Analysis of Commercial Assays for the detection of SARS-CoV-2 Antibodies or Antigens

Background: COVId-19 produced by SARS-CoV-2 infection has spread worldwide. There is a growing need for immunological assays to detect viral specific antibodies or viral antigen. Current standard of diagnosis is reverse-transcriptase polymerase chain reaction (RT-PCR) in nasopharyngeal swabs. However serological tests can be used to determine previous exposure to the virus and complement the diagnosis. IgM and IgG SARS-CoV-2 specific antibodies can be detected as early as one week after infection and assays can be useful to test large groups of individuals. This work revised the available information concerning assays that detect antibodies and antigens for SARS-CoV-2. Methods: Three sources of information were used: technical data sheets (TdS) web pages of the company’s products and published articles in Pubmed with reference to the use of diagnostic kits. All the information was revised until April 5th 2020. Results: There were 226 tests coming from 20 countries mainly from China. TdS were found only in 50 (22.1%). Most assays detect specific antibodies (n 180) based on immunochromatography methods (n 110) and use blood-derived samples (n 105). Assays for antibodies detection measured mainly IgM/IgG (n 112) and the most common procedure time was d as sensitivity and specificity was found only in 18.6% (n 42) of the assays. The majority of the tests are currently for in vitro diagnosis (IVd). A total 165 articles were found on PubMed 15 were included and only 4 used the commercial kits reviewed. Conclusions: due to the urgency of producing diagnostic tests for SARS-CoV-2 there is a broad offer of kits. Many tests need additional information for their application. The data collected may be useful in the selection of assays but more and higher quality information is needed.

Connexin 43 (CX43) is one of the major components of gap junctions, the structures responsible for the intercellular communication and transmission of the electrical impulse in the left ventricle. There is limited information on the histological changes of CX43 with age and their effect on electrophysiology, especially in humans. Here, we analyzed left ventricular biopsies from living donors starting at midlife to characterize age-related CX43 remodeling. We assessed its quantity, degree of lateralization, and spatial heterogeneity together with fibrotic deposition. We observed no significant age-related remodeling of CX43. Only spatial heterogeneity increased slightly with age, and this increase was better explained by biological age than by chronological age. Importantly, we found that CX43 features varied considerably among individuals in our population with no relevant relationship to age or fibrosis content, in contrast to animal species. We used our experimental results to feed computational models of human ventricular electrophysiology and to assess the effects of interindividual differences in specific features of CX43 and fibrosis on conduction velocity, action potential duration, and arrhythmogenicity. We found that larger amounts of fibrosis were associated with the highest arrhythmic risk, with this risk being increased when fibrosis deposition was combined with a reduction in CX43 amount and/or with an increase in CX43 spatial heterogeneity. These mechanisms underlying high arrhythmic risk in some individuals were not associated with age in our study population. In conclusion, our data rule out CX43 remodeling as an age-related arrhythmic substrate in the population beyond midlife, but highlight its potential as a proarrhythmic factor at the individual level, especially when combined with increased fibrosis.

Leuprolide acetate induces structural and functional recovery of injured spinal cord in rats

Gonadotropin-releasing hormone（Gn RH） and its synthetic analog leuprolide acetate, a Gn RH agonist, have neurotrophic properties. This study was designed to determine whether administration of leuprolide acetate can improve locomotor behavior, gait, micturition reflex, spinal cord morphology and the amount of microglia in the lesion epicenter after spinal cord injury in rats. Rats with spinal cord compression injury were administered leuprolide acetate or saline solution for 5 weeks. At the 5th week, leuprolide acetate-treated rats showed locomotor activity recovery by 38%, had improvement in kinematic gait and exhibited voiding reflex recovery by 60%, as compared with the 1st week. By contrast, saline solution-treated rats showed locomotor activity recovery only by 7%, but voiding reflex did not recover. More importantly, leuprolide acetate treatment reduced microglial immunological reaction and induced a trend towards greater area of white and gray matter in the spinal cord. Therefore, leuprolide acetate has great potential to repair spinal cord injury.

