Robust interface and excellent as-built mechanical properties of Ti–6Al–4V fabricated through laser-aided additive manufacturing with powder and wire

The feasibility of manufacturing Ti-6Al-4V samples through a combination of laser-aided additive manufacturing with powder(LAAM_(p))and wire(LAAM_(w))was explored.A process study was first conducted to successfully circumvent defects in Ti-6Al-4V deposits for LAAM_(p) and LAAM_(w),*** the optimized process parameters,robust interfaces were achieved between powder/wire deposits and the forged substrate,as well as between powder and wire *** characterization results revealed the epitaxial prior β grains in the deposited Ti-6Al-4V,wherein the powder deposit was dominated by a finerα′microstructure and the wire deposit was characterized by lamellar α *** mechanisms of microstructure formation and correlation with mechanical behavior were analyzed and *** mechanical properties of the interfacial samples can meet the requirements of the relevant Aerospace Material Specifications(AMS 6932)even without post heat *** fracture occurred within the interfacial area,further suggesting the robust *** findings of this study highlighted the feasibility of combining LAAM_(p) and LAAM_(w) in the direct manufacturing of Ti-6Al-4V parts in accordance with the required dimensional resolution and deposition rate,together with sound strength and ductility balance in the as-built condition.

Acoustic orbital angular momentum (OAM) provides an infinite set of orthogonal bases in Hilbert space and significantly boosts the link capacity and data transmission rate. High-capacity and low-crosstalk OAM detection within compact physical space is highly desired to effectively harness this spatial dimension of sound wave, but the existing mechanisms generally either operate only for finite OAMs with predetermined mode orders or rely on complicated active devices. Here, we propose a phase saddle point (PSP) transformation mechanism for distinguishing and demultiplexing acoustic OAM modes in a passive, mode-adaptive,high-efficiency, and high-accuracy paradigm. The phase mask characterized by a PSP in the specific area is created through the single-pass log-polar transformation without prior information of incident modes, whose interaction with incident vortex beams of different OAM modes will yield the translation of the stationary phase point and thereby generate the corresponding focused line spots at distinct lateral positions. We theoretically elucidate the mode-adaptive response of PSP transformation by deriving the quantitative relationship between the OAM order and line spot offset. This mode adaptability enables our mechanism to detect OAM of arbitrary mode orders and mode numbers with the inter-mode crosstalk being effectively suppressed, beyond attainable with the existing methods. The effectiveness of our proposed mechanism is validated by demonstrating several distinct examples of single-mode OAM sorting and multi-mode synthesized vortex beam demultiplexing. We anticipate this modeadaptive methodology, designed for compact dimensions and featuring with low crosstalk and uniform efficiency, will find important applications in OAM information processing.

Specialized pro-resolving lipid mediators including maresin 1 mediate resolution but the levels of these are reduced in Alzheimer's disease brain, suggesting that they constitute a novel target for the treatment of Alzheimer's disease to prevent/stop inflammation and combat disease pathology. Therefore, it is important to clarify whether they counteract the expression of genes and proteins induced by amyloid-β. With this objective, we analyzed the relevance of human monocyte–derived microglia for in vitro modeling of neuroinflammation and its resolution in the context of Alzheimer's disease and investigated the pro-resolving bioactivity of maresin 1 on amyloid-β42–induced Alzheimer's disease–like inflammation. Analysis of RNA-sequencing data and secreted proteins in supernatants from the monocyte-derived microglia showed that the monocyte-derived microglia resembled Alzheimer's disease–like neuroinflammation in human brain microglia after incubation with amyloid-β42. Maresin 1 restored homeostasis by down-regulating inflammatory pathway related gene expression induced by amyloid-β42 in monocyte-derived microglia, protection of maresin 1 against the effects of amyloid-β42 is mediated by a re-balancing of inflammatory transcriptional networks in which modulation of gene transcription in the nuclear factor-kappa B pathway plays a major part. We pinpointed molecular targets that are associated with both neuroinflammation in Alzheimer's disease and therapeutic targets by maresin 1. In conclusion, monocyte-derived microglia represent a relevant in vitro microglial model for studies on Alzheimer's disease-like inflammation and drug response for individual patients. Maresin 1 ameliorates amyloid-β42–induced changes in several genes of importance in Alzheimer's disease, highlighting its potential as a therapeutic target for Alzheimer's disease.

