Safety and efficacy of photodynamic therapy using BCECF-AM compared to mitomycin C in controlling post-operative fibrosis in a rabbit model of subscleral trabeculectomy

AIM： To evaluate the safety and efficacy of cellular photoablation using BCECF-AM [2, T-bis-（2-carboxyethyl） -5-（and-6）-carboxyfluorescein, acetoxymethyl ester mixed isomersl as a method to control postoperative fibrosis in subscleral trabeculectomy （SST） compared to mitomycin C （MMC） in a rabbit model. METhODS： A comparative prospective case-control animal study was conducted. Fourteen rabbits were subjected to SST with intraoperaUve use of wound modulating agents （MMC or BCECF-AM） of the right eye （study groups I and II respectively） and SST without use of intraoperative wound modulating agents for the left eye （control group II）. Two rabbits 4 eyes were considered as control group I with no surgical intervention. BCECF-AM was injected subconjunctivally 30min before surgery followed by intraoperative illumination with diffuse blue light for 10min. Antifibrotic efficacy was established by clinical response and histological examination. Clinical response was assessed by measuring intraocular pressure （lOP） at day 1, 3, 5, 7, 14, 21 postoperatively, Success was defined by 〉20.0% reduction in lOP from the preoperative values without anti-glaucoma medications. RESULTS： The mean percentage of reduction was 35.0% in the study group I with only one eye （14.3%） had 12.5% reduction. The mean percentage of reduction was 28.0% in the study group U with two eyes （28.6%） in study group II had 14.2% reduction each. Regarding the control group II, the mean percentage of reduction was 14.3% with 64.3% eyes had 〈20.0% reduction. Magnetohydrodynamic Effect on Free Convection of Three Dimensional Laminar Flow in Porous Annulus

A numerical study has been carried out to investigate heat transfer by free convection under the effect of MhD （magnetohydrodynamic） for steady state three-dimensional laminar flow in horizontal and vertical cylindrical annulus filled with saturated porous media （sand silica） with fins attached to the inner cylinder. A single electric coil placed around the inner cylinder to generate a magnetic field. The governing equations which used are continuity, momentum （using Darcy＇s law） and energy equations which are transformed to dimensionless equations. The finite difference approach is used to obtain all the computational results using Fortran 90 program. The parameters affected on the system are Rayleigh number ranging within （102 ~ Ra＊〈 104）, and MhD （Mn）（0 〈_ Mn 〈_ 100） and radius ratio Rr （0.225, 0.338 and 0.435）. The results obtained are presented graphically in the form of streamline and isotherm contour plots and the results show that heat transfer decrease with the increase of magnetohydrodynamic. It was found that the average Nusselt number increase with Ra＊ and decrease with h~ Mn and Rr. A correlation for the average Nusselt number in terms of Ra＊ and Mn, has been developed for the inner cylinder.

Purification and Biochemical Characterization of N-methyl-histidylesterase from Camel （Camelus dromedarius） Liver

Camel Liver Esterase （LE） was isolated through five consecutive steps： Extraction with Tris/hCl buffer, ph 8, precipitation with ammonium sulfate, affinity chromatography on Affi-gel-butyric acid and on Affi-gel-oleic acid, and preparative polyacrylamide gel electrophoresis （PAGE）. The Km values were found as： methyl butyrate （8.3 mM）, α-naphthyl acetate （3.65 mM）, 13-naphthyl myristate （66.7 mM）, p-nitrophenyl acetate （0.29 mM）, and phenyl acetate （5.26 mM）. The Ki of LE inhibition by bis（4-nitrophenyl） phosphate was 7.9 p.M, and 58 μM by phenyl-methyl-sulfonyl fluoride. The inhibition by these inhibitors is irreversible. p-hydroxymercuribenzoate or ethylenediamine-tetra-acetic acid did not inhibit the enzyme. LE showed a dimeric structure with molecular weight of 129 kD. The energy of activation of LE was 15.0, 5.5 and 10.75 Kcal, using the substrates： a-naphthyl acetate, p-nitrophenyl acetate, and methyl butyrate, respectively. The optimal ph for LE was between 8 and 10. The N-terminus was found as aspartic acid. The percentage of glycine residues （13.3%） was the highest whereas the percentage of cysteine residues （0.68%） was the lowest in LE. Amino acid composition shows that LE has -50% of its histidine residues as N-methylhistidines.

