Saponin mixture, obtained from Astragalus monspessulanus subsp. monspessulanus (Fabaceae) was investigated for possible protective effect on tert-butyl hydroperoxide (t-BuOOH)-induced cytotoxicity using primary isolated rat hepatocytes. The cells were isolated by two-stepped col-lagenase perfusion. Liver damage was induced by one hour incubation with t-BuOOH (75 μmol·L-1) and discerned by decreased cell viability, increased lactate dehydrogenase (LDH) leakage into the medium, increased production of malondialdehyde (MDA) and depletion of the cell protector glutathione (GSH). Cell pre-incubation with the saponin mixture (1 mg/mL and 5 mg/mL) significantly (p < 0.05) ameliorated t-BuOOH-induced liver damage, judged by preserved cell viability, decreased activity of LDH, decreased MDA production and restoration of GSH. The effect was concentration-dependent, more pronounced in the highest concentration and comparable with those of silymarin, used as a positive control. The observed cytoprotective effect could be explained by the influence of the saponins on the mitochondrial function, disturbed by t-BuOOH toxic metabolites.

This article describes the detection of DNA mutations using novel Au-Ag coated GaN substrate as SERS （surface-enhanced Raman spectroscopy） diagnostic platform. Oligonucleotide sequences corresponding to the BCR-ABL （breakpoint cluster region-Abelson） gene responsible for development of chronic myelogenous leukemia were used as a model system to demonstrate the discrimination between the wild type and Met244Val mutations. The thiolated ssDNA （single-strand DNA） was immobilized on the SERS-active surface and then hybridized to a labeled target sequence from solution. An intense SERS signal of the reporter molecule MGITC was detected from the complementary target due to formation of double helix. The SERS signal was either not observed, or decreased dramatically for a negative control sample consisting of labeled DNA that was not complementary to the DNA probe. The results indicate that our SERS substrate offers an opportunity for the development of novel diagnostic assays.

The problem of contamination of Lerma River in the State of Mexico, México, has been studied in an integral way, since it is one of the rivers of importance due to its geographical situation in the country. José Antonio Dam is the main receiving body of this river that is being contaminated by, industrial wastewater, of which about 70% is inorganic, as well as discharges municipal and agricultural, driving a lot of material in suspension. This dam acts as a regulator of avenues and its water is used for agricultural irrigation and its sediments are used as sludge to improve agricultural soil in the surroundings. One of the studies that have been carried out is to determine the geochemical distribution of heavy metals in the sediments of this dam, as well as its adsorptive capacity towards different metals. It has been observed that these sediments acts as metal contaminants sequester. Knowing the composition and morphology of these sediments can provide information to understand the mobility of different heavy metals it contains, and potential health risk, which can finally lead to the establishment of purification capacity of this sludge.

AIM: To identify predictors of sustained virological response in hemodialysed patients treated by PEGinterferon α for chronic hepatitis C, genotype ***: The sustained virological response(SVR) rate, IL28 B genotype, IFNL4 genotype, initial viral load(IVL) and other pretreatment variables in 39 endstage renal disease patients(ESRD) on maintenance haemodialysis(HD) infected with hepatitis C virus(HCV), genotype 1b, were compared with a control group of 109 patients with normal kidney function treated within the same period. All the patients were treatment nave and had well compensated liver disease. The ESRD patients received 135 μg of PEGylated interferon α-2a(Peg IFN-α) weekly and a reduced dose of ribavirin(RBV) was administered to 23/39 patients with an initial haemoglobin level > 10 g/d L. Control group patients were given standard doses of Peg IFN-α and RBV. SVR was assessed as HCV RNA negativity 24 wk post-treatment. Since the last decade deep brain stimulation has been proposed as an alternative treatment for patients who do not become seizure-free with the current pharmacological treatments and cannot undergo resective surgical procedure. However, the optimal stimulation parameters remain undetermined and active research in humans and animals is necessary. The present study was designed to investigate the effect of unilateral Low Frequency Stimulation (LFS) of hippocampus on seizure development by using the hippocampal rapid kindling method (hRK) in rats. We used male Wistar rats implanted with electrodes in the ventral hippocampus. All rats underwent hRK (biphasic square wave pulses, 20 Hz for 10 seconds) during three consecutive days (twelve stimulations per day). The control group (hRK;n = 6) received only RK stimulus, while the treated group (LFS-hRK;n = 8) received also LFS (biphasic square wave pulses, 1 Hz for 30 seconds) immediately before the RK stimulus, during three consecutive days. At the end of behavioral testing on day 3, 62% (P < 0.05) of the animals receiving LFS treatment were still not fully kindled staying in stages 0-III (P < 0.01). The number of stimulations needed to achieve generalized seizures (stage IV-V of Racine scale) was significantly higher (P < 0.05) in the LFS group with respect to control group. No significant differences in the cumulative daily afterdischarge duration were observed between both groups. These findings suggest that preemptive LFS can significantly decrease the incidence of hippocampus-kindled seizures and delay the progression and secondary generalization of focal seizures.

