Development of Alfalfa (Medicago sativa L.) Regeneration System and Agrobacterium-Mediated Genetic Transformation

The regeneration ability of four alfalfa （Medicago sativa L.） cultivars, Xinjiang Daye, Longdong, Gannong 1 and Gannong 3, was studied, and the effects of various cultivars, explant sources and medium recipes on regeneration were compared. The better callus forming frequency obtained from hypocotyls of Xinjiang Daye is 88.5% and regeneration frequency is 9.8% in our initial experiments. To further optimize regeneration system for genetic transformation, we therefore changed concentrations of plant growth regulators and supplemented with glutamine into callus-induction and shoot-regeneration media. Callus forming frequency and shoot differentiation frequency were increased to 100%. The time taken to generate transgenic plants （16 weeks） was shorter than that for previouse procedure （25 weeks） and regeneration frequency was promoted to 15.1%. The results show that addition of glutamine is particularly important for shortening period of regeneration and promoting regeneration frequency. For study of genetic transformation of alfalfa, Agrobacterium tumefaciens-mediated transformation of Xinjiang Daye was developed based on this optimized regeneration system. The plant expression vector carrying two glutamine synthetases （GS 1 and GS2） and △1-pyrroline-5-carboxylate synthetase （P5CS） gene was used for alfalfa in vitro transformation. Six transgenic alfalfa plantlets with resistance to PPT were obtained. The introduction of foreign genes into plants was assessed in the transformants by PCR analysis and Southern hybridizations.

In this study,high entropy metallic glasses(HEMGs)were developed through a combination of concepts for designing metallic glasses(main element+transition metal+metalloid element)and high-entropy alloys(more than five elements,each element having an atomic concentration between 5 at.%and 35 at.%).The developed metallic glass alloys are composed of Co-Fe-Ni main elements,transition metals(Cr,Mo)and metalloid elements(C,B).Moreover,the present work reports the thermal,mechanical and magnetic properties of(CoFeNi)(50)(CrMo)(50-x)(CB)x alloys with x=20,25,*** developed as-spun HEMGs exhibit typical paramagnetic properties even for a high amount of ferromagnetic elements(Co,Fe,and Ni)and have high elastic modulus(103–160 GPa)and hardness(14–27 GPa),thus possessing mechanical properties similar to well-known Co-based metallic glasses(Co-Cr-Mo-C-B system).In addition,some of the bulk samples prepared with a diameter of 2 mm form bulk metallic glasses with a high compressive strength around 3.5 *** mechanisms determining the stability of the supercooled liquid,as well as the paramagnetic and mechanical properties for the developed non-equiatomic HEMGs,are discussed.

Background The exact pathophysiological mechanism of transient global amnesia(TGA)is *** is debatable whether TGA is a risk factor for ***,here we investigated the possibility of TGA as a risk factor for stroke in a real-world setting using large-scale nationwide health claims *** We used health claims data from the Korean National Health Insurance Service(NHIS).Patients diagnosed with TGA between 2007 and 2013 were *** initially extracted patients without TGA who were preferentially matched for age and sex with the patients with TGA at a ratio of 10:1 from the whole ***,we performed 1:2 propensity score matching analysis to balance the baseline characteristics between the two *** the propensity score-matched dataset,we performed multivariable Cox regression analysis to investigate the association between TGA and stroke type,including ischaemic,haemorrhagic and all stroke *** Patients with TGA(n=14673)were selected from the NHIS *** extracting from the whole database(n=140486)and propensity score matching their data at a 1:2 ratio,a total of 10448 and 20442 patients were finally assigned to the TGA and control groups,*** multivariable Cox regression analysis demonstrated that the TGA group had a higher risk of ischaemic stroke and all types of stroke(adjusted HR=1.194;95%CI:1.043 to 1.368;and HR=1.197;95%CI:1.056 to 1.357,respectively).Conclusions Analysis of the nationwide claims database showed that TGA could be an important risk factor for stroke,especially for ischaemic stroke.

Wireless sensor nodes have the advantage of being low-cost,easily deployed and of good *** deployed in an underground mine with existing underground transmission systems a wireless sensor network can improve the collection of *** get good transmission performance for 2.4 GHz wireless sensor nodes at the working face we calculated the reflection properties of electromagnetic waves from a flat metal *** the cascade impedance method(CIM),we studied transmission attenuation and compared the results to actual *** results show that the effective transmission distance of 2.4 GHz wireless sensor nodes meets the stipulations of the ZigBee protocol.

Voltage-gated potassium channels comprise 12 subtypes （Kv1-Kv12）. Kv2.1, which is expressed in most mammalian central neurons, provides the majority of delayed-rectifier K＋ current in cortical and hippocampal pyramidal neurons, and plays an especially prominent role in repolarizing membrane potential, as well as in facilitation of exocytosis. Kv2.1-encoded K＋ efflux is essential for neuronal apoptosis programming. The human form of the Kv2.1 potassium channel contains large intracellular regions. The cytoplasmic C-terminal plays a key role in modulating Kv2.1 gating. The present manuscript summarized Kv2.1 structure and modulation in neurons and analyzed the roles of the cytoplasmic C-terminal.

Genome Analysis of the Bombyx mori Infectious Flacherie Virus Isolated in China

We reported here the complete genome of the Bombyx mori infectious flacherie virus （BmlFV）, BmlFV-CHN01, isolated in China and compared it with the Japanese strain IFV （BmIFV-JAN） sequence. BmIFV-JAN and BmIFV-CHN01 were 99% identical in nucleotide and amino acid sequences. The amino acid sequences of the three structural proteins （VP1, VP3 and VP4） and L protein were 100% identical between the two strains. Phylogenetic analyses based on conserved domains in RNA-dependent RNA polymerases （RdRps） by the neighbor-joining method showed that these two strains were from the same virus. The gain of the whole genome of the BmIFV isolated in China and its weak mutation character established a great foundation to the study of functional genome and the molecular epidemiology of BmIFV.
