Pro-resolving lipid mediator reduces amyloid-β42–induced gene expression in human monocyte–derived microglia

Specialized pro-resolving lipid mediators including maresin 1 mediate resolution but the levels of these are reduced in Alzheimer's disease brain, suggesting that they constitute a novel target for the treatment of Alzheimer's disease to prevent/stop inflammation and combat disease pathology. Therefore, it is important to clarify whether they counteract the expression of genes and proteins induced by amyloid-β. With this objective, we analyzed the relevance of human monocyte–derived microglia for in vitro modeling of neuroinflammation and its resolution in the context of Alzheimer's disease and investigated the pro-resolving bioactivity of maresin 1 on amyloid-β42–induced Alzheimer's disease–like inflammation. Analysis of RNA-sequencing data and secreted proteins in supernatants from the monocyte-derived microglia showed that the monocyte-derived microglia resembled Alzheimer's disease–like neuroinflammation in human brain microglia after incubation with amyloid-β42. Maresin 1 restored homeostasis by down-regulating inflammatory pathway related gene expression induced by amyloid-β42 in monocyte-derived microglia, protection of maresin 1 against the effects of amyloid-β42 is mediated by a re-balancing of inflammatory transcriptional networks in which modulation of gene transcription in the nuclear factor-kappa B pathway plays a major part. We pinpointed molecular targets that are associated with both neuroinflammation in Alzheimer's disease and therapeutic targets by maresin 1. In conclusion, monocyte-derived microglia represent a relevant in vitro microglial model for studies on Alzheimer's disease-like inflammation and drug response for individual patients. Maresin 1 ameliorates amyloid-β42–induced changes in several genes of importance in Alzheimer's disease, highlighting its potential as a therapeutic target for Alzheimer's disease.

The COVID-19 pandemic highlighted the urgent need to strengthen public health systems.In response,the United Nations Disaster Risk Reduction(UNDRR) Public Health System Resilience Scorecard(Scorecard) was applied in workshops across multiple countries.The aim of our research was to explore the workshop findings to develop priority strategies for strengthening public health system resilience.We conducted a workshop from 14 to 16 March 2023,at the UNDRR Global Education and Training Institute in Incheon,Republic of Korea.A sequential modified Delphi method was utilized to develop a set of prioritized resilience strategies.These were drawn from 70 strategies identified from 13 distinct workshops in eight countries.After two surveys,23 strategies were finalized.Ten received ratings of "High" or Very High" from89% of participants.These related to the inclusion of public health risks in emergency plans,integrating multidisciplinary teams into public health,enabling local transport mechanisms,and improving the ability to manage an influx of patients.The Scorecard provides an adaptable framework to identify and prioritize strategies for strengthening public health system resilience.By leveraging this methodology,our study demonstrated how resilience strategies could inform disaster risk reduction funding,policies,and actions.

BACKGROUND Phosphoglyceride crystal deposition disease(PCDD)is a rare acquired disease in which phospholipid crystals deposit in bone and soft tissue long after surgery,trauma,or repeated injections.CASE SUMMARY A 60-year-old-woman was referred to our department because of multiple abdominal masses after open splenectomy for idiopathic thrombocytopenic purpura 29 years earlier.All the masses showed marked fluorodeoxyglucose(FDG)uptake on ^(18)F-fluorodeoxyglucose positron emission tomography(^(18)F-FDGPET)and were strongly suspected to be malignant tumors.Surgical biopsies were performed,and the abdominal masses were found to be aligned vertically,three in a row,along the tissue layers cut in the patient’s previous surgery.Pathological finding of the specimens showed foreign body granuloma consisting of histiocytes and multinucleated giant cells accumulating around needle-like crystals.The crystals were confirmed as phosphoglyceride by Raman spectroscopy,and PCDD was diagnosed.To our knowledge,this is the first report of PCDD diagnosed by Raman spectroscopy.CONCLUSION We made a definitive diagnosis of PCDD in a patient with multiple tumors showing marked FDG uptake on ^(18)F-FDG-PET by incisional biopsy and composition analysis using Raman spectroscopy,a method that has not previously been reported for the diagnosis of PCDD.

Efficacy of ripasudil in reducing intraocular pressure and medication score for ocular hypertension with inflammation and corticosteroid

·AIM:To investigate the efficacy of ripasudil,a Rho kinase inhibitor,in reducing intraocular pressure(IOP)and medication scores of anti-glaucoma drugs in patients with ocular hypertension with inflammation and corticosteroid.·METHODS:The study included 11 patients diagnosed with ocular hypertension with inflammation and corticosteroid,all of whom were prescribed ripasudil eye drops and followed up for at least 2y after the initiation of treatment.IOP was measured using a non-contact tonometer before enrollment and at each follow-up visit.The medication score of glaucoma eye drops was calculated for each patient.·RESULTS:The mean IOP(26.4±2.9 mm Hg before treatment)significantly decreased after ripasudil therapy(13.7±3.3 mm Hg at 3mo)and remained stable in the low-teens during the 2-year follow-up period(P<0.0001).A significant decrease in the medication score was observed at 12mo or later after the initiation of ripasudil therapy(P<0.05).Both baseline medication scores and glaucomatous optic disc change rates were significantly higher in the five eyes that required glaucoma surgery during the 2-year observation period than the 10 eyes that did not require surgery.·CONCLUSION:Our results demonstrate the efficacy of ripasudil,in reducing IOP and the medication score over a 2-year treatment period in patients with ocular hypertension with inflammation and corticosteroid.Our findings also suggest that ripasudil could reduce the IOP in uveitic glaucoma patients with both lower baseline medication score and lower glaucomatous optic disc change rate.

