Disruption of Arabidopsis CHY1 Reveals an Important Role of Metabolic Status in Plant Cold Stress Signaling

To study cold signaling, we screened for Arabidopsis mutants with altered cold-induced transcription of a firefly luciferase reporter gene driven by the CBF3 promoter （CBF3-LUC）. One mutant, chy1-10, displayed reduced cold-induction of CBF3-LUC luminescence. RNA gel blot analysis revealed that expression of endogenous CBFs also was reduced in the chyl mutant, chy1-10 mutant plants are more sensitive to freezing treatment than wild-type after cold acclimation. Both the wild-type and chyl mutant plants are sensitive to darkness-induced starvation at warm temperatures, although chyl plants are slightly more sensitive. This dark-sensitivity is suppressed by cold temperature in the wildtype but not in chyl. Constitutive CBF3 expression partially rescues the sensitivity of chyl-10 plants to dark treatment in the cold. The chyl mutant accumulates higher levels of reactive oxygen species, and application of hydrogen peroxide can reduce cold-induction of CBF3-LUC in wild-type. Map-based cloning of the gene defective in the mutant revealed a nonsense mutation in CHY1, which encodes a peroxisomal β-hydroxyisobutyryl （HIBYL）-CoA hydrolase needed for valine catabolism and fatty acid β-oxidation. Our results suggest a role for peroxisomal metabolism in cold stress signaling, and plant tolerance to cold stress and darkness-induced starvation.

The phytohormone abscisic acid (ABA) modulates the expression of many genes important to plant growth and development and to stress adaptation. In this study, we found that an APETELA2/Ethylene Responsive Binding Protein (AP2/EREBP) type transcription factor, AtERF7, plays an important role in ABA responses. AtERF7 interacts with the protein kinase PKS3, which has been shown to be a global regulator of ABA responses. AtERF7 binds to the GCC box and acts as a repressor of gene transcription. AtERF7 interacts with the Arabidopsis homolog of human global co-repressor of transcription, AtSin3, which in turn may interact with HDA19, a histone deacetylase. The transcriptional repression activity of AtERF7 is enhanced by HDA19 and AtSin3. Arabidopsis plants overexpressing AtERF7 show reduced sensitivity of guard cells to ABA and increased transpirational water loss. In contrast, AtERF7 and AtSin3 RNA interference lines show increased sensitivity to ABA during germination. Together, our results suggest that AtERF7 plays an important role in ABA responses and it may be part of a transcriptional repressor complex and be regulated by PKS3.

Municipal solid waste(MSW) can have a negative impact on the environment and/or human health, if not properly managed. Though landfill is the most common method used to manage MSW, treating MSW using waste-toenergy(Wt E) technologies is an option that is receiving increasing interest. Pyrolysis is a Wt E technology that is capable of converting waste into products such as bio-gas, biooil and bio-char. Bio-char, which varies in composition based on the raw material pyrolysed, has the potential to be used as a fuel and as an applied material for catalysis or adsorption. In this work the removal of azo dye was investigated using biochar from pyrolysis of a cellulosic municipal solid waste. The bio-char was characterised using Scanning Electron Microscopy and Surface Area Analysis. The influence of p H on the dye removal was investigated over the range 4-7. The experimental data was analysed using four non-linear kinetic models(Pseudo first-order, pseudo second-order, Weber intraparticle diffusion and Elovich equation) and four non-linear isotherm models(Langmuir, Freundlich, Temkin and Sips), and ranked based on the statistical analysis tools in origin software. The dye removal kinetics most closely followed pseudo-first order kinetics whilst the adsorption isotherms were most closely fitted by the Temkin model over the p H range studied.

Ocular surface evaluation in eyes with chronic glaucoma on long term topical antiglaucoma therapy

AIM： To evaluate ocular surface changes and its correlation with the central corneal subbasal nerve fibre layer in chronic glaucoma patients. METHODS： A prospective comparative study of ocular surface evaluation was performed in 50 eyes of 25 patients using two or more antiglaucoma medications for at least 6too and 50 eyes of 25 normal subjects without any ocular problems as controls. The study parameters evaluated included visual acuity, intraocular pressure, ocular surface evaluation parameters [fluorescein break-up time （FTBUT）, Schirmer＇s I test, ocular surface staining scores and ocular surface disease index score （OSDI）], central corneal sensation （Cochet Bonnett aesthesiometer）, central subbasal nerve fiber layer density （SBNFLD） by confocal microscopy. RESULTS： The mean values in the glaucoma cases and control groups respectively were as follows： OSDI score （35.89±16.07/6.02±3.84; P=-0.001）, Schirmer＇s I test score （7.632.64 mm/12.86±1.93 mm; P=-0.001）, FTBUT （9.442.76s/11.8±1.88s; P=0.001）, corneal （5.7±2.33/ 1.1±0.58; P=0.001） and conjunctival staining score （5.061.94/0.84±0.46; P=0.001）, corneal sensitivity （4.68±0.44/5.07±0.37; P=0.076）, mean subbasal nerve fiber number （3.58±0.99/5.40±1.70; P=0.001）, SBNFL length （1101.44±287.56 pm/1963.70±562.56 pro; P=0.001） and density （6883.941798.03 pm/mm/12 273.153516.04 pm/mm2; P=O.O01）. Dry eye severity of level 2 and 3 was seen in 66% of glaucoma group. Corneal （/=0.86） and conjunctival staining （R2=0.71） and OSDI score （R=0.67） showed statistically significant negative correlation with central corneal SBNFLD while FTBUT （R==0.84）, corneal sensitivity （/=0.52） showed positive correlation to central corneal SBNFLD in the long term topical antiglaucoma medication group. CONCLUSION： Ocular surface changes and antiglaucoma therapy induced dry eye is found to be associated with decreased SBNFLD in eyes on long term topical antiglaucoma medications.
