BACKGROUND Primary sclerosing cholangitis(PSC)is characterized by chronic inflammation and it predisposes to cholangiocarcinoma due to lack of effective treatment options.Recombinant adeno-associated virus(rAAV)provides a promising platform for gene therapy on such kinds of diseases.A microRNA(miRNA)let-7a has been reported to be associated with the progress of PSC but the potential therapeutic implication of inhibition of let-7a on PSC has not been evaluated.AIM To investigate the therapeutic effects of inhibition of a miRNA let-7a transferred by recombinant adeno-associated virus 8(rAAV8)on a xenobiotic-induced mouse model of sclerosing cholangitis.METHODS A xenobiotic-induced mouse model of sclerosing cholangitis was induced by 0.1% 3,5-Diethoxycarbonyl-1,4-Dihydrocollidine(DDC)feeding for 2 wk or 6 wk.A single dose of rAAV8-mediated anti-let-7a-5p sponges or scramble control was injected in vivo into mice onset of DDC feeding.Upon sacrifice,the liver and the serum were collected from each mouse.The hepatobiliary injuries,hepatic inflammation and fibrosis were evaluated.The targets of let-7a-5p and downstream molecule NF-κB were detected using Western blot.RESULTS rAAV8-mediated anti-let-7a-5p sponges can depress the expression of let-7a-5p in mice after DDC feeding for 2 wk or 6 wk.The reduced expression of let-7a-5p can alleviate hepato-biliary injuries indicated by serum markers,and prevent the proliferation of cholangiocytes and biliary fibrosis.Furthermore,inhibition of let-7a mediated by rAAV8 can increase the expression of potential target molecules such as suppressor of cytokine signaling 1 and Dectin1,which consequently inhibit of NF-κB-mediated hepatic inflammation.CONCLUSION Our study demonstrates that a rAAV8 vector designed for liver-specific inhibition of let-7a-5p can potently ameliorate symptoms in a xenobiotic-induced mouse model of sclerosing cholangitis,which provides a possible clinical translation of PSC of human.

Organic long-persistent luminescence(LPL)materials,featuring low preparation cost,eco-friendly synthesis,and easy modification of functional groups,have exhibited extensive applications in information encryption,anti-counterfeiting,and biological imaging.Several design strategies including crystallization-inducement,H-aggregation,and host-guest doping to enhance persistent-room-temperature phosphorescence(RTP)effect by precisely controlling intersystem crossing(ISC)constant and suppressing nonradiative decay rates,those are important strategies to enable LPL performance.Among the strategies,researchers have made several efforts to enhance persistent-RTP effect by host-guest interaction,in which the host matrices provide a rigid environment for phosphor guest molecules.The interaction of the luminescent guest molecules with the host matrix can effectively reduce the vibration and rotation of the luminescent molecules,and suppress the non-radiative inactivation,thereby improving the phosphorescence quantum yield.This review aims to summarize several design strategies of pure organic LPL materials based on persistent-RTP effect through host-guest interaction,and describe some applications of pure organic LPL materials in different fields.

V0405 Dra: A New Deep and Low Mass Ratio Contact Binary with Extremely Fast Decrease in the Orbital Period

V0405 Dra is a W UMa-type binary star. Based on the TESS data, we have conducted an orbital period study and performed a light curve analysis for the system. The orbital period study reveals that the O-C curve for V0405 Dra exhibits secular decrease at an extremely high rate of d P/dt =-2.71 × 10-6day year-1, along with periodic variations characterized by an amplitude of A3= 0.0032 day and a period of P3= 1.413 years. The orbital periodic change is possibly due to the light-travel time effect resulting from an additional third body in the system, for which we estimate a minimum mass of M3= 0.77M⊙. By employing the 2013 version of the Wilson-Devinney（W-D） method to synthesize a light curve, we derived photometric solutions indicating that V0405 Dra is a new deep（ f = 68.7%） and low-mass ratio（q = 0.175） contact binary. The fast decrease in its orbital period is likely caused by mass transfer from the more massive primary star to the less massive secondary star, or due to angular momentum loss. With further mass transfer and loss of angular momentum, the binary will gradually evolve into a tighter contact configuration, eventually leading to a merger into a single star, following the evolutionary paths suggested for such deep and low mass ratio contact binaries.