Candidiasis is the most common opportunistic fungal infection in HIV patients, and its presence is ascribed mainly to the persistence of the original infecting strain. The latter might acquire genetic variations during interaction with the host, reflecting the adaptation of the strain. Here, we report the case of a 32-year-old man complaining of asthenia, irregular hyperpyrexia, and dry cough, who was admitted to the emergency unit. Laboratory examination showed positivity for HIV. Dark violet macular lesions and ulcerated lesions with verrucous erosion were observed at the tip of the nose, whereas an ulcer without exudates was noted in the pubic region. Candida albicans was recovered from the skin by scraping these lesions. Cultures from the bronchoalveolar lavage (BAL) were negative for bacteria and opportunistic fungi but were positive for Candida albicans. The isolates from the skin and BAL were typed by PCR-RFLP and Candida albicans was identified. Analysis by microsatellite length polymorphisms, established that the pubic isolate was a genetic variant of the isolate from the nose and mouth. This suggested a microevolutionary event. Despite clinical support, the patient died of multiple organ failure.

AIM To evaluate the efficacy and safety of the modified FOLFIRI3-aflibercept as second-line therapy in patients with metastatic colorectal *** This is a retrospective multicenter cohort, evaluating the efficacy and safety of the association of aflibercept with FOLFIRI3(day 1: aflibercept 4 mg/kg, folinic acid 400 mg/m^2, irinotecan 90 mg/m^2, 5-fluorouracil infusion 2400 mg/m^2 per 46 h; day 3: irinotecan 90 mg/m^2) in patients with previously treated metastatic colorectal cancer. The primary endpoint was overall response rate(ORR). Secondary endpoints were disease control rate(DCR), progression-free survival(PFS), overall survival(OS), and *** Among 74 patients treated in four French centers, nine were excluded due to prior use of aflibercept(n = 3), more than one prior treatment line in irinotecanna?ve patients(n = 3), and inadequate liver function(n = 3). In the "irinotecan-na?ve" patients(n = 30), ORR was 43.3% and DCR was 76.7%. Median PFS and OS were 11.3 mo(95%CI: 6.1-29.0) and 17.0 mo(95%CI: 13.0-17.3), respectively. The most common(> 5%) grade 3-4 adverse events were diarrhea(37.9%), neutropenia(14.3%), stomatitis and anemia(10.4%), and hypertension(6.7%). In the "pre-exposed irinotecan" patients(n = 35), 20(57.1%) received ≥ 2 prior lines of treatment. ORR was 34.3% and DCR was 60.0%. Median PFS and OS were 5.7 mo(95%CI: 3.9-10.4) and 14.3 mo(95%CI: 12.8-19.5), *** Minimally modified FOLFIRI has improvement dramatically the FOLFIRI3-aflibercept efficacy, whatever prior use of irinotecan.
