Introduction of Multiscaled Longitudinal Vortices by Fractal-Patterned Surface Roughness

To determine the type of surface roughness pattern that is suitable for adaptive suppression of the drag of an obstacle, we observed flow structures introduced by such obstacles. Several roughness patterns were tested: geometric patterns, fractal patterns, reptile-skin patterns, and patterns of circular cylinders arranged in a lattice and in a zigzag manner. A suitable pattern for adaptive control of flow is one that generates longitudinal vortices with nonconstant distances. The preferred instability mode of a laminar boundary layer is expected to be selected automatically from fluctuations involving many frequencies and caused by fractal patterns. Snake- and reptile-skin patterns may have a similar ability as fractal patterns because they consist of multiscale patterns. The longitudinal vortices generated from peculiar positions and concave corners in patterns were observed. The distance between these vortices is not constant because the onset of vortices is at concave corners in fractal patterns. These vortices have differing strengths and easily cause nonlinear interactions, so they can disturb a laminar boundary layer with several higher-harmonic frequencies. The velocity profiles of the laminar boundary-layer flow over the fractal patterns were measured by using hydrogen bubbles. The results show the down-wash flow between the longitudinal vortices, which means that the vortices may effectively suppress the boundary layer separation in an adverse pressure gradient.

An effective method for laboratory acoustic emission detection and location using template matching

In this paper, a template matching and location method, which has been rapidly adopted in microseismic research in recent years, is applied to laboratory acoustic emission(AE) monitoring. First, we used traditional methods to detect P-wave first motions and locate AE hypocenters in three dimensions. In addition, we selected events located with sufficient accuracy(normally corresponding AE events of relatively larger energy, showing clear P-wave first motion and a higher signal-to-noise ratio in most channels) as template events. Then, the template events were used to scan and match other poorly located events in triggered event records or weak events in continuous records. Through crosscorrelation of the multi-channel waveforms between the template and the event to be detected, the weak signal was detected and located using a grid-searching algorithm(with the grid centered at the template hypocenter). In order to examine the performance of the approach, we calibrated the proposed method using experimental data of different rocks and different types of experiments. The results show that the proposed method can significantly improve the detection capability and location accuracy, and can be applied to various laboratory and in situ experiments, which use multi-channel AE monitoring with waveforms recorded in either triggering or continuous mode.

Curvature-driven lipid sorting: coarse-grained dynamics simulations of a membrane mimicking a hemifusion intermediate

How membrane curvature influences lipid distribution is under intensive research. In this short report, after a brief review of recent studies, the results of our coarse-grained (CG) molecular dynamics simulations of membranes with “hemifused ribbons” geometry are discussed. When membranes of a binary mixture of (dipalmitoyl-phosphatidylcholine (DPPC) / diol-eoyl-phosphatidylethanolamine (DOPE) were used, DOPE accumulated in the negatively curved region of the monolayer that formed as the proximal monolayers fused (i.e., cis leaflets). However, the enrichment was dependent on the presence of tethering molecules which kept the curvature high (the curvature radius of ~1 nm), placing the cis monolayers ~2-2.5 nm from each other. Simulations in which DOPE was replaced with dioleoyl-phosphatidylcholine (DOPC) showed an insignificant degree of DOPC accumulation, suggesting the importance of lateral interaction among DOPE molecules for the curvature sorting. The above composition was not close to a demixing point and our radial distribution function analysis suggested that the DOPE accumulation was not assisted by the lipid phase separation which has been shown to promote curvature-driven lipid sorting. Relevance of curvature-driven lipid sorting to biological membrane fusion is discussed.

