Photosynthetic and Molecular Markers of CO_2-mediated Photosynthetic Downregulation in Nodulated Alfalfa

elevated CO2 leads to a decrease in potential net photosynthesis in long-term experiments and thus to a reduction in potential growth. This process is known as photosynthetic downregulation. There is no agreement on the definition of which parameters are the most sensitive for detecting CO2 acclimation. In order to investigate the most sensitive photosynthetic and molecular markers of CO2 acclimation, the effects of elevated CO2, and associated elevated temperature were analyzed in alfalfa plants inoculated with different Sinorhizobium meliloti strains. Plants （Medicago sativa L. cv. Aragon） were grown in summer or autumn in temperature gradient greenhouses （TGG）. At the end of the experiment, all plants showed acclimation in both seasons, especially under elevated summer temperatures. This was probably due to the lower nitrogen （N） availability caused by decreased N2-fixation under higher temperatures. Photosynthesis measured at growth CO2 concentration, rubisco in vitro activity and maximum rate of carboxylation were the most sensitive parameters for detecting downregulation. Severe acclimation was also related with decreases in leaf nitrogen content associated with declines in rubisco content （large and small subunits） and activity that resulted in a drop in photosynthesis. Despite the sensitivity of rubisco content as a marker of acclimation, it was not coordinated with gene expression, possibly due to a lag between gene transcription and protein translation.

Background:The island of Hispaniola,shared by Haiti and the Dominican Republic(DR),is the only remaining malaria-endemic island in the Caribbean and accounts for 95%of the lymphatic filariasis(LF)burden in the Americas.Both countries aim to eliminate the diseases by 2020.Migration from Haiti,where both diseases are more prevalent,may promote transmission in the DR.Historically,Haitian migrant labourers live in rural Dominican agricultural'company towns'called boteyes,many of which received mass drug administration(MDA)for LF elimination.This study sought to determine the prevalence of malaria and LF in bateyes of the DR and to describe related risk faaors for disease.Methods:From March to April 2016,a cross-seaional,cluster survey was conduaed across Dominican boteyes stratified into three regions:southwest,north and east.A household questionnaire(n=776),captured demographics,ethnic origin,mobility patterns,malaria intervention coverage,and knowledge,and recent fever and treatment-seeking.Two individuals per household(n=1418)were tested for malaria parasites by microscopy and rapid diagnostic test(RDT)and LF antigen by filariasis test strip(FTS).Population-level estimates and confidence intervals(Cl)were computed adjusting for the survey design.Two-sided Hests compared differences in knowledge scores.Results:No(0%)blood sample was Plosm odium-positw e by microscopy or RDT.Six individuals were FTS-positive(0.5%;95%CI:0.2-1.5),but none(0%)of these were microfllariae-positive.Most botey residents were born in the DR(57.8%),documented(85.0%),and permanent residents(85.1%).Very few respondents(9.4%)reported travel to Haiti in the past year.Overall,half(53.8%)of respondents owned a bed net,and 82.3%of net owners reported using it the previous night.Indoor residual spraying(IRS)differed by region(range:4.7%-61.2%).Most of those with recent fever sought care(56.0%),yet only 30.5%of those seeking care were tested for malaria.Compared to Dominican-born populations,Haitian-born respondents more frequently reported recent fe

Pseudostress-BasedMixed Finite element Methods for the Stokes Problem in Rn with Dirichlet Boundary Conditions.I:A Priori error Analysis

We consider a non-standard mixed method for the Stokes problem in Rn,n∈{2,3},with Dirichlet boundary conditions,in which,after using the incompressibility condition to eliminate the pressure,the pseudostress tensor s and the velocity vector u become the only unknowns.Then,we apply the Babuˇska-Brezzi theory to prove the well-posedness of the corresponding continuous and discrete formulations.In particular,we show that Raviart-Thomas elements of order k≥0 for s and piecewise polynomials of degree k for u ensure unique solvability and stability of the associated Galerkin scheme.In addition,we introduce and analyze an augmented approach for our pseudostress-velocity formulation.The methodology employed is based on the introduction of the Galerkin least-squares type terms arising from the constitutive and equilibrium equations,and the Dirichlet boundary condition for the velocity,all of them multiplied by suitable stabilization parameters.We show that these parameters can be chosen so that the resulting augmented variational formulation is defined by a strongly coercive bilinear form,whence the associated Galerkin scheme becomes well posed for any choice of finite element subspaces.For instance,Raviart-Thomas elements of order k≥0 for s and continuous piecewise polynomials of degree k+1 for u become a feasible choice in this case.Finally,extensive numerical experiments illustrating the good performance of the methods and comparing them with other procedures available in the literature,are provided.

