The deubiquitinating gene Usp29 is dispensable for fertility in male mice

The balanced actions between ubiquitination and deubiquitination precisely control the levels of various proteins vital for spermatogenesis. Ubiquitin-specific processing proteases(USPs) are the largest family of deubiquitinatingenzymes(DUBs),containing more than 50 members. So far, the functions of only a few USPs in male fertility have been studied, the roles of the majority are yet unknown. The present study aimed to explore the function of Usp29(ubiquitin-specific protease 29) in male fertility. We found that Usp29 showed predominant expression in mouse testis, and its m RNA expression started to increase at 14 days postpartum(dpp), with a peak at 28 and 35 dpp. Using CRISPR/Cas9 technology, we generated Usp29 knockout mice(Usp29^(–/–)). Usp29^(–/–)mice exhibited no overt developmental anomalies. Further examination revealed that Usp29^(–/–)mice had normal fertility and showed no detectable difference in the testis/body weight ratio, testicular and epididymal histology as well as epididymal sperm count from the wild-type littermates. Moreover, Usp29 is not a pseudogene in mice. Taken together, our study first reported that though Usp29 is predominantly expressed in the testis, it is not essential for male fertility in mice.

Meiosis is pivotal for sexual reproduction and fertility. Meiotic programmed DNA double-strand breaks(DSBs) initiate homologous recombination, ensuring faithful chromosome segregation and generation of gametes. However, few studies have focused on meiotic DSB formation in human ***, we report four infertile siblings born to a consanguineous marriage, with three brothers suffering from non-obstructive azoospermia and one sister suffering from unexplained infertility with normal menstrual cycles and normal ovary sizes with follicular activity. An autosomal recessive mutation in TOP6BL was found co-segregating with infertility in this family. Investigation of one male patient revealed failure in programmed meiotic DSB formation and meiotic arrest prior to pachytene stage of prophase *** models carrying similar mutations to that in patients recapitulated the spermatogenic abnormalities of the patient. Pathogenicity of the mutation in the female patient was supported by observations in mice that meiotic programmed DSBs failed to form in mutant oocytes and oocyte maturation failure due to absence of meiotic recombination. Our study thus illustrates the phenotypical characteristics and the genotype-phenotype correlations of meiotic DSB formation failure in humans.

Clinicopathological features and medium-term outcomes of histologic variants of primary focal segmental glomerulosclerosis in adults:A retrospective study

Optimizing Fully Convolutional Encoder-Decoder Network for Segmentation of Diabetic Eye Disease

In the emerging field of image segmentation,Fully Convolutional Networks(FCNs)have recently become ***,their effectiveness is intimately linked with the correct selection and fine-tuning of hyperparameters,which can often be a cumbersome manual *** main aim of this study is to propose a more efficient,less labour-intensive approach to hyperparameter optimization in FCNs for segmenting fundus *** this end,our research introduces a hyperparameter-optimized Fully Convolutional Encoder-Decoder Network(FCEDN).The optimization is handled by a novel Genetic Grey Wolf Optimization(G-GWO)*** algorithm employs the Genetic Algorithm(GA)to generate a diverse set of initial *** leverages Grey Wolf Optimization(GWO)to fine-tune these positions within the discrete search *** on the Indian Diabetic Retinopathy Image Dataset(IDRiD),Diabetic Retinopathy,Hypertension,Age-related macular degeneration and Glacuoma ImageS(DR-HAGIS),and Ocular Disease Intelligent Recognition(ODIR)datasets showed that the G-GWO method outperformed four other variants of GWO,GA,and PSO-based hyperparameter optimization *** proposed model achieved impressive segmentation results,with accuracy rates of 98.5%for IDRiD,98.7%for DR-HAGIS,and 98.4%,98.8%,and 98.5%for different sub-datasets within *** results suggest that the proposed hyperparameter-optimized FCEDN model,driven by the G-GWO algorithm,is more efficient than recent deep-learning models for image segmentation *** thereby presents the potential for increased automation and accuracy in the segmentation of fundus images,mitigating the need for extensive manual hyperparameter adjustments.

All the compositions in the(1-x)Mg_(0.95)Ni_(0.05)Ti_(0.98)Zr_(0.02)O_3–xCa_(0.6)La_(0.8/3)TiO_3(0 ≤ x ≤ 0.2) series were fabricated using solid state sintering route. Mg_(0.95)Ni_(0.05)Ti_(0.98)Zr_(0.02)O_3 possessed excellent microwave dielectric properties with ε_r ≈ 17.1, Q_uf_0 ≈ 195855 GHz, and τ_f ≈-46 ppm/℃. t f was tuned through zero by mixing with Ca_(0.6)La_(0.8/3)TiO_3. In the present study, τ_f ≈-2 ppm/℃ with er ≈ 23.9 and high Q_uf_0 ≈ 115870 GHz was achieved for x = 0.15, i.e., for a mixture of 85% Mg_(0.95)Ni_(0.05)Ti_(0.98)Zr_(0.02)O_3–xCa_(0.6)La_(0.8/3)TiO_3.

