Prion diseases are a group of neurodegenerative diseases that are fatal. The study of these unique diseases in China is hampered by a lack of resources. Amongst the most important resources for biological study are monoclonal antibodies. Here, we characterize a panel of monoclonal antibodies specific for cellular prion protein by enzyme-linked immunosorbent assay(ELISA), immunofluorescent staining, flow cytometry, and western blotting. We identify several antibodies that can be used for specific applications and we demonstrate that there is no prion protein expression in human pancreatic ductal epithelial cells(HPDC).

Lactobacillus plantarum J26 alleviates alcohol-induced oxidative liver injury by regulating the Nrf2 signaling pathway

Oxidative stress is one of the main ways to cause alcohol-induced liver injury,and alcoholic liver disease(ALD)has been a common health problem *** acid bacteria(LAB)is also considered as a potential treatment to alleviate alcohol-induced liver *** plantarum J26 is a LAB isolated from Chinese traditional fermented dairy products with excellent probiotic *** study aimed to establish a mice model of alcoholic liver injury through acute-on-chronic alcohol feeding and to study the alleviating effect of pre-intake of *** J26 on alcohol-induced oxidative liver injury and focus on its potential mechanism of alleviating *** results showed that pre-intake of *** J26 could improve liver pathological changes,reduce lipid accumulation,increase mitochondrial ATP and mitochondrial(mtDNA)levels,and alleviate liver *** addition,pre-intake *** J26 can improve the level of short-chain fatty acids(SCFAs)in the intestines in mice,short chain fatty acids can be used as a signaling molecule activation of nuclear factor E2-related factor 2(Nrf2)signaling pathway to alleviate liver oxidative stress,and maintain mitochondrial homeostasis by regulating the expression of genes related to mitochondrial dynamics and autophagy,thereby reducing cell apoptosis to alleviate alcohol-induced oxidative liver injury.

V0405 Dra is a W UMa-type binary *** on the TESS data,we have conducted an orbital period study and performed a light curve analysis for the *** orbital period study reveals that the O-C curve for V0405 Dra exhibits secular decrease at an extremely high rate of d P/dt=-2.71×10^(-6)day year^(-1),along with periodic variations characterized by an amplitude of A_(3)=0.0032 day and a period of P_(3)=1.413 *** orbital periodic change is possibly due to the light-travel time effect resulting from an additional third body in the system,for which we estimate a minimum mass of M_(3)=0.77M_(⊙).By employing the 2013 version of the Wilson-Devinney(W-D)method to synthesize a light curve,we derived photometric solutions indicating that V0405 Dra is a new deep(f=68.7%)and low-mass ratio(q=0.175)contact *** fast decrease in its orbital period is likely caused by mass transfer from the more massive primary star to the less massive secondary star,or due to angular momentum *** further mass transfer and loss of angular momentum,the binary will gradually evolve into a tighter contact configuration,eventually leading to a merger into a single star,following the evolutionary paths suggested for such deep and low mass ratio contact binaries.

The notorious dendrite and infinite volume change seriously restrict the advancement of lithium metal anodes(LMAs),during the long-term process of stripping/***,the nanosheets of metal fluoride(CoF_(2))and metal nitride(CoN)with magnificent lithiophilicity on the nickel(Ni)foam are designed as the“regulator”to uniform the Li plating and build stronger solid electrolyte interface(SEI)layer for dendrite free *** Ni foam offers abundant space to receive deposited Li *** CoN nanosheets can guarantee the fast transfer of electrons,which provides a stable interface of Li^(+)***,the nanosheet structure with lithiophilicity would accelerate the move of Li+and decrease the nucleation barrier,due to the high lattice-matching of Li and ***,the CoF_(2) could increase the content of F(LiF)in the SEI layer,which enhances the strength and avoids the destruction of SEI *** the cooperation of CoN and CoF_(2),the composited anode(Li/NF@CNCF)exhibits ultra-long cycle performance(more than 1200 h)and fantastic structure stability at 1 mA·cm^(-2) with 1 mAh·cm^(-2).Based on the LiFePO_(4) and Li/NF@CNCF,the full cells deliver excellent specifical capacity and steady coulombic *** strategy contributes an effective approach to alleviate the issues of lithium metal anodes in the field of LMAs.

Gut-liver-brain axis is a three-way highway of information interaction system among the gastrointestinal tract,liver,and nervous *** the past few decades,breakthrough progress has been made in the gut liver brain axis,mainly through understanding its formation mechanism and increasing treatment *** this review,we discuss various complex networks including barrier permeability,gut hormones,gut microbial metabolites,vagus nerve,neurotransmitters,immunity,brain toxic metabolites,β-amyloid(Aβ)metabolism,and epigenetic regulation in the gut-liver-brain *** therapies containing antibiotics,probiotics,prebiotics,synbiotics,fecal microbiota transplantation(FMT),polyphenols,low FODMAP diet and nanotechnology application regulate the gut liver brain ***,some special treatments targeting gut-liver axis include farnesoid X receptor(FXR)agonists,takeda G protein-coupled receptor 5(TGR5)agonists,glucagon-like peptide-1(GLP-1)receptor antagonists and fibroblast growth factor 19(FGF19)*** gut-brain axis embraces cognitive behavioral therapy(CBT),antidepressants and tryptophan metabolism-related *** liver-brain axis contains epigenetic regulation and Aβmetabolism-related *** the future,a better understanding of gut-liver-brain axis interactions will promote the development of novel preventative strategies and the discovery of precise therapeutic targets in multiple diseases.

