Ferroptosis inhibitors reduce celastrol toxicity and preserve its insulin sensitizing effects in insulin resistant HepG2 cells

Objective:Research has shown that celastrol can effectively treat a variety of diseases,yet when passing a certain dosage threshold,celastrol becomes toxic,causing complications such as liver and kidney damage and erythrocytopenia,among *** this dichotomy in mind,it is extremely important to find ways to preserve celastrol’s efficacy while reducing or preventing its ***:In this study,insulin-resistant Hep G2(IR-Hep G2)cells were prepared using palmitic acid and used for in vitro ***-Hep G2 cells were treated with celastrol alone or in combination with Nacetylcysteine(NAC)or ferrostatin-1(Fer-1)for 12,24 or 48 h,at a range of *** counting kit-8assay,Western blotting,quantitative reverse transcription-polymerase chain reaction,glucose consumption assessment,and flow cytometry were performed to measure celastrol’s cytotoxicity and whether the cell death was linked to ***:Celastrol treatment increased lipid oxidation and decreased expression of anti-ferroptosis proteins in IR-Hep G2 *** downregulated glutathione peroxidase 4(GPX4)m *** docking models predicted that solute carrier family 7 member 11(SLC7A11)and GPX4 were covalently bound by ***,we found for the first time that the application of ferroptosis inhibitors(especially NAC)was able to reduce celastrol’s toxicity while preserving its ability to improve insulin sensitivity in IR-Hep G2 ***:One potential mechanism of celastrol’s cytotoxicity is the induction of ferroptosis,which can be alleviated by treatment with ferroptosis *** findings provide a new strategy to block celastrol’s toxicity while preserving its therapeutic effects.

Calcium alumino-titanate(CAT)can be used as a raw material owing to its excellent refractoriness and thermal ***-containing taphole clay(THC)with different particle sizes was prepared and *** results indicated that THC with CAT aggregate has better thermo-mechanical performance than that with a fine CAT powder because of different sintering *** CAT aggregate and SiO_(2)reacted and formed liquid phase covering their surface,enhancing the bonding of the particle–matrix and promoting the internal transformation of the ***,the mullite was formed in aggregate with the interwoven network *** CAT powder has a higher specific surface area,and its surface energy decreases with the particle *** could cause a reaction between CAT and SiO_(2)at relatively low temperature,making it unable to form a core–shell structure similar to that of the aggregate,with the excessive liquid phase affecting the performance of the ***,the liquid phase was formed to close the pores and cover the SiC surface,and this was found to enhance the oxidation resistance.A corrosion test confirmed that the THC refractories with CAT aggregates showed better corrosion resistance than other *** electron microscopy and energy-dispersive spectrometry results indicated that Ti(C,N)transformed by TiO_(2)in CAT was deposited at the interface between the molten slag and the refractory,and it efficiently prevented the penetration and reaction owing to its high melting point and small wetting angle with the slag.

Based on fundamental principle of wind turbine and Betz theory, the wind turbine characteristic curve is discussed in this paper. The curve pointed to the fundamental theory of three points comparing algorithm. A novel improved algorithm is put forward to overcome the shortcomings and deficiencies of normal method applied in the wind turbine. The correctness and feasibility of the method are fully verified by MATLAB simulation experiment.

Knockout Brown Norway （BN） rat could be a useful disease model for human disorders,however,a failure to derive embryonic stem （ES） cells disturbs the further development of the *** this study,we reported a case of successful derivation of the BN rat ES cells with the derivation efficiency comparable to that of Sprague Dawley （SD） *** BN rat ES cells expressed the key transcription factors,and were able to form embryonic bodies （EBs） when being differentiated in *** injecting the BN rat ES cells into the SD rat blastocysts,high-contribution chimeric rats were generated and could survive to their *** success in generating pluripotent rat ES cells will benefit the generation of the knockout rats in the future.

Periodic Variations of Drag Coefficient for the ANDE Spherical Satellites During its Lifetime

A drag coefficient(C_d) inversion method is introduced to study the variations of the drag coefficient for orbital satellites with spherical *** coefficients of the four micro satellites in the Atmospheric Neutral Density Experiment(ANDE) are compiled out with this new method. The Lomb-Scargle Periodgram(LSP) analysis of the four ANDE satellites' C__d series has shown that there are obvious 5,7,9,and 27 days' period in those *** results are found through comparing the LSP analysis with series of the daily solar radio flux at 10.7cm(F_(10.7) index),the Ap index,and the daily averaged solar wind speed at *** series in the same time interval have an obvious period of about 27 days,which has already been explained as the association with the 27 days' solar *** oscillating periods less than 27 days are found in series of C_D,Ap and solar wind speed at 1 AU,e.g.,the 5,7,9 days ***,these short periods disappeared in the time series of F_(10.7) *** same periodicities of 5,7,9 days in Ap and solar wind are presented at the same time interval during the declining phase of solar cycle *** in the ascending phase of solar cycle 24,these short oscillations are not so obvious as that in the declining phase of solar cycle *** results provide definite evidence that the C_d variations with period of 5,7 and 9 days are produced by a combination of space weather effects caused by the solar wind and geomagnetic activity.