Coarse-grained simulations of branched bilayer membranes: effects of cholesterol-dependent phase separation on curvature-driven lipid sorting

Our recent coarse-grained (CG) molecular dynamics (MD) simulations of membranes with a hemifused-ribbon (λ-shaped) geometry showed curvature-driven demixing leading to enrich ment in dioleoyl-phosphatidylethanolamine (DOPE) in a negatively-curved region (at C = –0.8 nm–1) of a DOPE/dipalmitoyl-phosphati-dylcholine (DPPC) membrane. Here we extend the analysis with respect to lipid composition and simulation time. Simulations of 12 – 20 μs effective time show that, compared with DOPE of the DOPE/DPPC system, a DPPC/dilinoleyl-PC [di(18:2)PC] membrane showed a similar degree of enrichment of di(18:2)PC in the curved region with C=–0.8 nm–1. For the latter mixture, even weak negative curvatures (C=–0.5 – 0.6 nm–1) caused significant degrees of di(18:2)PC enrichment. In agreement with recent studies of a planar bilayer, a ternary DPPC/ di(18:2)PC/cholesterol 0.42:0.28:0.3 mixture phase-separated into nanoscale raft-like liquid-ordered (Lo) and non-raft liquid-disordered (Ld) phases on a sub-microsecond time scale. The Lo domains were preferentially localized at planar portions, whereas the Ld domains were positioned mainly in curved regions of the membrane. Unlike binary dioleoylphosphatidylcho-line (DOPC)/cholesterol and DPPC/cholesterol mixtures, which showed only a slight enrich ment of cholesterol in the curved region, the ternary mixtures showed considerable migra tion of cholesterol and DPPC from the curved to the planar region. A pronounced degree of lipid segregation due to the preferential distribution of the Ld and Lo domains in the curved and planar regions, respectively, was observed even when the curvature of the fused monolayers (originally ‘cis’ leaflets) was weakened (C= –0.5 nm-1). Overall, the results are consistent with theoretical predictions based on spontaneous curvature of the constituent lipids and the difference in rigidity between the Ld and Lo domains, whereas lipid-lipid interactions, such as PE-PE or DPPC-cholesterol, as well as propensity fo

Molecular dynamics simulation analyses of viral fusion peptides in membranes prone to phase transition: effects on membrane curvature, phase behavior and lipid-water interface destabilization

To gain insight into the atomistic details of membrane fusion induced by fusogenic peptides, molecular dynamic simulations of synthetic peptides, derived from viral fusion proteins, contained in lipid bilayers were performed. A 20 amino acid peptide from the N-terminus of the influenza HA fusion peptide (WT20) assumed the oblique orientation at the interface between water and the membrane made up of dipalmitoylphosphatidylcholine (DPPC)/palmitic acid (PA), as reported previously for different membranes. Simulations of WT20 embedded in bilayer membranes made up of dioleoylphos-phatidylethanolamine (DOPE) and DPPC/PA showed a positive curvature-inducing effect, whereas WT20 showed a negative curvature-inducing effect on a DPPC bilayer. In phase re-constitution analyses starting from a random mixture of DPPC, PA and water molecules, WT20 weakly stabilized an inverted hexagonal phase. In the latter analyses WT20 preferentially assumed a transmembrane orientation as opposed to the interfacial orientation, regardless of the phase to which the system settled (lamellar vs. inverted hexagonal). In another set of analyses using systems containing a water layer between the apposed DPPC/PA (and DOPE) monolayers, the behavior of WT20 during the formation of an intermembrane connection (or stalk) was examined. Comparison among the mutants supports a view that the oblique orientation of WT20 facilitates the perturbation of the lipid-water interface and the stalk formation. Taken together, these results imply that the influenza HA fusion peptide can have substantial effects on the membrane curvature and can assume a wide range of orientation/position in membranes depending on the local environment of the lipid/water system. Its movability and oblique orientation appear to be associated with its ability to perturb membrane/water interfaces.