BACKGROUND Despite continuous changes in treatment methods,the survival rate for advanced hepatocellular carcinoma(HCC)patients remains low,highlighting the importance of diagnostic methods for HCC.AIM To explore the efficacy of texture analysis based on multi-parametric magnetic resonance(MR)imaging(MRI)in predicting microvascular invasion(MVI)in preoperative HCC.METHODS This study included 105 patients with pathologically confirmed HCC,categorized into MVI-positive and MVI-negative groups.We employed Original Data Analysis,Principal Component Analysis,Linear Discriminant Analysis(LDA),and Non-LDA(NDA)for texture analysis using multi-parametric MR images to predict preoperative MVI.The effectiveness of texture analysis was determined using the B11 program of the MaZda4.6 software,with results expressed as the misjudgment rate(MCR).RESULTS Texture analysis using multi-parametric MRI,particularly the MI+PA+F dimensionality reduction method combined with NDA discrimination,demonstrated the most effective prediction of MVI in HCC.Prediction accuracy in the pulse and equilibrium phases was 83.81%.MCRs for the combination of T2-weighted imaging(T2WI),arterial phase,portal venous phase,and equilibrium phase were 22.86%,16.19%,20.95%,and 20.95%,respectively.The area under the curve for predicting MVI positivity was 0.844,with a sensitivity of 77.19%and specificity of 91.67%.CONCLUSION Texture analysis of arterial phase images demonstrated superior predictive efficacy for MVI in HCC compared to T2WI,portal venous,and equilibrium phases.This study provides an objective,non-invasive method for preoperative prediction of MVI,offering a theoretical foundation for the selection of clinical therapy.

6,6’-Bieckol induces apoptosis and suppresses TGF-β-induced epithelial-mesenchymal transition in non-small lung cancer cells

Objective:In this study,the aim was to investigate the inhibitory effect of 6,6’-bieckol on the migration and epithelial-mesenchymal transition(EMT)of non-small cell lung cancer(NSCLC)cells,and explore its potential molecular mechanisms.Methods:Cell migration was measured using a CCK8,wound healing,and transwell migration assay.Apoptosis was determined using an Annexin V/propidium iodide staining.Western blotting and immunofluorescence were used to examine the expression level of apoptosis-related proteins and EMT marker proteins.Results:The results showed that 6,6’-bieckol inhibited migration and induced apoptosis of NSCLC cells.Furthermore,6,6’-bieckol had significantly up-regulated the E-cadherin and down-regulated Snail1 and Twist1 transcriptional levels.6,6’-Bieckol might inhibit TGF-β-induced EMT by down-regulating Snail1 and Twist1 and up-regulating E-cadherin in lung cancer cells.Conclusion:It is suggested that 6,6’-bieckol has the potential to be developed as a therapeutic candidate for lung cancer.

Chro-austempering treatment of a medium-carbon high-strength bainitic steel

The integrated processing of chromizing and austempering(termed chro-austempering)treatments was proposed.The microstructure and properties of a medium-carbon high-strength bainitic steel treated by chro-austempering treatments were investigated by metallography,scanning vibrating electrode technique,electrochemical workstation,and microhardness test.The results show that the high-strength bainitic steel with carbide-free bainite as matrix and the chromized layers on surfaces was successfully fabricated by chro-austempering treatment.The hardness of surface layers was about 3.5 times that of the bainite matrix.Meanwhile,the corrosion started from exposed bainitic matrix and proceeded along the depth direction,testifying that the surface corrosion resistance was significantly improved by chro-austempering treatment due to the formation of Cr_(7)C_(3)and(Cr,Fe)7_(7)C_(3)on the surface.

The association between mutations in ubiquitin-specific protease 26(USP26)and male infertility:a systematic review and meta-analysis

During recent decades,the association between mutations in ubiquitin-specific protease 26(USP26)and male infertility remains doubtful.We conducted this meta-analysis to evaluate the association between mutations in USP26 and male infertility according to the Preferred Reporting Items for Systematic Reviews and Meta-Analyses(PRISMA)2020 guidelines.It was registered in the International Prospective Register of Systematic Reviews(PROSPERO;CRD42021225251).PubMed,Web of Science,and Scopus were systematically searched for comparative clinical studies,which were written in English and provided eligible data.Studies were included when they compared USP26 mutations in azoospermic,oligozoospermic,and asthenozoospermic patients with controls with normal sperm parameter values or whose partners had experienced spontaneous pregnancy.Pooled odds ratio(OR)with 95%confidence interval(CI)was calculated with random effect models.Overall,twelve studies with 3927 infertility patients and 4648 healthy controls were included.The association between overall USP26 mutations and infertility was not significant(OR=1.60,95%CI:0.51-5.01).For specific mutations,the pooled ORs were 1.65(95%CI:1.02-2.69)for cluster mutation(including 370-371insACA,494T>C,and 1423C>T),1.80(95%CI:0.35-9.15)for c.576G>A,1.43(95%CI:0.79-2.56)for c.1090C>T,and 3.59(95%CI:2.30-5.59)for c.1737G>A.Our results suggest that several mutations(cluster mutation,c.1737G>A)may play roles in male infertility,while others(c.576G>A and c.1090C>T)do not show notable associations with male infertility.More high-quality clinical researches are needed for validation.

