Kinetics of Iododeoxyuridine release from sodium alginate hydrogel in vitro

Objective To investigate the kinetics of Iododeoxyuridine(IUdr)release from sodium alginate hydrogel cross-linked with varying amounts of calcium chloride, and to optimize sustained release for further periadventitial I125-labeled IUdr delivery to suppress intimal hyperplasia following angioplasty in vivo. Methods Four hydrogels,composed of 0.16 mEq sodium alginate and 200 g IUdr, were cross-linked with calcium chloride to yield ion equivalence (IE) ratios (Calcium: alginate) of 3:1, 4:1, 5:1, or 6:1. 2 ml of normal saline was placed on top of each hydrogel and allowed to remain in contact at 37℃ for up to 30 days. At set time intervals, the concentration and amount of IUdr in the eluate were assayed by high performance liquid chromatography using UV detection and Water symmetry C18 column. The data for accumulated release rate and concentration in the eluate were calculated based on the calibration curve of peak area versus IUdr concentration. The hydrogel morphologic degradations were also observed. results The hydrogels entrapped 92.9%, 98.6%, 98.4% and 98.6% of the IUdr with 3:1, 4:1, 5:1 and 6:1 IE ratios, respectively. IUdr concentration in eluates from 3:1 IE ratio hydrogel decreased faster than that from other hydrogels over time (P ratio hydrogels produced more than 10 μm IUdr concentrations in eluates for the first 8 days, while the 3:1 IE ratio hydrogel for 4 days. IUdr release rates of the 4:1, 5:1 and 6:1 IE ratio hydrogels were very close, however they were lower than that of the 3:1 IE hydrogel in the first 48 hours (P ratio hydrogels had 100% and 88% degradation, but no significant degradation was observed in the other hydrogels. Conclusion The sodium alginate hydrogel with 4:1 IE ratio exhibited an optimal IUdr sustained release and almost complete degradation in 30 days. (J Intervent radiol, 2006, 15: 293-298)

Assessment of oxidative stress indices in a marine macro brown alga Padina tetrastromatica(Hauck) from comparable polluted coastal regions of the Arabian Sea,west coast of India

Oxidative stress and antioxidant defence systems were assessed in a marine brown alga Padina tetrastromatica, commonly occurring from the tropics. Lipid peroxidation （LPX） and H2O2 were measured as oxidative stress markers, and antioxidant defences were measured as catalase （CAT）, glutathione S-transferase （GST） and ascorbic acid （ASA）, in order to understand their dissimilarity with respect to pollution levels from selective locations along the central west coast of India. A significant increased levels of LPX, H2O2, CAT and GST were observed in samples from relatively polluted localities （Colaba and Karwar） when compared to less polluted locality （Anjuna）, while AsA concentration was higher in algal samples from worst polluted region of Colaba. Heavy metals such as Cd and Pb were also higher in the vicinity of polluted areas compared to reference area. Variation of oxidative stress indices in response to accumulation of heavy metals within P. tetrastromatica could be used as molecular biomarkers in assessment and monitoring environmental quality of ecologically sensitive marine habitats.

Monozygotic twinning after donor egg intracytoplasmic sperm injection-A case report

We are reporting a case of monozygotic twinning after donor egg intracytoplasmic sperm injection (ICSI). A 34-year-old lady presented to our centre with primary infertility due to severe endometriosis and low egg reserve. ICSI with donor eggs was planned. A 24 years old voluntary oocyte donor was investigated and stimulated under antagonist protocol. Total 21 eggs and all in metaphaseⅡwere retrieved. These eggs were injected with the patient's husband sperms by ICSI. Patient's endometrial lining was prepared under hormonal replacement therapy protocol. Two expanded blastocysts were transferred on day 5 of progesterone. Beta human chorionic gonadotropin was positive after 10 days. The first antenatal scan at 6 weeks could pick up only two sacs indicating twin conception. repeat scan at 12 weeks revealed tri-amniotic triplet conception with two foetuses sharing the same placenta (triamniotic pregnancy with monochorionic twins). The patient was counselled about risks associated with triplet conception and was advised of embryo reduction. Two mono chorionic twins were reduced under ultrasonography guidance. Single pregnancy continued till 21 weeks after which the patient miscarried spontaneously. It is difficult to identify the subset of patients at risk for monozygotic twinning;hence, all patients should be counselled about possibility of monozygotic twinning while deciding the number of embryos to be transferred.