目的:生物信息学分析筛选出骨肉瘤关键基因,探讨骨肉瘤潜在分子标记物。方法:从GEO数据库中下载GSE14359、GSE16088、GSE160913个数据库集,利用R语言“SVA”软件包进行合并;差异分析利用R语言“limma”软件包进行分析,并对差异基因运用R语言“clusterProfiler”进行GO和KEGG富集分析。随后,对最显著富集的KEGG通路基因进行筛选出最关键的Top5基因并进行生存分析,筛选对预后具有影响意义的关键基因,然后运用实时荧光定量聚合酶链式反应(qRT-PCR)验证最关键基因的差异性表达。结果:通过数据合并共得到12547个基因的数据集,分析出723个表达基因有差异;差异基因的KEGG富集最显著通路是PI3K-Akt信号通路,并且该通路的排名前5的关键基因为ITGA3、ITGA4、ITGAV、COL1A1、COL1A2。排名前5的关键基因中,与ITGA3低表达组比较,ITGA3高表达组生存预后较好(P<0.05),在143B、MNNG、U2OS 3个骨肉瘤细胞系中ITGA3相对表达量低于正常成骨细胞系hFOB1.19(P<0.01)。结论:ITGA3在骨肉瘤中呈现出低表达,这可能成为预测骨肉瘤患者预后的潜在生物标记物。

Widely used spin-coated nickle oxide （NiOx） based perovskite solar cells often suffer from severe interfacial reactions between the NiOxand adjacent perovskite layers due to surface defect states,which inherently impair device performance in a long-term view,even with surface molecule *** this study,we developed high-quality magnetron-sputtered NiOxthin films through detailed process optimization,and compared systematically sputtered and spin-coated NiOxthin film surfaces from materials to *** sputtered NiOxfilms exhibit improved crystallinity,smoother surfaces,and significantly reduced Ni3+or Ni vacancies compared to their spin-coated ***,the interface between the perovskite and sputtered NiOxfilm shows a substantially reduced density of defect *** solar cells （PSCs） fabricated with our optimally sputtered NiOxfilms achieved a high power conversion efficiency （PCE） of up to 19.93%and demonstrated enhanced stability,maintaining 86.2% efficiency during 500 h of maximum power point tracking under one standard sun ***,with the surface modification using (4-(2,7-dibromo-9,9-dimethylacridin-10（9H）-yl)butyl)p hosphonic acid （DMAcPA）,the device PCE was further promoted to 23.07%,which is the highest value reported for sputtered NiOxbased PSCs so far.

目的探讨环状RNA hsa_circ_0003568在骨肉瘤(OS)中的表达并采用生物信息学方法进行circRNA-miRNA-mRNA的网络构建,分析其可能的机制及临床意义。方法采用实时荧光定量聚合酶链反应(RT-qPCR)检测hsa_circ_0003568在OS细胞系及正常成骨细胞中的表达。随后,基于circRNA-miRNA和miRNA-mRNA构建circRNA-miRNA-mRNA网络。通过多种circRNA软件数据库进行分析hsa_circ_0003568靶向的miRNA,并进一步通过miRNA数据软件预测其靶基因,选择至少4个以上软件支持的靶向miRNA及靶基因,并通过GEPIA数据库软件对靶基因进行生存分析筛选出显著预后差异的靶基因从而分析hsa_circ_0003568潜在的临床意义。结果RT-qPCR检测结果表明:与正常对照组相比,hsa_circ_0003568在OS细胞系中显著升高(P<0.05)。测序发现hsa_circ_0003568的成环连接点位于IST1前体RNA的第5和6外显子上。生物信息学分析结果显示:hsa_circ_0003568同时靶向两个miRNA(miR-140-3p及miR-502-5p),构建了hsa_circ_0003568、miR-140-3p/miR-502-5p及13个靶基因的circRNA-miRNA-mRNA网络,此外,GEPIA数据库分析中发现CDK6和NFYA两个靶基因,与OS患者的生存预后显著相关(P<0.05),并且NFYA同时是miR-140-3p及miR-502-5p的潜在靶基因。结论高表达的hsa_circ_0003568可能通过miR-140-3p及miR-502-5p调控基因CDK6及NFYA参与OS的发生发展过程。

In this work, we provide a comprehensive review on the formation, evolution, properties, and effects of supercritical geofluid. In Earth's interior, enhanced miscibility between H2O and silicate by the addition of special components or by the increase of pressure and temperature gives rise to supercritical geofluid with a significant amount of both H2O and silicate *** formation of supercritical geofluid in magmatic-hydrothermal systems, typified by pegmatite system, is governed by meltfluid critical curve. The formation of supercritical geofluid in metamorphic systems, typified by subducted slab, is governed by the second critical end point. Experimental results suggest that the presence of boron and fluorine in pegmatite system makes it possible to form supercritical geofluid at crustal depths, but the release of supercritical geofluid from subducted slab is withheld until almost 100 km depth. A major presence of both H2O and depolymerized structural units（monomers, dimers, etc.） endows supercritical geofluid with unique physical properties including low density, low elastic moduli, low viscosity, high diffusivity,and high electrical conductivity. Supercritical geofluid can effectively mobilize a variety of elements even including high field strength elements and heavy rare earth elements. The chemical signatures of supercritical geofluid can be inherited by metasomatized mantle and mantle-derived melts, and this could give an explanation of the oxidation of arc magmas. Phase separation of supercritical geofluid through the mechanism of spinodal decomposition leads to formation of a melt network. Multiphase fluid inclusions recovered from subduction zone rocks and pegmatites are possible relics of supercritical geofluid. Supercritical geofluid can cause electrical anomaly and low seismic velocity near the top of subducted slab, and can be linked with intermediate-focus earthquakes.
