Germinal centers(GCs)are essential for the establishment of long-lasting antibody *** B cells rely on post-transcriptional RNA mechanisms to translate activation-associated transcriptional programs into functional changes in the cell ***,the critical proteins driving these key mechanisms are still ***,we show that the RNA binding proteins TIA1 and TIAL1 are required for the generation of long-lasting GC ***1-and TIAL1-deficient GC B cells fail to undergo antigen-mediated positive selection,expansion and differentiation into B-cell clones producing high-affinity ***,TIA1 and TIAL1 control the transcriptional identity of dark-and light-zone GC B cells and enable timely expression of the prosurvival molecule ***,we demonstrate here that TIA1 and TIAL1 are key players in the post-transcriptional program that selects high-affinity antigen-specific GC B cells.

Jet grouting is one of the most popular soil improvement techniques,but its design usually involves great uncertainties that can lead to economic cost overruns in construction *** high dispersion in the properties of the improved material leads to designers assuming a conservative,arbitrary and unjustified strength,which is even sometimes subjected to the results of the test *** present paper presents an approach for prediction of the uniaxial compressive strength(UCS)of jet grouting columns based on the analysis of several machine learning algorithms on a database of 854 results mainly collected from different research *** selected machine learning model(extremely randomized trees)relates the soil type and various parameters of the technique to the value of the compressive *** the complex mechanism that surrounds the jet grouting process,evidenced by the high dispersion and low correlation of the variables studied,the trained model allows to optimally predict the values of compressive strength with a significant improvement with respect to the existing ***,this work proposes for the first time a reliable and easily applicable approach for estimation of the compressive strength of jet grouting columns.

NGLY1 deficiency is an ultra-rare autosomal recessively inherited disorder. Characteristic symptoms include among others, developmental delays, movement disorders, liver function abnormalities, seizures, and problems with tear formation. Movements are hyperkinetic and may include dysmetric, choreo-athetoid, myoclonic and dystonic movement elements. To date, there have been no quantitative reports describing arm movements of individuals with NGLY1 deficiency. This report provides quantitative information about a series of arm movements performed by an individual with NGLY1 deficiency and an aged-matched neurotypical participant. Three categories of arm movements were tested: 1) open ended reaches without specific end point targets;2) goal-directed reaches that included grasping an object;3) picking up small objects from a table placed in front of the participants. Arm movement kinematics were obtained with a camera-based motion analysis system and “initiation” and “maintenance” phases were identified for each movement. The combination of the two phases was labeled as a “complete” movement. Three-dimensional analysis techniques were used to quantify the movements and included hand trajectory pathlength, joint motion area, as well as hand trajectory and joint jerk cost. These techniques were required to fully characterize the movements because the NGLY1 individual was unable to perform movements only in the primary plane of progression instead producing motion across all three planes of movement. The individual with NGLY1 deficiency was unable to pick up objects from a table or effectively complete movements requiring crossing the midline. The successfully completed movements were analyzed using the above techniques and the results of the two participants were compared statistically. Spain is the country with the most Biosphere Reserves in the world,52,and Asturias,with 7,ranks first among the single-province Autonomous *** tourist interest in these natural spaces is increasing,but little or nothing is known about how the residents of these natural spaces perceive the tourist *** main objective of this research was to evaluate the residents’perceived changes in protected mountain areas in Asturias concerning the environmental,sociocultural,and economic dimensions related to tourism activity with survey methods and various quantitative *** findings obtained were the following:firstly,there were significant differences between the residents’sociodemographic characteristics and their perceptions of tourism impacts;secondly,there were positive relationships between the residents’perceived tourism economic,environmental and sociocultural impacts and their general attitude towards tourism development;and thirdly,the more positive the residents’attitude towards tourism development,the more likely residents actively support its further *** findings indicate the willingness of residents to get involved and participate in the tourism activity,so policymakers should implicate residents in decisionmaking on local tourism *** conclusion,long-term monitoring of residents’evolution of their perceptions and attitudes is necessary to achieve sustainable tourism development.