Blood transfusion for the treatment of acute anaemia in inflammatory bowel disease and other digestive diseases

Allogeneic blood transfusion (ABT) is frequently used as the first therapeutic option for the treatment of acute anaemia in patients with inflammatory bowel disease (IBD), especially when it developed due to gastrointestinal or perioperative blood loss, but is not risk-free. Adverse effects of ABT include, but are not limited to, acute hemolytic reaction (wrong blood or wrong patient), febrile non-hemolytic transfusional reaction, bacterial contamination, transfusion-related acute lung injury, transfusion associated circulatory overload, transfusion-related immuno-modulation, and transmission of almost all infectious diseases (bacteria, virus, protozoa and prion), which might result in increased risk of morbidity and mortality. Unfortunately, the main physiological goal of ABT, i.e. to increase oxygen consumption by the hypoxic tissues, has not been well documented. In contrast, the ABT is usually misused only to increase the haemoglobin level within a f ixed protocol [mostly two by two packed red blood cell (PRC) units] independently of the patient' s tolerance to normovolemic anaemia or his clinical response to the transfusion of PRC units according to a "one-by-one" administration schedule. evidencebased clinical guidelines may promote best transfusion practices by implementing restrictive transfusion protocols, thus reducing variability and minimizing the avoidable risks of transfusion, and the use of autologous blood and pharmacologic alternatives. In this regard, preoperative autologous blood donation (PABD) consistently diminished the frequency of ABT, although its contribution to ABT avoidance is reduced when performed under a transfusion protocol. In addition, interpretation of utility of PABD in surgical IBD patients is hampered by scarcity of published data. However, the role of autologous red blood cells as drug carriers is promising. Biometric and ICL-related risk factors associated to sub-optimal vaults in eyes implanted with implantable collamer lenses

Background:To identify biometric and implantable collamer lens(ICL)-related risk factors associated with suboptimal postoperative vault in eyes implanted with phakic ICL.Methods:This study reports a retrospective case series of the first operated eye in 360 patients implanted with myopic spherical or toric ICL.Preoperatively,white-to-white(WTW),central keratometry(Kc)and central corneal thickness(CCT)were measured using the Pentacam.Anterior-segment optical coherence tomography(AS-OCT,Visante)was applied preoperatively for measuring the horizontal anterior-chamber angle-to-angle distance(ATA),internal anterior chamber depth(ACD),crystalline lens rise(CLR),anterior-chamber angle(ACA)and postoperatively the vault.eyes were divided into three vault groups:low(LVG:≤250μm),optimal(OVG:>250 andegression(MLR)was used to find the sub-optimal vault predictors.Results:MLR showed that CLR,ICL size minus the ATA(ICL size-ATA),age,ICL spherical equivalent(ICLSe)and ICL size as contributing factors for sub-optimal vaults(pseudo-R2=0.40).Increased CLR(OR:1.01,CI:1.00-1.01)and less myopic ICLSe(OR:1.22,CI:1.07-1.40)were risk factors for low vaults.Larger ICL size-ATA(OR:41.29,CI:10.57-161.22)and the 13.7mm ICL(OR:7.08,CI:3.16-15.89)were risk factors for high vaults,whereas less myopic ICLSe(OR:0.85,CI:0.76-0.95)and older age(OR:0.92,CI:0.88-0.98)were protective factors.Conclusion:High CLR and low ICLSe were the major risk factors in eyes presenting low vaults.In the opposite direction,ICL size-ATA was the major contributor for high vaults.This relationship was more critical in higher myopic ICLSe,younger eyes and when 13.7mm ICL were used.The findings show that factors influencing the vault have differentiated weight of influence depending on the type of vault(low,optimal or high).

Direct soil temperature(ST)measurement is time-consuming and costly;thus,the use of simple and cost-effective machine learning(ML)tools is helpful.In this study,ML approaches,including KStar,instance-based K-nearest learning(IBK),and locally weighted learning(LWL),coupled with resampling algorithms of bagging(BA)and dagging(DA)(BA-IBK,BA-KStar,BA-LWL,DA-IBK,DA-KStar,and DA-LWL)were developed and tested for multi-step ahead(3,6,and 9 d ahead)ST forecasting.In addition,a linear regression(LR)model was used as a benchmark to evaluate the results.A dataset was established,with daily ST time-series at 5 and 50 cm soil depths in a farmland as models’output and meteorological data as models’input,including mean(T_(mean)),minimum(Tmin),and maximum(T_(max))air temperatures,evaporation(eva),sunshine hours(SSH),and solar radiation(SR),which were collected at Isfahan Synoptic Station(Iran)for 13 years(1992–2005).Six different input combination scenarios were selected based on Pearson’s correlation coefficients between inputs and outputs and fed into the models.We used 70%of the data to train the models,with the remaining 30%used for model evaluation via multiple visual and quantitative metrics.Our?ndings showed that T_(mean)was the most effective input variable for ST forecasting in most of the developed models,while in some cases the combinations of variables,including T_(mean)and T_(max)and T_(mean),T_(max),Tmin,eva,and SSH proved to be the best input combinations.Among the evaluated models,BA-KStar showed greater compatibility,while in most cases,BA-IBK and-LWL provided more accurate results,depending on soil depth.For the 5 cm soil depth,BA-KStar had superior performance(i.e.,Nash-Sutcliffe efficiency(NSe)=0.90,0.87,and 0.85 for 3,6,and 9 d ahead forecasting,respectively);for the 50 cm soil depth,DA-KStar outperformed the other models(i.e.,NSe=0.88,0.89,and 0.89 for 3,6,and 9 d ahead forecasting,respectively).The results con?rmed that all hybrid models had higher prediction capabilities than the LR model.