Nigella sativa oil as a treatment for gingivitis: A randomized active-control trial

Objective: To assess the clinical anti-inflammatory and antimicrobial efficacy of Nigella sativa oil compared with chlorhexidine in patients with gingivitis. Methods: A double-blind, randomized clinical trial was conducted in patients having chronic generalized gingivitis. Patients were randomly assigned to receive Nigella sativa oil(n=18) or chlorhexidine(n=19). The following assessments were made on day 0 and day 15: plaque index, gingival index, gingival IL-6 and IL-18 levels were measured using ELISA, plaque colony-forming units, and alpha-hemolytic Streptococcus strains. Data were analyzed using parametric and non-parametric tests and Fisher’s exact ***: Both interventions reduced plaque index and gingival index scores(Phe Nigella sativa oil group was better at lowering IL-6(P=0.007 6) than the chlorhexidine group(P=0.145), although there was no change in IL-18 levels(P>0.05). The post-intervention plaque index and gingival index scores and inflammatory cytokine levels between the two groups were not significantly different. Both interventions caused a significant reduction in the plaque colony-forming units(Phogenic bacteria: Streptococcus mitis, Streptococcus oralis, Streptococcus sanguinis, and Streptococcus parasanguinis in the chlorhexidine group(50%)(P=0.103 1), and the Nigella sativa oil group(20%)(P=0.739 5). Conclusions: Nigella sativa oil had anti-inflammatory and antibacterial activities, reducing biofilm formation and disrupting the colonization of pathogenic bacteria essential for the progression of periodontal disease. Nigella sativa oil could offer an alternative therapy for treating gingivitis and may prevent associated systemic diseases and improve overall health outcomes.

An Overview of Nanoparticles from Medicinal Plants: Synthesis, Characterization and Bio-Applications

Presenting and definition the most important points about nanoparticles for medicinal plants and the most important vital applications of them. The scoping review was performed according to systemic style. The research articles included the selected studies dealing with primary information on nanotechnology and medicinal plants published between 2000 and 2023. The search was at grassroots platforms such as Web of Science and PubMed. Many studies correlated the properties of plants’ nanoparticles such as stability, surface area, and high reactivity, and their small size. It was found that nanoparticles (NPs) have size ranging from some nanometers to 100 nm and their morphology is controlled because of their tiny size, NPs have a big surface area, which makes them suitable for many applications. Green nanotechnology has the potential to become an industry with very high green credentials as it is increasingly commercialized. In general, nanoparticles derived from medicinal plants offer a promising avenue for various bio-applications. Their green synthesis, biocompatibility, and potential therapeutic properties make them an attractive area of research with the potential to impact fields ranging from medicine to agriculture.

Melatonin and/or rowatinex attenuate streptozotocin-induced diabetic renal injury in rats

The study aimed to explore the prophylactic effect of melatonin, rowatinex; a naturally occurring renal drug, and its combination on diabetic nephropathy in type 2 diabetic rats. Diabetes was induced by intraperitoneal injection of a single dose of streptozotocin(50 mg/g body weight). Three days before diabetes induction, rats were daily treated with melatonin, rowatinex and their combination continuously for 8 weeks. Evaluation was done through measuring blood urea nitrogen(BUN), serum uric acid, serum creatinine, urine creatinine, creatinine clearance, nitric oxide(NO), malondialdehyde(MDA), superoxide dismutase(SOD), glutathione(GSh), total antioxidant capacity(TAC),kidney injury molecule-1(KIM-1), heat shock protein-70(hSP-70), caspase-3, transforming growth factor β1(TGFβ1), DNA degradation by the comet assay and total protein contents. histopathologic study was also done for the kidney and the pancreas. Drastic changes in all measured parameters of the diabetic rats were observed. Treatment with melatonin and rowatinex showed amelioration to variable degrees. In conclusion, melatonin showed the most potent effect on protecting rats from deleterious action of diabetic nephropathy followed by its combination with rowatinex.

Appendectomy in Pediatrics the Value of Peritoneal Fluid Smear and Its Bacteriological Profile

Introduction: Acute appendicitis is the most common cause of abdominal pain requiring surgery in children. In most instances the infecting organisms are normal inhabitants of the lumen of appendix. Surgery allows easy microbiological sampling. Aspiration of fluid or pus in a syringe is preferred. Swabs are less suitable and only to be used when sampling with a syringe is not feasible. Antimicrobial susceptibility testing of the isolated bacteria and particularly of the anaer- obes can be important to adjust therapy in case of the presence of multi resistant bacteria. Objective: The aim of the study was to determine the bacteriological profile of acute appendicitis in children. Materials and Methods: Study design is a prospective descriptive study including children hospitalized for acute appendicitis. Tissue samples (a speci- men of the appendix), peritoneal fluid swab from the appendicial fossa and the peritoneal exudates (if exists) obtained at surgery from 54 children with suspected acute appendicitis operated at the pediatric surgery unit at the Maternity and Child Teaching hospital in Al-Qadisiya province from the period 1st of June 2007 to the end of May 2011, were exam- ined histologically and by culture for aerobic and anaerobic bacteria. Results: Out of these, 39 boys (72.2%) and 15 (27.7% girls). Their age ranged between (1.8 - 13) years, with a mean of 6.9 years. Of the all patients studied 34 pre- sented with suppurative and phlegmnous appendicitis, 8 with gangrenous appendicitis, 6 with septic complications of appendicitis and 6 had normal appendices. Only 80 of the 108 swabs taken yielded a positive culture (74.07%). Con- clusion: Although in our study no antibiotic regime was changed on the basis of a positive culture swab and the perito- neal culture swabs do not improve immediate postoperative therapy based on surgical impression and rapid histological reporting, however, the routine use of peritoneal culture swabs may be of value in identifying pat