Changes in Secondary Metabolites and Free Amino Acid Content in Tomato with <i>Lamiaceae</i>Herbs Companion Planting

The present experiment was conducted to evaluate the influence of Lamiaceae herbs companion planting on growth and secondary metabolites changes in tomato plants. Furthermore, the free amino acid changes in tomato due to companion planting were also evaluated using tomato-basil companionship as a model. Four-week-old seedlings of tomato were grown in a pot containing autoclaved commercial soil with basil, peppermint and hyssop as a companion plant separately in different density. Four weeks after companion planting, tomato plants under 1:1 companionship with the herbs showed significant increase in dry weights of shoots compared to control. Higher density of the herbs on the other hand expressed a growth suppression on tomato possibly due to nutrient competition. By the LC-MS analysis, shikimic acid and apigenin were identified as the major secondary compounds found in tomato plants and 1:1 companionship with basil seemed to have a positive influence on their content in tomato shoots. On the other hand, in case of peppermint and hyssop, the increase was observed in all parts of tomato plants. In addition, promotion in several free amino acid contents was also observed in tomato plants with basil companion planting compared to control. Thus, tomato plants grown with herb companion planting in 1:1 ratio seems to have a positive impact on growth of tomato. This positive influence might be related to the increase in some secondary metabolites and changes in the free amino acids observed in this study.

Observations of Wood Cell Walls with a Scanning Probe Microscope

Scanning Probe Microscopes (SPMs) observe specimen surfaces with probes by detecting the physical amount of a material between the cantilever and the surface. SPMs have a high resolution and can measure mechanical characteristics such as stiffness, adsorptive properties, and viscoelasticity. These features make it easy to identify the surface structure of complex materials;therefore, the use of SPMs has increased in recent years. Wood cell walls are primarily composed of cellulose, hemicellulose, and lignin. It is believed that hemicellulose and lignin surround the cellulose framework;however, their detailed formation remains unknown. Therefore, we observed wood cell walls via scanning probe microscopy to try to reveal the formation of the cellulose framework. We determined that the size of the cellulose microfibril bundle and hemicellulose lignin module composite was 18.48 nm based on topography and that the size of the cellulose microfibril bundle was 15.33 nm based on phase images. In the viscoelasticity image, we found that the viscoelasticities of each cell wall of the same cell were not the same. This is because the cellulose microfibrils in each cell wall lean in different directions. The angle between the leaning of the cellulose microfibril and the cantilever affects the viscoelasticity measurement.

BACKGROUND The success of liver resection relies on the ability of the remnant liver to *** of the knowledge regarding the pathophysiological basis of liver regeneration comes from rodent studies,and data on humans are ***,there is limited knowledge about the preoperative factors that influence postoperative *** To quantify postoperative remnant liver volume by the latest volumetric software and investigate perioperative factors that affect posthepatectomy liver *** A total of 268 patients who received partial hepatectomy were *** were grouped into right hepatectomy/trisegmentectomy(RH/Tri),left hepa-tectomy(LH),segmentectomy(Seg),and subsegmentectomy/nonanatomical hepatectomy(Sub/Non)*** regeneration index(RI)and late rege-neration rate were defined as(postoperative liver volume)/[total functional liver volume(TFLV)]×100 and(RI at 6-months-RI at 3-months)/RI at 6-months,*** lower 25th percentile of RI and the higher 25th percentile of late regeneration rate in each group were defined as“low regeneration”and“delayed regeneration”.“Restoration to the original size”was defined as regeneration of the liver volume by more than 90%of the TFLV at 12 months *** The numbers of patients in the RH/Tri,LH,Seg,and Sub/Non groups were 41,53,99 and 75,*** RI plateaued at 3 months in the LH,Seg,and Sub/Non groups,whereas the RI increased until 12 months in the RH/Tri *** to our multivariate analysis,the preoperative albumin-bilirubin(ALBI)score was an independent factor for low regeneration at 3 months[odds ratio(OR)95%CI=2.80(1.17-6.69),P=0.02;per 1.0 up]and 12 months[OR=2.27(1.01-5.09),P=0.04;per 1.0 up].Multivariate analysis revealed that only liver resection percentage[OR=1.03(1.00-1.05),P=0.04]was associated with delayed ***,multivariate analysis demonstrated that the preoperative ALBI score[OR=2.63(1.00-1.05),P=0.02;per 1.