Normalized urban heat island (UHI) indicators: Classifying the temporal variation of UHI for building energy simulation (BES) applications

Despite known effects of urban heat island(UHI)on building energy consumption such as increased cooling energy demand,typical building energy simulation(BES)practices lack a standardized approach to incorporate UHI into building energy *** seasonal and diurnal variation of UHI makes the task of incorporating UHI into BES an especially challenging task,often limited by the availability of detailed hourly temperature data at building *** paper addresses the temporal variation of UHI by deriving four normalized UHI indicators that can successfully capture the seasonal and diurnal variation of *** accuracy of these indicators was established across four climate types including hot and humid(Miami,FL),hot and dry(Los Angeles,CA),cold and dry(Denver,CO),and cold and humid(Chicago,IL),and three building types including office,hospital,and *** four indicators are mean summer daytime UHI,mean summer nighttime UHI,mean winter daytime UHI,and mean winter nighttime UHI,which can accurately predict cooling,heating,and annual energy consumption with mean relative error of less than 1%.Not only do these indicators simplify the application of UHI to BES but also,they provide a new paradigm for UHI data collection,storage,and usage,specifically for the purpose of BES.

Plant extract-loaded bacterial cellulose composite membrane for potential biomedical applications

Bacterial cellulose(BC)has been extensively explored as biomaterial for various biomedical applications owing to its non-toxic nature and unique structural morphology and impressive physicochemical and mechanical ***,its high production cost and lack of antimicrobial activity have restricted its large-scale production and therapeutic ***,the current study is aimed to devise a strategy for low-cost BC production and develop its composite with bioactive materials to bless it with antimicrobial ***,5 mm thick reticulated fibrous and highly porous BC was produced by utilizing the wasted rotten tomatoes as the production *** produced bacterial cellulose waste(BCW)(i.e.,produced from wastes)was ex-situ modified with bioactive plant extract(PE)obtained from Euclea schimperi,and the bactericidal activity of the developed BCW/PE was evaluated against Gram-positive Staphylococcus aureus and Gram-negative Escherichia coli through disc diffusion and colony forming unit(CFU)count *** BCW/PE composite showed high bactericidal activities against *** and produced clear inhibition zone whereas negligible activity was observed against ***,indicating its bactericidal activity mainly against the Gram-positive ***,this study illustrates that there is a huge potential for developing valuable biomaterials from food wastes and utilizing their liquid holding capabilities for value-added applications in medical and pharmaceutical fields.

The role of graveyards in species conservation and beta diversity:a vegetation appraisal of sacred habitats from Bannu,Pakistan

Graveyards or sacred groves are often places of natural vegetation protected by spiritual believers because of their sacred beliefs and indigenous culture.A study of graveyards was conducted to determine their role in species conservation,community formation,and associated indicators and species composition using multivariate statistical *** was hypothesized that variations in the age of graveyards would give rise to diverse plant communities under the impact of various edaphic and climatic *** ecological techniques were applied to determine various phytosociological *** the data were put in MS Excel for analysis in PCORD and CANOCO softwares for cluster analysis(CA),two-way cluster analysis(TWCA),indicator species analysis and canonical correspondence *** and TWCA through Sorenson distance measurements identified five major graveyard plant communities:(1)FicusBougainvillea-Chenopodium;(2)Acacia-Datura-Convolvulus;(3)Ziziphus-Vitex-Abutilon;(4)Acacia-Lantana-Salsola;and(5)*** such as Capparis decidua,Herniaria hirsuta,Salvadora oliedes and Populus euphratica were only present inside graveyards rather than outside and advocate the role of graveyards in species *** impact of different environmental and climatic variables plus the age of the graveyards were also assessed for comparison of plant communities and their respective indicator *** results indicate that higher chlorine concentration,age of graveyards,low soil electrical conductivity,lower anthropogenic activities,higher nitrogen,calcium and magnesium concentrations in the soil,and sandy soils were the strong environmental variables playing a significant role in the formation of graveyard plant communities,their associated indicators and species distribution *** results could further be utilized to evaluate the role of edaphic and climatic factors,indicator species and conservation management practices at a gr

Myeloproliferative neoplasms(MPNs)occur due to the abnormal proliferation of one or more terminal myeloid cell lines in peripheral *** suffering from MPNs display a high burden of cardiovascular risk factors,and thrombotic events are often the cause of death in this population of ***,we provide a brief overview of dyslipidemia and metabolic syndrome and their epidemiology in MPNs and examine the common molecular mechanisms between dyslipidemia,metabolic syndrome,and MPNs,with a special focus on cardio-vascular risk,atherosclerosis,and thrombotic ***,we investigate the impact of dyslipidemia and metabolic syndrome on the occurrence and survival of thrombosis in MPN patients,as well as the management of dyslipidemia in MPNs,and the impact of MPN treatment on serum lipid concentrations,particularly as side/adverse effects reported in the context of clinical trials.