Main factors affecting lacertid lizard thermal ecology

The thermal ecology of ectotherms has been studied for almost 2 centuries,but additional attention is currently being paid to it,to understand how organisms deal with the environment in a climate change context.A consensus is still far away due to the large number of factors involved and their complex *** this context,3 analyses in lacertid lizards were carried out:(i)a meta-analysis,to test for differences between body and air temperatures from 71 populations;(ii)a meta-analysis concerning correlations between body and air temperatures from 60 populations;and(iii)a multimodel inference of thermoregulation effectiveness indices from 45 *** importance of different factors,including body size,habitat,insularity,altitude,climate and season,was evaluated in all analyses to model the response variables.A strong seasonality effect was observed,with a consistent pattern of less effective thermoregulation in summer compared to other *** was the second most important factor,with a consistent higher thermoregulation effort in populations occurring at high elevations(>1000 m above sea level).Other factors,such as insularity or body size,can also be important,but did not exhibit a clear ***,thermoregulation was less affected by climate and habitat type.

The ability to operate despite failure will become increasingly important as the use of optical sensor networks grows, and the amount of sensing information to be handled by a sensor network is increasing, especially for safety and security applications. In this review, the four categories of protection to allow service to be reestablished after a failure （dedicated/shared and line/path） are thoroughly discussed. This paper also presents an overview of the most representative robust fiber-optic sensor systems, discussing their schemes, pros and cons.

Background:Small populations that have been isolated by conflict make vaccination and surveillance difficult,threatening polio *** circulation is caused by asymptomatic *** is currently not clear whether the dynamics of waning immunity also influence the risk of silent circulation in the absence of *** circulation can,nevertheless,be present following a declaration of elimination as a result of inadequate acute flaccid paralysis surveillance(AFPS)or environmental surveillance(ES).Methods:We have constructed a stochastic model to understand how stochastic effects alter the ability of small populations to sustain virus circulation in the absence of *** analyzed how the stochastic process determinants of the duration of silent circulation that could have been detected by ES were affected by R0,waning dynamics,population size,and AFPS sensitivity in a discrete individual stochastic model with homogeneous contagiousness and random *** measured the duration of silent circulation both by the interval between detected acute flaccid paralysis(AFP)cases and the duration of circulation until ***:As R0 increased and population size increased,the interval between detected AFP cases and the duration of circulation until elimination *** AFPS detection rates decreased,the interval between detected AFP cases *** was up to a 22%chance of silent circulation lasting for more than 3 years with 100%AFP *** duration of silent circulation was not affected by the waning immunity ***:We demonstrated that small populations have the potential to sustain prolonged silent *** in these areas should be intensified before declaring *** further validate these conclusions,it is necessary to realistically relax the simplifying assumptions about mixing and waning.

Optical properties of phosphate glasses co-doped with Yb^(3+) and silver nanoparticles

We report the fabrication and spectroscopic characterization of Yb^3＋-doped phosphate glass, also containing silver nitrate. Scanning electron microscopy（SEM） provides the evidence of the formation of silver nano-particles（SNPs）, which are formed as a consequence of melting and thermal decomposition of Ag NO3. Absorption spectra of the samples in the visible-to-near-infrared spectral range reveal the presence of bands centered at 410 nm associated with the SNP-plasmon resonance, and at 976 nm due to the Yb^3＋. Under 916-nm laser-diode pumping, the effect of the SNP reflects that： i） the fluorescence in the 950-nm–1150-nm spectral range is strongly enhanced（～ 30 times）, while the fluorescence decay time associated with the ^2F5/2→^2F7/2transition of Yb^3＋ increases 25%, and ii） the basic lasing properties（saturation pumping intensity, the emission and absorption cross sections） are substantially improved.

Langerin-mediated internalization of a modified peptide routes antigens to early endosomes and enhances cross-presentation by human Langerhans cells

The potential of the skin immune system to generate immune responses is well established, and the skin is actively exploited as a vaccination site. Human skin contains several antigen-presenting cell subsets with specialized functions. In particular, the capacity to cross-present exogenous antigens to Cd8^＋ T cells is of interest for the design of effective immunotherapies against viruses or cancer. Here, we show that primary human Langerhans cells （LCs） were able to cross-present a synthetic long peptide （SLP） to Cd8^＋ T cells. In addition, modification of this SLP using antibodies against the receptor langerin, but not dectin-1, further enhanced the cross-presenting capacity of LCs through routing of internalized antigens to less proteolytic early endosome antigen 1 ^＋ early endosomes. The potency of LCs to enhance Cd8^＋ T-cell responses could be further increased through activation of LCs with the toll-like receptor 3 ligand polyinosinic：polycytidylic acid （phC）. Altogether, the data provide evidence that human LCs are able to cross-present antigens after langerin-mediated internalization. Furthermore, the potential for antigen modification to target LCs specifically provides a rationale for generating effective anti-tumor or anti-viral cytotoxic T lymphocyte responses.