Efficient parallelization of SPH algorithm on modern multi-core CPUs and massively parallel GPUs

Smoothed Particle Hydrodynamics (SPH) is fast emerging as a practically usefulcomputational simulation tool for a wide variety of engineering problems. SPH isalso gaining popularity as the back bone for fast and realistic animations in graphicsand video games. The Lagrangian and mesh-free nature of the method facilitates fastand accurate simulation of material deformation, interface capture, etc. Typically,particle-based methods would necessitate particle search and locate algorithms tobe implemented efficiently, as continuous creation of neighbor particle lists is acomputationally expensive step. Hence, it is advantageous to implement SPH, on modernmulti-core platforms with the help of High-Performance Computing (HPC) tools. Inthis work, the computational performance of an SPH algorithm is assessed on multicore Central Processing Unit (CPU) as well as massively parallel General PurposeGraphical Processing Units (GP-GPU). Parallelizing SPH faces several challenges suchas, scalability of the neighbor search process, force calculations, minimizing threaddivergence, achieving coalesced memory access patterns, balancing workload, ensuringoptimum use of computational resources, etc. While addressing some of these challenges,detailed analysis of performance metrics such as speedup, global load efficiency, globalstore efficiency, warp execution efficiency, occupancy, etc. is evaluated. The OpenMP andCompute Unified Device Architecture (CUDA) parallel programming models have beenused for parallel computing on Intel Xeon(r) E5-2630 multi-core CPU and NVIDIAQuadro M4000 and NVIDIA Tesla p100 massively parallel GPU architectures. Standardbenchmark problems from the Computational Fluid Dynamics (CFD) literature are chosen for the validation. The key concern of how to identify a suitable architecturefor mesh-less methods which essentially require heavy workload of neighbor search andevaluation of local force fields from neighbor interactions is addressed.

Growth inhibition mediated by PSP94 or CrISP-3 is prostate cancer cell line specific

The prostate secretory protein of 94 amino acids （PSP94） has been shown to interact with cysteine-rich secretory protein 3 （CrISP-3） in human seminal plasma. Interestingly, PSP94 expression is redaced or lost in the majority of the prostate tumours, whereas CrISP-3 expression is upregulated in prostate cancer compared with normal prostate tissue. To obtain a better understanding of the individual roles these proteins have in prostate tumourigenesis and the functional relevance of their interaction, we ectopically expressed either PSP94 or CrISP-3 alone or PSP94 along with CrISP-3 in three prostate cell lines （PC3, WPE1-NB26 and LNCaP） and performed growth inhibition assays. reverse transcription-polymerase chain reaction and Western blot analysis were used to screen prostate cell lines for PSP94 and CrISP-3 expression. Mammalian expression constructs for human PSP94 and CrISP-3 were also generated and the expression, localization and secretion of recombinant protein were assayed by transfection followed by Western blot analysis and immunofluorescence assay. The effect that ectopic expression of PSP94 or CrISP-3 had on cell growth was studied by clonogenic survival assay follokving transfection. To evaluate the effects of coexpression of the two proteins, stable clones of PC3 that expressed PSP94 were generated. They were subsequently transfected with a CrISP-3 expression construct and subjected to clonogenic survival assay. Our results showed that PSP94 and CrISP-3 could each induce growth inhibition in a cell line specific manner. Although the growth of CrISP-3-positive cell lines was inhibited by PSP94, growth inhibition mediated by CrISP-3 was not affected by the presence or absence of PSP94. This suggests that CrISP-3 may participate in PSP94-independent activities during prostate tumourigenesis.

A Deep Learning Modeling Framework to Capture Mixing Patterns in reactive-Transport Systems

Prediction and control of chemical mixing are vital for many scientific areas such as subsurface reactive transport, climate modeling, combustion, epidemiology, and pharmacology. Due to the complex nature of mixing in heterogeneous andanisotropic media, the mathematical models related to this phenomenon are not analytically tractable. Numerical simulations often provide a viable route to predict chemical mixing accurately. However, contemporary modeling approaches for mixing cannot utilize available spatial-temporal data to improve the accuracy of the future prediction and can be compute-intensive, especially when the spatial domain is large andfor long-term temporal predictions. To address this knowledge gap, we will present inthis paper a deep learning (DL) modeling framework applied to predict the progress ofchemical mixing under fast bimolecular reactions. This framework uses convolutionalneural networks (CNN) for capturing spatial patterns and long short-term memory(LSTM) networks for forecasting temporal variations in mixing. By careful design ofthe framework—placement of non-negative constraint on the weights of the CNN andthe selection of activation function, the framework ensures non-negativity of the chemical species at all spatial points and for all times. Our DL-based framework is fast,accurate, and requires minimal data for training. The time needed to obtain a forecastusing the model is a fraction (≈ O(10−6)) of the time needed to obtain the result using a high-fidelity simulation. To achieve an error of 10% (measured using the infinitynorm) for capturing local-scale mixing features such as interfacial mixing, only 24%to 32% of the sequence data for model training is required. To achieve the same levelof accuracy for capturing global-scale mixing features, the sequence data required formodel training is 64% to 70% of the total spatial-temporal data. Eosinophilic enteritis presenting as a rare cause for ileo-ileal intussusception

Eosinophilic enteritis,a relatively rare entity,usually involves gastric antrum or proximal small bowel. Our case is rarer in its involvement of the distal small bowel and presents unusually as intussusception. The disease if diagnosed in the initial stages responds well to medical treatment but if associated with complications or misdiagnosed,surgical modality is the treatment of choice. In our case,the patient presented with acute intestinal obstruction due to intussusception and emergency laparotomy with ileoileal anastomosis was done. Histopathology confirmed the diagnosis as eosinophilic enteritis. This case with such a presentation is discussed here.
