic material of similar chemical and microstructural composition existing in the commercial market and with higher cost called Synergy Odontit® β-TCP. The materials were implanted separately into 5 mm diameter defects in the tibias of New Zealand White rabbits at 30, 60, and 90 days. The results obtained showed that the host tissue well accepted the new biphasic material;the presence of new bone formation was observed. A more complete resorption was observed for the new microcrystalline biphasic material compared to for a commercial β-TCP material.

Neutron Induced Fission Track Estimation of Uranium Concentration and Its Associated Health Hazards in Drinking Water of the Faisalabad Industrial City

Drinking water plays a major role regarding quality of human life because polluted water is the main cause of many types of diseases. Besides other pollutants, high concentration of uranium above a certain level in drinking water is also hazardous. As water and food are the main sources of uranium intake, it is absolutely imperative that their level of concentrations is monitored regularly for safety of the general public. In this regard, trace amount of uranium in drinking water samples collected from different locations of Faisalabadcity has been carried out using Neutron Induced Fission Track Technique. The water samples along with the standard of known uranium concentration were poured and dried over Lexan track detectors and then irradiated with thermal neutrons in Reactor. After etching, the tracks produced in the detectors as a result of 235U (n, f) reaction were counted under an optical microscope. The uranium concentration was determined by counting and comparing the observed fission track density in the samples and the standard. The observed uranium concentration in the studied water samples varied from (1.04 ± 0.30) μg·L-1 to (21.08 ± 2. 95) μg·L-1 with an average value of (7.39 ± 1.30) μg·L-1 and is found to be within safe limits as far as uranium related health hazards are concerned.