This study aims to analyze rapeseed oil’s effectiveness as a coolant of a high-voltage direct current(HVdC)converter transformer while comparing it with mineral *** employed a numerical model,assisted by Computational Fluid dynamics(CFd)tools,to simulate the convective heat transfer inside a HVdC converter transformer,which is an oil-directed air forced(OdAF)*** model seeks to obtain the temperature and velocity fields at the critical points of the fluid *** simulation domain reproduces a 2d cross-sectional model in the center of the *** results show that rapeseed oil requires more time to gain heat from the core and windings due to its lower thermal ***,the average temperature of rapeseed and mineral oils will not significantly differ once the channel temperature reaches a stable *** indicates that the rapeseed oil can effectively replace mineral oil under the same heating ***,a slower flow of the rapeseed oil allows the fluid to smoothly enter the cooling ***,the rapeseed oil cools these channels by pushing the fluid in them and absorbs heat from the sources earlier than mineral *** improvement in cooling indicates that rapeseed oil is better than mineral oil as a coolant and electrical insulating fluid for power transformers.

Plants experience different abiotic/biotic stresses,which trigger their molecular machinery to cope with them. Besides general mechanisms prompted by many stresses, specific mechanisms have been introduced to optimize the response to individual threats. However, these key mechanisms are difficult to identify. Here, we introduce an in-depth species-specific transcriptomic analysis and conduct an extensive meta-analysis of the responses to related species to gain more knowledge about plant responses. The spider mite Tetranychus urticae was used as the individual species, several arthropod herbivores as the related species for meta-analysis, and Arabidopsis thaliana plants as the common host. The analysis of the transcriptomic data showed typical common responses to herbivory, such as jasmonate signaling or glucosinolate biosynthesis. Also, a specific set of genes likely involved in the particularities of the Arabidopsis-spider mite interaction was discovered. The new findings have determined a prominent role in this interaction of the jasmonateinduced pathways leading to the biosynthesis of anthocyanins and tocopherols. Therefore, tandem individual/general transcriptomic profiling has been revealed as an effective method to identify novel relevant processes and specificities in the plant response to environmental stresses.

Trace elements, isotopic modeling and U-Th-Pb SHRIMP zircon age constraints are used to reconstruct the eruption history and magmatic processes of the Piedra Parada Caldera. In the early Eocene, the crystal-poor Barda Colorada ignimbrite(BCI), having >>15% micro-porphyritic crystals with respect to magmatic components, erupted a volume estimated in more than 300 km^3. The Piedra Parada caldera is located in the Patagonian Andes foreland, at the southern end of the calderas field of the Pilcaniyeu Volcanic Belt(PVB). This belt is related to an extensional tectonic setting as a result of the collision of the Farallon-Aluk ridge with South America, which enabled the development of a transform ocean/continental plate margin followed by the detachment of the Aluk plate and the opening of a slab window. The BCI extra-caldera Plateau is a >100 m thick deposit, having a lower unit with high silica(Si O_2> 76 wt.%),potassium poor rhyolitic composition(trondhjemitic like magma), and an upper unit with normal to high potassium rhyolitic composition(granitic like magma). A trace elements modeling of the BCI units shows that the BCI lower and upper units did not evolve from fractionation or immiscibility in the shallow magma reservoir. The BCI also have a primitive isotopic signature(initial87 Sr/86 Sr =0.7031-0.7049 and ε_(Nd)= +3.4 to +3.65). Thus, tectonic, compositional and isotopic constraints suggest the fast ascent of high silica magmas to a shallow reservoir, and point to an upper mantle origin for these rhyolitic magmas in a transitional(Orogenic-Anorogenic) tectono-magmatic setting. U-Th-Pb SHRIMP zircon crystallization ages of the Syn-caldera stage BCI units(56 -51.5 Ma) show a protracted life of 5 Ma for this caldera reservoir. The age of 52.9 ± 0.3 Ma is considered the best fit for the possible maximum age for the caldera collapse.
