Reliability of design approaches for axially loaded offshore piles and its consequences with respect to the North Sea

In the near future, several offshore wind farms are planned to be built in the North Sea. Therefore, jacket and tripod constructions with mainly axially loaded piles are suitable as support structures. The current design of axial bearing resistance of these piles leads to deviant results regarding the pile resistance when different design methods are adopted. Hence, a strong deviation regarding the required pile length must be addressed. The reliability of a design method can be evaluated based on a model error which describes the quality of the considered design method by comparing measured and predicted pile bearing resistances. However, only few pile load tests are reported with regard to the boundary conditions in the North Sea. This paper presents 6 large-scale axial pile load tests which were incorporated within a new model error approach for the current design methods used for the axial bearing resistance,namely API Main Text method and cone penetration test(CPT)-based design methods, such as simplified ICP-05, offshore UWA-05, Fugro-05 and NGI-05 methods. Based on these new model errors, a reliabilitybased study towards the safety was conducted by performing a Monte-Carlo simulation. In addition,consequences regarding the deterministic pile design in terms of quality factors were evaluated. It is shown that the current global safety factor(GSF) prescribed and the partial safety factors are only valid for the API Main Text and the offshore UWA-05 design methods; whereas for the simplified ICP-05,Fugro-05 and NGI-05 design methods, an increase in the required embedded pile length and thus in the GSF up to 2.69, 2.95 and 3.27, respectively, should be considered to satisfy the desired safety level according to DIN EN 1990 of b ? 3.8. Further, quality factors for each design method on the basis of all reliability-based design results were derived. Hence, evaluation of each design method regarding the reliability of the pile capacity prediction is possible.

High demand for firewood leads to overuse of walnut-fruit forests in Kyrgyzstan

After Kyrgyzstan gained independence in 1991, the importance of the primary sector for food- and energy-supply increased significantly. This has led to a discussion about the sustainability of current firewood use. We investigated firewood collection and use practices in three selected villages and analysed differences between the annual increment of woody biomass and firewood consumption for heating during winter months. The calculated individual firewood consumption is on average 3.90 kg/capita/heating day and the calculation of differences between increment and consumption shows that in minimum one village the surrounding forests are overused for firewood collection. Pressure on the forest and the overuse of preferred tree species for firewood can lead to an overuse of the resource and to a decrease in the genetic diversity of these species in the walnut-fruit forests which are considered as a biodiversity hotspot of international significance due to the diversity of woody.

Thinning effects on walnut stem and crown diameter growth and fruiting in the walnut-fruit forests of Kyrgyzstan

The walnut-fruit forests(Juglans regia L.) in southern Kyrgyzstan are considered to be biodiversity hotspots and of considerable economic and ecological importance to local populations. The fruiting ability and stability of these forests are, however, decreasing because of infrequent silvicultural interventions and non-existent natural regeneration. Continuous inventories and walnut harvest measurements were carried out on 18 trial and control plots over the course of 13 year. Results showed that i) thinning had a positive influence on diameter at breast height(dbh) increase, ii) dbh growth and crown area increases are strongly and positively correlated, iii) large crowns enhanced the production of fruits, and iv) temperature conditions had a major influence on the production of fruits. The above-mentioned points are particularly relevant for young plantations but are also valid for relatively old plantations(> 50 years). We conclude that regular thinning substantially improves diameter growth, fruit production and ability of trees to withstand wind and snow damage in both young stands and in older walnut-fruit forests.

