Steatosis and insulin resistance in hepatitis C: A way out for the virus?

The hepatitis C virus (HCV) induces lipid accumulation in vitro and in vivo. The pathogenesis of steatosis is due to both viral and host factors. Viral steatosis is mostly reported in patients with genotype 3a, whereas meta-bolic steatosis is often associated with genotype 1 and metabolic syndrome. Several molecular mechanisms responsible for steatosis have been associated with the HCV core protein, which is able to induce gene expres-sion and activity of sterol regulatory element binding protein 1 (SrEBP1) and peroxisome proliferator-activat-ed.receptor γ (PPArγ), increasing the transcription of genes involved in hepatic fatty acid synthesis. Steatosis has been also implicated in viral replication. In infected cells, HCV core protein is targeted to lipid droplets which serve as intracellular storage organelles. These studies have shown that lipid droplets are essential for virus assembly. Thus, HCV promotes steatosis as an eff icient mechanism for stable viral replication. Chronic HCV in-fection can also induce insulin resistance. In patients with HCV, insulin resistance is more strongly associated with viral load than visceral obesity. HCV seems to lead to insulin resistance through interference of intracellular insulin signalling by HCV proteins, mainly, the serine phosphorylation of insulin receptor-1 (IrS-1) and im-pairment of the downstream Akt signalling pathway. The HCV core protein interferes with in vitro insulin signal-ling by genotype-specif ic mechanisms, where the role of suppressor of cytokine signal 7 (SOCS-7) in genotype 3aand mammalian target of rapamycin (mTOr) in geno-type 1 in IrS-1 downregulation play key roles. Steatosis and insulin resistance have been associated with f ibrosis progression and a reduced.rate of sustained.response to peginterferon plus ribavirin.

Soil under dead or live organic matter systems:Effect of European shag(Phalacrocorax aristotelis L.)nesting on soil nematodes and nutrient mineralization

Here we studied whether soil systems differ if they are under the influence of live(plants)or dead organic matter systems(nest)in terms of C and N mineralization,microbiological characteristics and nematode trophic group *** analyzed physicochemical and microbiological properties of soils inside and outside nests of the European shag(Phalacrocorax aristotelis L.)on the Cies Islands(NW Spain).We sampled fresh soil below dead(nests)and live organic matter(plants)(paired samples,n=7).Soil of nests had lower organic matter and higher electric conductivity and dissolved organic C and extractable N contents than the soil of *** biomass and activity were greater in soil of nests than in soil of *** abundance of nematode trophic groups(bacterivores,fungivores,omnivores and herbivores)differred between soils of nests and plants,and the soil of plants supported a more abundant and diverse nematode *** present results points to that source of organic matter promote differences in the decomposer community,being more efficient in soil of nests because C mineralization is ***,this occurred independently of the complexity of the systems,higher in the soil of plants with more groups of nematodes.

PSTCNN: Explainable COVID-19 diagnosis using PSO-guided self-tuning CNN

Since 2019,the coronavirus disease-19(COVID-19)has been spreading rapidly worldwide,posing an unignorable threat to the global economy and human *** is a disease caused by severe acute respiratory syndrome coronavirus 2,a single-stranded.rNA virus of the genus *** virus is highly infectious and relies on its angiotensin-converting enzyme 2-receptor to enter *** the increase in the number of confirmed COVID-19 diagnoses,the difficulty of diagnosis due to the lack of global healthcare resources becomes increasingly *** learning-based computer-aided diagnosis models with high generalisability can effectively alleviate this *** tuning is essential in training such models and significantly impacts their final performance and training ***,traditional hyperparameter tuning methods are usually time-consuming and *** solve this issue,we introduce Particle Swarm Optimisation to build a PSO-guided Self-Tuning Convolution Neural Network(PSTCNN),allowing the model to tune hyperparameters ***,the proposed approach can reduce human ***,the optimisation algorithm can select the combination of hyperparameters in a targeted manner,thus stably achieving a solution closer to the global ***,the PSTCNN can obtain quite excellent results,with a sensitivity of 93.65%±1.86%,a specificity of 94.32%±2.07%,a precision of 94.30%±2.04%,an accuracy of 93.99%±1.78%,an F1-score of 93.97%±1.78%,Matthews Correlation Coefficient of 87.99%±3.56%,and Fowlkes-Mallows Index of 93.97%±1.78%.Our experiments demonstrate that compared to traditional methods,hyperparameter tuning of the model using an optimisation algorithm is faster and more effective.

Property and Activity of Molybdates Dispersed on Silica Obtained from Various Synthetic Proced.res

The synthesis and characterization of several dispersed molybdena catalysts on silica support (MoO3-SiO2) prepared from a variety of precursors (Mo(VI)-acetylacetonate, oxo-peroxo Mo-species, hydrated ammonium heptamolybdate) and preparation methods (deposition of the Mo-phase on finite SiO2 support by aqueous and methanol impregnations, by adsorption, by oxo-peroxo route-like, and by one-step synthesis of MoO3-SiO2 system with molecular precursors) are presented. The molybdena concentration on silica was comprised in a large interval (1.5 - 14 wt%) depending on the preparation method which governed the Mo-loading on silica. Convenient comparisons among samples at similar Mo-concentration have been made discussing the morphologic-structural (XrD, XPS, UV-vis-DrS, and N2-adsorption) and physicochemical (TG-DTG, TPr, and n-butylamine-TPD) sample properties. Polymeric octahedral polymolybdate aggregates predominated in the samples prepared by aqueous and methanol impregnations, which were at high Mo-concentration. On the contrary, isolated Mo(VI) species in distorted Td symmetry predominated in the sample prepared by adsorption which was at very low Mo-concentration. The sample acidity was composed of a weak acidy site population, associated with the silica support, and a strong acid site population associated with the Mo-dispersed phase. Oxidation tests of formaldehyde, an oxygen-containing VOC (Volatile Organic Compound), were performed to determine the prevalent redox or acidic function of the Mo-species at the surface of the catalysts.