BACKGROUND Benign paroxysmal positional vertigo(BPPV)is a form of temporary vertigo induced by moving the head to a specific position.It is a self-limited,peripheral,vestibular disease and can be divided into primary and secondary forms.Congenital nystagmus(CN),an involuntary,rhythmic,binocular-symmetry,conjugated eye movement,is found at birth or within 3 mo of birth.According to the pathogenesis,CN can be divided into sensory-defect nystagmus and motordefect nystagmus.The coexistence of BPPV and CN is rarely seen in the clinic.CASE SUMMARY A 62-year-old woman presented to our clinic complaining of a 15-d history of recurrent positional vertigo.The vertigo lasting less than 1 min occurred when she turned over,sometimes accompanied by nausea and vomiting.Both the patient and her father had CN.Her spontaneous nystagmus was horizontal to right;however,the gaze test revealed variable horizontal nystagmus with the same degree when the eyes moved.The patient’s Dix-Hallpike test was normal,except for persistent nystagmus,and the roll test showed severe variable horizontal nystagmus,which lasted for about 20 s in the same direction as her head movement to the right and left,although the right-side nystagmus was stronger than the left-side.Since these symptoms were accompanied by nausea,she was diagnosed with BPPV with CN and treated by manual reduction.CONCLUSION Though rare,if BPPV with CN is correctly identified and diagnosed,reduction treatment is comparably effective to other vertigo types.

肠源性IgA^(+)浆细胞是肠道黏膜免疫发挥免疫效应的关键。肠源性IgA^(+)浆细胞的产生受肠道菌群和饮食调控,不仅在肠道黏膜局部发挥免疫作用,还可迁徙至肠道外的组织或器官,通过依赖产生IgA抗体或分泌细胞因子等非抗体产生效应来调节免疫和炎症反应,在多种疾病中发挥免疫调节的保护或者致病效应。肠道黏膜免疫失衡在IgA肾病中的致病效应逐渐被关注。IgA肾病高血清IgA小鼠肠道固有层IgA^(+)浆细胞的增多与循环中异常糖基化IgA1增多有关。IgA肾病患者循环中的长寿命浆细胞数量增加,并抑制白细胞介素-6发挥致病作用。但肠源性IgA^(+)浆细胞是否迁移至肾脏局部促进疾病进展尚不清楚。现就肠源性IgA^(+)浆细胞在IgA肾病肠道黏膜免疫致病效应及机制的相关研究进展进行综述。

Real-time monitoring flexible hydrogels based on dual physically cross-linked network for promoting wound healing

To achieve smart and personalized medicine, the development of hydrogel dressings with sensing properties and biotherapeutic properties that can act as a sensor to monitor of human health in real-time while speeding up wound healing face great challenge. In the present study, a biocompatible dual-network composite hydrogel(DNCGel) sensor was obtained via a simple process. The dual network hydrogel is constructed by the interpenetration of a flexible network formed of poly(vinyl alcohol)(PVA) physical cross-linked by repeated freeze-thawing and a rigid network of iron-chelated xanthan gum(XG) impregnated with Fe^(3+) interpenetration. The pure PVA/XG hydrogels were chelated with ferric ions by immersion to improve the gel strength(compressive modulus and tensile modulus can reach up to 0.62 MPa and0.079 MPa, respectively), conductivity(conductivity values ranging from 9 × 10^(-4) S/cm to 1 × 10^(-3)S/cm)and bacterial inhibition properties(up to 98.56%). Subsequently, the effects of the ratio of PVA and XG and the immersion time of Fe^(3+) on the hydrogels were investigated, and DNGel3 was given the most priority on a comprehensive consideration. It was demonstrated that the DNCGel exhibit good biocompatibility in vitro, effectively facilitate wound healing in vivo(up to 97.8% healing rate) under electrical stimulation, and monitors human movement in real time. This work provides a novel avenue to explore multifunctional intelligent hydrogels that hold great promise in biomedical fields such as smart wound dressings and flexible wearable sensors.