Fusarium wilt is one of the most serious diseases of banana plants caused by soil-borne pathogen Fusarium oxysporum ***. cubense(FOC). In this study a pot experiment was conducted to evaluate the effects of different bio-organic fertilizers(BIOs) on Fusarium wilt of banana, including the investigations of disease incidence, chitinase and β-1,3-glucanase activities of banana plants, and FOC populations as well as soil rhizosphere microbial community. Five fertilization treatments were considered, including chemical fertilizer containing the same N, P and K concentrations as the BIO(control), and matured compost mixed with antagonists Paenibacillus polymyxa SQR-21 and Trichoderma harzianum T37(BIO1), Bacillus amyloliquefaciens N6(BIO2), Bacillus subtilis N11(BIO3), and the combination of N6 and N11(BIO4). The results indicated that the application of BIOs significantly decreased the incidence rate of Fusarium wilt by up to 80% compared with the control. BIOs also significantly promoted plant growth, and increased chitinase andβ-1,3-glucanase activities by 55%–65% and 17.3%–120.1%, respectively, in the banana roots. The population of FOC in the rhizosphere soil was decreased significantly to about 104 colony forming units g-1with treatment of BIOs. Serial dilution plating and denaturing gradient gel electrophoresis analysis revealed that the application of BIOs increased the densities of bacteria and actinomycetes but decreased the number of fungi in the rhizosphere soil. In general, the application of BIOs revealed a great potential for the control of Fusarium wilt disease of banana plants.

Three dimensional velocity structure and accurate earthquake location in Changning–Gongxian area of southeast Sichuan

In order to understand the crustal structure and tectonic background of the Changning–Gongxiang area, southeastern Sichuan Province, where a series of moderate-to-strong earthquakes occurred in recent years, we utilized the seismic phase data both from a local dense array and from the regional seismic networks;we used the tomoDD program to invert for the high-resolution three-dimensional velocity structure within the depth range of 0–10 km and for accurate hypocentral locations in this area. We analyzed the seismogenic structures for the events of Xingwen M5.7 in 2018 and Gongxian M5.3 and Changning M6.0 in 2019. The results show that:(1) widespread lateral inhomogeneity exists in the velocity structure of the study area, and the location of the velocity anomaly is largely consistent with known structures. In the range of distinguishable depth, the inhomogeneity decreases with increasing depth, and the velocity structure anomalies in some areas are continuous in depth;(2) earthquakes occurred in clusters, showing the characteristics of zonal folding trends in the NW-SE and NE-SW directions;the focal depth in the area is generally shallow in both the sedimentary cap and the crystalline basement. The seismogenic structures of small earthquake clusters are different in size and occurrence in different sections, and the clusters occurred mostly in regions with high P-or S-wave velocities;(3) synthesis of a variety of data suggests that the seismogenic structures of the Xingwen M5.7 and Changning M6.0 earthquakes are associated with slip faults that trend NW-SE in, respectively, the south wing and the axis of the Changning–Shuanghe anticline, while that of the Gongxian M5.3 earthquake is associated with thrust faults that trend N-S in the Jianwu syncline region. The dynamic sources of the three earthquakes are all from the SE pushing of the qinghai–Tibet block on the Sichuan basin;(4) the risk of future strong earthquakes in this area must be reevaluated in light of the

Neuroinflammation,commonly associated with various central nervous system(CNS)diseases such as postoperative cognitive dysfunction(POCD),is primarily mediated by the disruption of biological signals in ***,the effective treatment of CNS diseases remains an ongoing challenge as biological signals show limited microglia-targeting *** this study,taking advantage of the highly expressed lipoprotein receptor-related protein-1(LRP1)on the microglia,a nanobiosignal delivery system modified by LRP1 high-affinity peptide ligand RAP12(RAP:receptor-associated protein)was constructed to specifically regulate neuroinflammation via targeting *** uptake of the RAP12 modified-nanobiosignaler by microglia increased significantly,indicating its microglia-targeting *** in vitro/vivo studies proved that the“nanobiosignaler”significantly reduced the secretion of pro-inflammatory cytokines,induced specific M2(anti-inflammatory type)microglia differentiation,and remarkably alleviated cognitive function impairment in the mice model when compared with unmodified *** was indicated that the“nanobiosignaler”could target microglia to deliver the biological signal and inhibit the excessive activation of ***,the cell-targeted biological signal transmission system inspired by“nanobiosignaler”has broad application prospects in the future.

Several studies have suggested that interleukin(IL)-15 is a promising adjuvant that promotes cellular immunity when administered with human immunodeficiency virus(HIV)*** we evaluated the effect of IL-15 plasmid on HIV-specific immune responses,especially cellular immunity,in eight rhesus *** monkeys were immunized three times with HIV DNA vaccine with or without IL-15 plasmid and boosted with recombinant Tiantan strain vaccinia virus-based HIV vaccine(rTV)22 weeks after the first *** we did not detect any significant differences in the HIV-specific CD81 T-cell response between monkeys with IL-15 coimmunization and monkeys with HIV vaccine alone,our results showed that the frequency of effector CD8^(+) memory T cells in the peripheral blood was significantly higher in monkeys with IL-15 coimmunization than those with HIV vaccine alone at almost all of the time points ***,the titers of anti-HIV antibodies were higher in Group T than those in Group C after rTV *** findings in rhesus monkeys suggest that IL-15 may be useful as a cytokine adjuvant for HIV vaccine.