Hypertension is one of the most important modifiable risk factors for stroke,and greater than 50%of all stroke events are estimated to be attributable to elevated blood pressure(BP).Randomized trials and meta-analyses have demonstrated that reducing BP is the most effective and generalizable strategy for preventing recurrent stroke.Based on currently available evidence,BP should be reduced to below 140/90 mmHg in all patients during the chronic post-stroke phase and to below 130/80 mmHg when welltolerated.

Su Manshu＇s 苏曼殊（1884-1918） The Lone Swan （Duanhong lingyanji 断鸿零雁记, 1911, 1912） is best known for a sustained use of subjective voice and a thematic emphasis on tragic love. Critics have often credited the novella＇s intensely tragic narrative for spearheading a new kind of literary subjectivity that became a cornerstone of modern Chinese literature as heralded by the May Fourth critics in the late 1910s and the 1920s. However, very few have examined this new subjectivity as an effect of Su＇s critical engagement with a late Qing nationalist narrative. Su＇s novella was an appropriation of the anti-Manchu revolutionary narrative of a nation, which hinged on a paradoxical mode of envisaging a new China through a temporal return to the past and by means of a tragic sacrifice of the individual. Following a brief analysis of Su＇s early piece published in The People＇s Journal （Minbao 民报）, this article demonstrates how The Lone Swan elaborated on an excess of individual sacrifice, while developing the new, mourning subjectivity as a witness to the unfinished revolutionary enterprise of forging a powerful nation. Su＇s narrative of cultural devastation resonates with Lu Xun＇s （1881-1936） late Qing work, but, in the May Fourth period that immediately followed, this sense of despair would become an unequivocal obiect for overcoming.

Effect of weld line positions on the tensile deformation of two-component metal injection moulding

Knowledge of the mechanical properties of two-component parts is critical for engineering functionally graded components. In this study, mono-and two-component tensile test specimens were metal injection moulded. Three different weld line positions were generated in the two-component specimens. Linear shrinkage of the two-component specimens was greater than that of the mono-component specimens because the incompatibility of sintering shrinkage of both materials causes biaxial stresses and enhances sintering. The mechanical properties of 316L stainless steel were affected by the addition of a coloured pigment used to identify the weld line position after injection moulding. For the two-component specimens, the yield stress and ultimate tensile stress were similar to those of 316L stainless steel. Because 316L and 630(also known as 17-4PH) stainless steels were well-sintered at the interface, the mechanical properties of the weaker material(316L stainless steel) were dominant. However, the elongations of the two-component specimens were lower than those of the mono-component specimens. An interfacial zone with a microstructure that differed from those of the mono-material specimens was observed; its different microstructure was attributed to the gradual diffusion of nickel and copper.

AIM:To examine the effects of long-term proton pump inhibitor (PPI) therapy on body weight (BW) and body mass index (BMI) in patients with gastroesophageal reflux disease (GERD).METHODS: The subjects were 52 patients with GERD and 58 sex-and age-matched healthy controls. GERD patients were treated with PPI for a mean of 2.2 years (range, 0.8-5.7 years), and also advised on lifestyle modifications (e.g. selective diet, weight management). BW, BMI and other parameters were measured at baseline and end of study.RESULTS: Twenty-four GERD patients were treated daily with 10 mg omeprazole, 12 with 20 mg omeprazole, 8 with 10 mg rabeprazole, 5 with 15 mg lansoprazole, and 3 patients with 30 mg lansoprazole. At baseline, there were no differences in BW and BMI between reflux patients and controls. Patients with GERD showed increases in BW (baseline: 56.4±10.4 kg, end: 58.6±10.8 kg, mean±SD, P<0.0001) and BMI (baseline: 23.1±3.1 kg/m2, end: 24.0±3.1 kg/m2, P<0.001), but no such changes were noted in the control group. Mean BW increased by 3.5 kg (6.2% of baseline) in 37 (71%) reflux patients but decreased in only 6 (12%) patients during treatment.CONCLUSION: Long-term PPI treatment was associated with BW gain in patients with GERD. Reflux patients receiving PPI should be encouraged to manage BW through lifestyle modifi cations.

Comparison of Fitness Tracking Using Three Different Smartwatches during Free Activities in Daily Life

Background: Young women of reproductive age experience various physiological changes, which they measure and track using various devices, including fitness trackers and smartwatches. However, fitness tracking assessment methods are ambiguous because they may differ from model to model. Objective: This study aimed to compare the stress level, heart rate, sleep time, number of steps, and distance traveled, which were calculated using fitness tracking methods for daily-life free activity installed in various smartwatches. Materials and Methodology: Healthy women in their 20s to 30s were recruited for this study, which was conducted from December 2021 to June 2022. The finalized participants wore three different smartwatch models (Mi smartband 6, vivosmart®4, and Band 6) simultaneously on their person for 48 hours and performed their daily activities and recorded them on an hour-based activity chart. Each smartwatch’s measured data (e.g., age, height, weight, and oral medications) were extracted into five datasets: heart rate, stress level, number of steps, distance, and sleep time. Data analyses were conducted using Spearman’s rank correlation coefficient ρ (for comparing heart rates) and Bland-Altman plots (for assessing heart rate agreement). The smartwatches’ fitness trackers were compared using the mean absolute percentage error. Results: The correlation coefficient showed that vivosmart®4 and Band 6 had a higher heart rate agreement (ρ = 0.684). The Bland-Altman plots showed high agreement between Band 6, Mi smartband 6, and vivosmart®4. The heart rate measurement method used under free movement was found to be consistent. The examined smartwatches were able to measure heart rate at the same level even under daily-life free movements. Conclusion: Several different smartwatches’ calculated measured values for heart rate had a high agreement.