Expression of toxin-related human mono-ADP-ribosyltransferase 3 in human testes

Aim： To investigate wether the corresponding protein of mono-ADP-ribosyltransferase 3 （ART3） mRNA is expressed in human testes and, if so, whether the expression is cell type-specific. Methods： ART3 mRNA was determined in human testes and sperm by reverse transcription-polymerase chain reaction （RT-PCR）. The glycosylphosphatidylinositol linkage of ART3 was shown by treating ART3-transfected HEK-293-T cells with phospholipase C. Fluorescent activated cell sorter （FACS）-analyses were used to detect ART3 on mature spermatozoa and immunohistological studies to detect the protein in testes. Results： ART3 protein was shown to be present in testes. It was found on spermatocytes only. It was absent from spermatogonia, spermatids and spermatozoa. The absence of ART3 from spermatozoa was confirmed by FACS-analysis. ART3 protein was detected neither within a seminoma nor on Leydig cells. Conclusion： Here we show for the first time that ART3 protein is expressed in testes in particular on spermatocytes, indicating that ART3 exerts a specific function only required at a particular stage of spermatogenesis.

Spatial and temporal variability of soil water in drylands:plant water potential as a diagnostic tool

Arid and semi-arid regions are characterized by low rainfall and high potential evaporative demand. Here, water is the major limiting factor for plant growth and productivity. Soil and surface hydrology properties （e.g. field capacity, infikration rates） effectively control the water re-distribution in the ecosystem, a fact that is aggravated in arid environments. Information of the spatial and temporal accessibility of soil water in desert ecosystems is limited. The purpose of the studies is the application of plant water potential to estimate the spatial and temporal variations of soil water availability in different arid ecosystems of the Negev （Israel） and southern Morocco. As model plants the evergreen shrubs Retama raetam, Thymelaea hirsuta and trees （Acacia tortilis） were chosen. Seasonal and spatial variations of the pre-dawn water potential （ψpd） were examined as diagnostic tool to determine water availability on the landscape level. The seasonal differences in the pre-dawn water potential were less pronounced on the dune compared to the interdune. This showed a better water availability on the dune slope. Also in the investigated wadis systems spatial differences of the water potential could be detected and related to the vegetation pattern.

On the Impact of Precursor Unfolding during Protein Import into Chloroplasts

Protein translocation across membranes is a fundamental cellular process. The majority of the proteins of organelles such as mitochondria and chloroplasts is synthesized in the cytosol and subsequently imported in a posttranslational manner. The precursor proteins have to be unfolded at least for translocation, but it has also been assumed that they are unfolded during transport to the organelle in the cytosol. Unfolding is governed by chaperones and the translocon itself. At the same time, chaperones provide the energy for the import process. The energetic properties of the chloroplast translocon were studied by import of the Ig-like module of the muscle protein titin fused to the transit peptide of the chloroplast targeted oxygen evolving complex subunit of 33 kDa （OE33）. Our results suggest that p（OE33）titin is folded prior to import and that translocation is initiated by unfolding after having bound to the translocon at the chloroplast surface. Using a set of stabilizing and destabilizing mutants of titin previously analyzed by atomic force microscopy and as passenger for mitochondrial translocation, we studied the unfolding force provided by the chloroplast translocon. Based on these results, a model for translocation is discussed.

Acute myeloid leukemia(AML)is the most common form of acute leukemia in adults and the second most common form of acute leukemia in *** this,very little improvement in survival rates has been achieved over the past few *** is partially due to the heterogeneity of AML and the need for more targeted therapeutics than the traditional cytotoxic chemotherapies that have been a mainstay in therapy for the past 50 *** the past 20 years,research has been diversifying the approach to treating AML by investigating molecular pathways uniquely relevant to AML cell proliferation and *** we review the development of novel therapeutics in targeting apoptosis,receptor tyrosine kinase(RTK)signaling,hedgehog(HH)pathway,mitochondrial function,DNA repair,and c-Myc *** has been an impressive effort into better understanding the diversity of AML cell characteristics and here we highlight important preclinical studies that have supported therapeutic development and continue to promote new ways to target AML *** addition,we describe clinical investigations that have led to FDA approval of new targeted AML therapies and ongoing clinical trials of novel therapies targeting AML survival *** also describe the complexity of targeting leukemia stem cells(LSCs)as an approach to addressing relapse and remission in AML and targetable pathways that are unique to LSC *** comprehensive review details what we currently understand about the signaling pathways that support AML cell survival and the exceptional ways in which we disrupt them.