Background:Lung hypoplasia,pulmonary persistent hypertension of the newborn and its morphological changes are the main features in congenital diaphragmatic hernia(CDH).This study was undertaken to investigate if antenatal use of sildenafil and/or bosentan attenuates vascular remodeling,promotes branching,and improves alveolarization in experimental nitrofeninduced ***:Nitrofen(100 mg)was gavage-fed to pregnant rats at post conception day(PCD)9 to induce *** rats were randomized to 5 groups:1)control;2)nitrofen;3)nitrofen+sildenafil 100 mg/kg per day at PCD 16-20;4)nitrofen+bosentan 30 mg/kg per day,at PCD 16-20,and 5)nitrofen+bosentan+sildenafil,same doses and administration *** cesarean delivery,the offsprings were sacrifi *** diaphragmatic defect and pulmonary hypoplasia were identified,and the lungs were *** wall thickness,bronchiolar density and alveolarization were ***:The offsprings with CDH were characterized by severe pulmonary hypoplasia(lung weight-to-body weight ratio:0.0263[95%confidence interval(CI)0.0242-0.0278)]in the nitrofen group versus 0.0385(95%CI 0.0355-0.0424)in the control group(P=0.0001).Pulmonary arterial wall thickness was decreased to 3.0(95%CI 2.8-3.7)μm in the nitrofen+sildenafil group versus 5.0(95%CI 4.1-4.9)μm in the nitrofen group(P=0.02).Terminal bronchioles increased to 13.7(95%CI 10.7-15.2)μm in the nitrofen+bosentan group in contrast to 8.7(95%CI 7.2-9.4)μm in the nitrofen group(P=0.002).More significant differences(P=0.0001)were seen in terminal bronchioles in the nitrofen+sildenafil+bosentan group than in the nitrofen group[14.0(95%CI 12.5-15.4)μm versus 8.5(95%CI 7.1-9.3)μm].Pulmonary arterial wall thickness was also decreased in the former ***:In this rat model,antenatal treatment with sildenafil attenuates vascular *** promotes the development of terminal bronchioles in nitrofen-induced CDH.

Carbon stocks in a highly fragmented landscape with seasonally dry tropical forest in the Neotropics

Background:Global modeling of carbon storage and sequestration often mischaracterizes unique ecosystems such as the seasonally dry tropical forest of the central region of the Gulf of Mexico,because species diversity is usually underestimated,as is their carbon *** this study,aboveground and soil carbon stocks were estimated to determine the climate mitigation potential of this highly degraded landscape(rest cover).results:Tree species in the study area had carbon content values that were 30%–40%higher than the standard value proposed by the IPCC(i.e.,50%).Tropical oak forest in the region,despite its restricted distribution and low species richness,accounted for the highest mean carbon stocks per unit *** main factors driving spatial variability in carbon stocks were:maximum precipitation,soil organic matter,clay and silt *** strong relationship was found between aboveground carbon stocks and soil organic carbon in the study ***-fication of carbon stocks is an important consideration in the assessment of the conservation value of remnants of native vegetation in human-modified ***:This study demonstrates the importance of the highly fragmented tropical dry regions of the Neo-tropics in maintaining landscape functionality and providing key ecosystem services such as carbon *** results also highlight how crucial field-based studies are for strengthening the accuracy of global ***,this approach reveals the real contribution of ecosystems that are not commonly taken into account in the mitigation of climate change effects.

背景社区获得性肺炎(CAP)风险在初级保健中的信息是有限的。我们评估了不同生活方式和共病条件作为初级保健成人CAP的危险因素。方法一项回顾性观察对照研究。使用国家初级保健数据监测系统检索了2009至2013年在西班牙初级保健部门诊断的成人CAP病例。匹配年龄和匹配性别的对照组采用发病率密度抽样(比例2∶1)。以百分比和比值比来表示关联分析。为了避免偏倚效应,建立二项回归模型。结果51 139例患者和102 372例对照组做比较。平均年龄为61.4(19.9)岁。危险因素与CAP的显著相关性如下:艾滋病[比值比(95%可信区间)5.21(4.35~6.27)]、慢性阻塞性肺病(COPD)[2.97(2.84~3.12)]、哮喘[2.16(2.07~2.26)]、吸烟[1.96(1.91~2.02)]和口腔卫生问题[1.45(1.41~1.49)]。患者组中有危险因素存在的平均患病率为82.2%,对照组为69.2%。CAP患病率的增加与危险因素和年龄的累积有关:1个危险因素时,在18~60岁老人中的风险相关性是1.42,在>60岁人中是1.57;2个危险因素时,是1.88和2.35;≥3个危险因素时,是3.11和4.34。结论成人CAP初级保健中的危险因素普遍较高。主要与危险因素相关的是艾滋病、COPD、哮喘、吸烟、口腔卫生问题。我们的风险叠加结果可以帮助临床医生确定肺炎风险较高的患者。