Although metal oxide compounds are considered as desirable anode materials for potassium-ion batteries(PIBs)due to their high theoretical capacity,the large volume variation remains a key issue in realizing metal oxide anodes with long cycle life and excellent rate *** this study,polypyrroleencapsulated Sb_(2)WO_(6)(denoted Sb_(2)WO_(6)@PPy)microflowers are synthesized by a one-step hydrothermal method followed by in-situ polymerization and coating by *** the nanosheet-stacked Sb_(2)WO_(6)microflower structure,the improved electronic conductivity,and the architectural protection offered by the PPy coating,Sb_(2)WO_(6)@PPy exhibits boosted potassium storage properties,thereby demonstrating an outstanding rate property of 110.3 m A h g^(-1)at 5 A g^(-1)and delivering a long-period cycling stability with a reversible capacity of 197.2 m A h g^(-1)after 500 cycles at 1 A g^(-1).In addition,the conversion and alloying processes of Sb_(2)WO_(6)@PPy in PIBs with the generation of intermediates,K_(2)WO_(4)and K_(3)Sb,is determined by X-ray photoelectron spectroscopy,transmission electron microscopy,and exsitu X-ray diffraction during potassiation/*** functional theory calculations demonstrate that the robust coupling between PPy and Sb_(2)WO_(6)endues it with a much stronger total density of states and a built-in electric field,thereby increasing the electronic conductivity,and thus effectively reduces the K^(+)diffusion barrier.

Tumor Necrosis Factor α(TNFα) is best known as a mediator of inflammation and immunity, and also plays important roles in tumor biology. However, the role of TNFα in tumor biology is complex and not completely understood. In a human melanoma cell line, M2, and a lung carcinoma cell line, A549, TNFα up-regulates prion protein(PrP) level, and promotes tumor cell migration in a PrP dependent manner. Silencing PRNP abrogates TNFα induced tumor cell migration;this phenotype is reversed when PRNP is re-introduced. Treatment with TNFα activates nuclear factor kappa B(NF-κB)signaling, which then mitigates autophagy by reducing the expression of Forkhead Box P3(FOXP3). Down regulation of FOXP3 reduces the transcription of synaptosome associated protein 29(SNAP29), which is essential in the fusion of autophagosome and lysosome creating autolysosome. FOXP3 being a bona fide transcription factor for SNAP29 is confirmed in a promoter binding assay. Accordingly, silencing SNAP29 in these cell lines also up-regulates PrP, and promotes tumor cell migration without TNFα treatment. But, when SNAP29 or FOXP3 is silenced in these cells, they are no longer respond to TNFα. Thus, a reduction in autophagy is the underlying mechanism by which expression of PrP is up-regulated,and tumor cell migration is enhanced upon TNFα treatment. Disrupting the TNFα-NF-κB-FOXP3-SNAP29 signaling axis may provide a therapeutic approach to mitigate tumor cell migration.

A universal cooperative assembly-oriented strategy for VS4 nanorod decoration on carbon nanostructures with enhanced magnesium storage properties

VS4has a unique layered atomic chain structure and has the potential to become a high-performance cathode material of magnesium-ion batteries with a high capacity and long cycle life. However, low conductivity and sluggish Mg2+diffusivity during cycling limit its practical application in large-scale energy storage. Herein, a cooperative assembly-directed strategy is adopted to synthesize VS4nanorods grown in situ on carbon nanotubes（CNTs/VS4）. VS4nanorods are tightly anchored to CNTs through V–O–C interface covalent bonds, and CNTs can enhance the electronic conductivity of the nanocomposite. In addition,the ion insertion reaction using Mg2+and Mg Cl+as carriers reduces the polar barrier for divalent Mg2+ion transport. This rationally designed architecture promotes ion diffusion and electron transfer, thus facilitating reaction kinetics. The cooperative assembly-oriented strategy can endow CNTs/VS4with excellent magnesium storage properties, including a high reversible capacity of 223.2 m Ah g-1at a current density of 50 m A g-1, a remarkable discharge capacity of 91.8 m Ah g-1even at 2,000 m A g-1, and an impressive capacity retention of 85.2% after 1,000 cycles at 500 m A g-1. Moreover, this strategy can serve as a general synthetic method for the complexation of VS4with other carbon nanostructures.