AIM To demonstrate the non-inferiority(15% non-inferiority limit) of monotherapy with tenofovir disoproxil fumarate(TDF) vs the combination of lamivudine(LAM) plus adefovir dipivoxil(ADV) in the maintenance of virologic response in patients with chronic hepatitis B(CHB) and prior failure with LAM.MeTHODS This study was a Phase IV prospective, randomized, open, controlled study with 2 parallel groups(TDF and LAM+ADV) of adult patients with hepatitis B e antigen(HBe Ag)-negative CHB, prior failure with LAM, on treatment with LAM+ADV for at least 6 mo, without prior resistance to ADV and with an undetectable viral load at the start of the study, in 14 Spanish hospitals. The follow-up time for each patient was 48 wk after randomization, with quarterly visits in which the viral load, biochemical and serological parameters, adverse effects, adherence to treatment and consumption of hospital resources were analysed.ReSULTS Forty-six patients were evaluated [median age: 55.4 years(30.2-75.2); 84.8% male], including 22 patients with TDF and 24 with LAM+ADV. During study development, hepatitis B virus DNA(HBV-DNA) remained undetectable, all patients remained HBe Ag negative, and hepatitis B surface antigen(HBs Ag) positive. Alanine aminotransferase(ALT) values at the end of the study were similar in the 2 groups(25.1± 7.65, TDF vs 24.22 ± 8.38, LAM+ADV, P = 0.646). No significant changes were observed in creatinine or serum phosphorus values in either group. No significant differences between the 2 groups were noted in the identification of adverse effects(Aes)(53.8%, TDF vs 37.5%, LAM+ADV, P = 0.170), and none of the Aes which occurred were serious. Treatment adherence was 95.5% and 83.3% in the TDF and the LAM+ADV groups, respectively(P = 0.488). Objective: To identify and synthesize the most recent available evidence of effectiveness of acupuncture on pain, physical function and health-related quality of life(HRQoL) in patients with rheumatoid arthritis(RA). Methods: A comprehensive search of 12 Western and Chinese databases was undertaken from their inception up to end of 2016. Randomized controlled trials(RCTs), concerning patients with RA treated with needle acupuncture, written in english, Portuguese, German or Chinese were included. Primary outcomes included pain, physical function and HRQoL. Secondary outcomes included morning stiffness, functional impairment, number of tender and swollen joints and serum concentrations of inflamatory markers. Methodological quality was assessed by three independent reviewers using the standardized critical appraisal instrument from the Joanna Briggs Institute Meta-Analysis of Statistics Assessment and Review Instrument. Results: Twenty-two studies met the inclusion criteria. Of those, 9 studies were excluded after assessment of their methodological quality. The remaining 13 original RCTs included 974 patients. Ten of these studies published in China, showed favorable statistical significant effects of acupuncture in relieving symptoms of RA compared with controls. Conclusions: evidence suggests that acupuncture interventions may have a positive effect in pain relief, physical function and HRQo L in RA patients. However, due to the heterogeneity and methodologic limitations of the studies included in this systematic review, evidence is not strong enough to produce a best practice guideline.

The aim of this study was to compare mandibular form （i.e., size and shape） between patients with agenesis of the lower second premolar （P2） and a control group with no agenesis. Three hypotheses were tested：（H1） agenesis causes a change in mandibular morphology because of inadequate alveolar ridge development in the area of the missing tooth （mandibular plasticity）; （H2） agenesis is caused by spatial limitations within the mandible （dental plasticity）; and （H3） common genetic/ epigenetic factors cause agenesis and affect mandibular form （pleiotropy）. A geometric morphometric analysis was applied to cross-sectional images of computed tomography （CT） scans of three matched groups （n= 50 each）：（1） regularly erupted P2; （2） agenesis of P2 and the primary second molar in situ; and （3） agenesis of P2 and the primary second molar missing for 〉3 months. Cross-sections of the three areas of interest （first premolar, P2, first molar） were digitized with 23 landmarks and superimposed by a generalized Procrustes analysis. On average, the mandibular cross-sections were narrower and shorter in patients with P2 agenesis compared with that in the control group. Both agenesis groups featured a pronounced submandibular fossa. These differences extended at least one tooth beyond the agenesis-affected region. Taken together with the large interindividual variation that resulted in massively overlapping group distributions, these findings support genetic and/or epigenetic pleiotropy （H3） as the most likely origin of the observed covariation between mandibular form and odontogenesis. Clinically, reduced dimensions and greater variability of mandibular form, as well as a pronounced submandibular fossa, should be expected during the treatment planning of patients with P2 agenesis.
