Interleukin-12 as a Genetic Adjuvant Enhances Hepatitis C Virus NS3 DNA Vaccine Immunogenicity

Hepatitis C virus (HCV) chronic infection is a worldwide health problem, and numerous efforts have been invested to develop novel vaccines. An efficient vaccine requires broad immune response induction against viral proteins. To achieve this goal, we constructed a DNA vaccine expressing nonstructural 3 (NS3) gene (pcDNA3.1-HCV-NS3) and assessed the immune response in C57BL/6 mice. In this study, the NS3 gene was amplified with a nested-reverse transcriptase-polymerase chain reaction (RT-PCR) method using sera of HCV-infected patients with genotype 1a. The resulting NS3 gene was subcloned into a pcDNA3.1 eukaryotic expression vector, and gene expression was detected by western blot. The resultant DNA vaccine was co-administered with interleukin-12 (IL-12) as an adjuvant to female C57BL/6 mice. After the final immunizations, lymphocyte proliferation, cytotoxicity, and cytokine levels were assessed to measure immune responses. Our data suggest that co-administration of HCV NS3 DNA vaccine with IL-12 induces production of significant levels of both IL-4 and interferon (IFN)-γ (p<0.05). Cytotoxicity and lymphocyte proliferation responses of vaccinated mice were significantly increased compared to control (p<0.05). Collectively, our results demonstrated that co-administration of HCV NS3 and IL-12 displayed strong immunogenicity in a murine model.

In this paper,21 substituents with various electron donating and electron withdrawing characters were placed in the available positions of Licoisoflavone in order to study their effect on the three O–H bond dissociation enthalpies and ionisation potential via DFT/B3LYP *** show that the B-ring is more important than the A-ring from the BDE and IP point of view because the radical structure formed after hydrogen atom abstraction from the 4-OH group can be stabilized by the resonance between the three *** results show that intramolecular hydrogen bonding effects are able to considerably stabilize the parents and *** NBO analysis results also confirmed the intramolecular hydrogen bond *** formation of strong intramolecular hydrogen bonds and suitable spin density distributions in several radicals result in low BDE and IP *** calculated BDE and IP values have been correlated with Hammett *** found dependencies are suitably linear,which can be important for the synthesis of novel antioxidants based on Licoisoflavone.

Optimal control homotopy perturbation method for cancer model

In this paper, we introduced the optimal control homotopy perturbation method (OCHPM) by using the homotopy perturbation method (HPM). Every one, by using of the proposed method, can obtain numerical solutions of mathematical modeling for cancer-immunotherapy. In this paper, in order to prove the preciseness and efficiency of the OCHPM method, we compared the obtained numerical solutions with HPM. The results obtained showed that the OCHPM method is powerful to generate the numerical solutions for some therapeutic models.

Modeling of forest soil and litter health using disturbance and landscape heterogeneity indicators in northern Iran

This paper focuses on the indicators of soil and litter health, disturbance, and landscape heterogeneity as a tool for prediction of ecosystem sustainability in the northern forests of Iran. The study area was divided into spatial homogenous sites using slope, aspect, and soil humidity classes. Then a range of sites along the disturbance gradient was selected for sampling. Chemical and physical indicators of soil and litter health were measured at random points within these sites. Structural equation modeling(SEM) was applied to link six constructs of landscape heterogeneity, three constructs of disturbance(harvest, livestock, and human accessibility), and soil and litter health. The results showed that with decreasing accessibility, the total N and organic matter content of soil increased and effective bulk density decreased. Harvesting activities increased soil organic matter. Therefore, it is concluded that disturbances through harvesting and accessibility inversely affect the soil health. Unexpectedly, it was found that the litter total C and C:N ratio improved with an increase in the harvest and accessibility disturbances, whereas litter bulk density decreased. Investigation of tree composition revealed that in the climax communities, which are normally affected more by harvesting activities, some species like Fagus orientalis Lipsky with low decomposition rate are dominant. The research results showed that changes in disturbance intensity are reflected in litter and soil indicators, whereas the SEM indicated that landscape heterogeneity has a moderator effect on the disturbance to both litter and soil paths.

The electrooxidation of L-cysteine（L-Cys） was studied using a benzoylferrocene（BF） modified multi-wall carbon nanotube paste electrode（BFCNPE） using cyclic voltammetry（CV）,square wave voltammetry（SWV） and chronoamperometry（CHA）.Under optimum pH in CV the oxidation of L-Cys occurs at a potential about 215 mV less positive than that at the surface of unmodified carbon paste *** catalytic oxidation peak currents were dependent on the L-Cys concentration and a linear calibration curve was obtained in the range 0.7-350.0 mmol/L of L-Cys with SWV *** detection limit（3s） was determined as 0.1 mmol/*** method was also used for the determination of L-Cys in some real samples.

Screening for probiotic characters in lactobacilli isolated from chickens revealed the intra-species diversity of Lactobacillus brevis

Considering the importance of the poultry industry and the increasing interest in alternative growth promoters,probiotics are considered as a potential candidate for use in the poultry *** this study,Lactobacillus species were isolated from 21 rectal swabs of 11 healthy 6-day-old and 10 healthy 21-day-old chickens and their fecal and feed *** isolates were characterized and their probiotic characteristics,including resistance to gastric acid and bile salts,biofilm formation and adherence to epithelium or mucus,amylase and protease activity and production of inhibitory compounds,were *** 31 acid and bile resistant lactobacilli,only 2 Lactobacillus brevis and 1 Lactobacillus reuteri strains showed significant probiotic *** isolates indicated detectable attachment to Caco-2 cells and significant antibacterial activities against Gram-positive and Gram-negative ***,phenotypic and genotypic diversity of lactobacilli isolates were studied by Phene Plate(PhP)system(PhP-LB)and random amplified polymorphic DNA(RAPD)-PCR,***-LB results of 24 *** isolates showed a high phenotypic variation among the *** comparison,results of RAPD-PCR highlighted a low ***,it seems that combination of the 2 techniques(PhP and RAPD-PCR)could result in a significant discriminatory power than each of them used alone.

AIM： To find out if a functional promoter polymorphism in the IL-8 gene along with cagA status and polymorphisms in vacA gene influence the type of diseases in Iranian patients infected by Hpylori. METHODS： IL-8 -251 A/T polymorphism was genotyped by oligonucleotide allele specific PCR （ASO-PCR） in a sample of 233 patients with Hpylori infection undergoing upper gastrointestinal endoscopy. The presence of cagA gene and polymorphisms in vacA gene was also determined by PCR. Association of these genetic polymorphisms with the development of gastritis, peptic ulcers as well as gastric cancer was tested. RESULTS： When the patients with different clinical manifestations were compared according to the presence of cagA gene or various vacA genotypes, only the vacA genotypes were significantly different among gastritis, peptic ulcer and gastric cancer patients （χ^2= 17.8; P=0.001）. Furthermore, there was a significant difference in the frequency of IL-8 -251 A/T genotypes between patients with gastric cancer and benign diseases （χ^2=10.47;P=0.005） CONCLUSION： The IL-8 -251 A/T polymorphism and the polymorphisms in H pylor/ vacA gene are involved in limiting the infection outcome to gastritis and peptic ulcer or in favoring cancer onset in Iranian patients.
