Peptidic defective interfering gene nanoparticles against Omicron,Delta SARS-CoV-2 variants and influenza A virus in vivo

Defective interfering genes(DIGs)are short viral genomes and interfere with wild-type viral ***,we demonstrate that the new designed SARS-CoV-2 DIG(CD3600)can significantly inhibit the replication of SARS-CoV-2 including Alpha,Delta,Kappa and Omicron variants in human HK-2 cells and influenza DIG(PAD4)can significantly inhibit influenza virus replication in human A549 *** dose of influenza DIGs prophylactically protects 90%mice from lethal challenge of A(H1N1)pdm09 virus and CD3600 inhibits SARS-CoV-2 replication in hamster lungs when DIGs are administrated to lungs one day before viral *** further investigate the gene delivery vector in the respiratory tract,a peptidic TAT2-P1&LAH4,which can package genes to form small spherical nanoparticles with high endosomal escape ability,is demonstrated to dramatically increase gene expression in the lung ***2-P1&LAH4,with the dual-functional TAT2-P1(gene-delivery and antiviral),can deliver CD3600 to significantly inhibit the replication of Delta and Omicron SARS-CoV-2 in hamster *** peptide-based nanoparticle system can effectively transfect genes in lungs and deliver DIGs to inhibit SARS-CoV-2 variants and influenza virus in vivo,which provides the new insight into the drug delivery system for gene therapy against respiratory viruses.

siRNA, miRNA and HIV: promises and challenges

Small interfering RNA (siRNA) and microRNA (miRNA) are small RNAs of 18-25 nucleotides (nt) in length that play important roles in regulating gene expression. They are incorporated into an RNA-induced silencing complex (RISC) and serve as guides for silencing their corresponding target mRNAs based on complementary base-pairing. The promise of gene silencing has led many researchers to consider siRNA as an anti-viral tool. However, in long-term settings, many viruses appear to escape from this therapeutical strategy. An example of this may be seen in the case of human immunodeficiency virus type-1 (HIV-1) which is able to evade RNA silencing by either mutating the siRNA- targeted sequence or by encoding for a partial suppressor of RNAi (RNA interference). On the other hand, because miRNA targeting does not require absolute complementarity of base-pairing, mutational escape by viruses from miRNA- specified silencing may be more difficult to achieve. In this review, we discuss stratagems used by various viruses to avoid the cells’ antiviral si/mi-RNA defenses and notions of how viruses might control and regulate host cell genes by encoding viral miRNAs (vmiRNAs).

An Analysis of Hong Kong Commercial Culture under the Influence of Mainland Visitors

The colonial history of Hong Kong created her own special culture,this kind of special culture can be found in many areas,including language,education,politics,justice and commercial sector. From the time when the economic opening of China's Mainland in 1978,the interaction and cooperation between mainland and Hong Kong have became closer and closer in the past few *** since handover in 1997,the change of migration policies of both governments(mainland and Hong Kong) leaded the growing number of mainland visitors to Hong *** Kong commercial culture is therefore changed by the mainland *** commercial sector,they also used to adjust their culture in order to adopt the needs of mainland visitors. This research is aimed to investigate the contemporary(visible) commercial culture of Hong Kong retail sale and catering *** whether their commercial culture has been adjusted to be an appropriate level in order to serve mainland visitors or *** site-visiting and participated observation were used as research *** experiment would be done in tourist-concentrated areas and control experiment would be done in non-tourist-concentrated *** research and several measuring indicators would be used to assess the commercial culture of those retail sale shops and *** function of control experiment,which would be done in non-tourist-concentrated areas,is aimed to compare and analyze the different commercial culture between tourist-concentrated and non-tourist-concentrated areas. Based on our data analysis,readers can know much about the contemporary commercial cultural change in Hong *** parts were noted and suggestions would be recommended to related official departments and Hong Kong commercial areas.

Appended corrole manganese complexes:Catalysis and axial-ligand effect

A series of N-base appended corroles and their manganese complexes were synthesized and their binding constants with three different nitrogenous ligands, triethylamine, N-methylimidazole and pyridine, were evaluated by spectroscopy. Kinetic studies indicated that the presence of appended N- donor ligands may cause a significant enhancement of the rate of oxygen atom transfers （OAT） from （oxo）manganese（V） corrole to alkene, and the stronger axial ligand binding has impact on the rate of the oxidation reaction. Turnover frequency （TOF） for the catalytic oxidation of alkenes by appended manganese corroles varies with the following ligand order： acetamido 〈 pyridyl 〈 imidazolyl. The influence of the external axial ligands on the catalytic epoxidation was investigated by using appended acetamido manganese corrole as catalyst, with the results revealing that N-methylimidazole gave the best enhancement on the yields of total oxidation products among the investigated nitrogenous ligands.

Probiotic-based nanoparticles for targeted microbiota modulation and immune restoration in bacterial pneumonia

While conventional bacterial pneumonia mainly centralizes avoidance of bacterial colonization,it remains unclear how to restore the host immunity for hyperactive immunocompetent primary and immunocompromised secondary bacterial ***,probiotic-based nanoparticles of OASCLR were formed by coating chitosan,hyaluronic acid and ononin on living Lactobacillus *** nanoparticles could effectively kill various clinic common pathogens and antibacterial efficiency was> 99.97%.Importantly,OASCLR could modulate lung microbiota,increasing the overall richness and diversity of microbiota by decreasing pathogens and increasing probiotic and commensal ***,OASCLR could target inflammatory macrophages by the interaction of OASCLR with the macrophage binding site of CD 44 and alleviate overactive immune responses for hyperactive immunocompetent ***,OASCLR could break the state of the macrophage’ s poor phagocytic ability by upregulating the expression of the extracellular matrix assembly,immune activation and fibroblast activation in immunocompromised *** macrophage’ s phagocytic ability was increased from 2.61% to 12.3 %.Our work provides a potential strategy for hyperactive immunocompetent primary and immunocompromised secondary bacterial pneumonia.

Theophylline(TP)is a very well established orally or intravenously delivered antiasthma drug with many beneficial *** study aims to improve asthma treatment by creating a dry powder inhalable(DPI)formulation of TP to be delivered directly to the lung,avoiding the side effects associated with conventional oral *** DPI TP formulation was investigated for its physico-chemical characteristics using scanning electron microscopy,laser diffraction,thermal analysis and dynamic vapour ***,aerosol performance was assessed using the Multi Stage Liquid Impinger(MSLI).In addition,a Calu-3 cell transport assay was conducted in vitro using a modified ACI to study the impact of the DPI formulation on lung epithelial *** showed DPI TP to be physico-chemically stable and of an aerodynamic size suitable for lung *** aerosolisation performance analysis showed the TP DPI formulation to have a fine particle fraction of 29.70±2.59%(P<0.05)for the TP formulation containing 1.0%(w/w)sodium stearate,the most efficient for *** the deposition of TP DPI on Calu-3 cells using the modified ACI,results demonstrated that 56.14±7.62%of the total TP deposited(13.07±1.69μg)was transported across the Calu-3 monolayer over 180 min following deposition,while 37.05±12.62%of the deposited TP was retained in the *** could be due to the presence of sodium stearate in the current formulation that increased its lipophilicity.A DPI formulation of TP was developed that was shown to be suitable for inhalation.
