Structural modifications of Helicobacter pylori lipopolysaccharide:An idea for how to live in peace

In this review, we discuss the findings and concepts underlying the “persistence mechanisms” of Helicobacter pylori (H. pylori), a spiral-shaped, Gram-negative rod bacterium that was discovered as a gastric pathogen by Marshall and Warren in 1984. H. pylori colonizes the gastric mucosa of nearly half of the human population. Infections appear in early childhood and, if not treated, persist for life. The presence or absence of symptoms and their severity depend on multiple bacterial components, host susceptibility and environmental factors, which allow H. pylori to switch between pathogenicity and commensalism. Many studies have shown that H. pylori components may facilitate the colonization process and the immune response of the host during the course of H. pylori infection. These H. pylori-driven interactions might result from positive or negative modulation. Among the negative immunomodulators, a prominent position is occupied by a vacuolating toxin A (VacA) and cytotoxin-associated gene A (CagA) protein. However, in light of the recent studies that are presented in this review, it is necessary to enrich this panel with H. pylori lipopolysaccharide (LPS). Together with CagA and VacA, LPS suppresses the elimination of H. pylori bacteria from the gastric mucosa by interfering with the activity of innate and adaptive immune cells, diminishing the inflammatory response, and affecting the adaptive T lymphocyte response, thus facilitating the development of chronic infections. The complex strategy of H. pylori bacteria for survival in the gastric mucosa of the host involves both structural modifications of LPS lipid A to diminish its endotoxic properties and the expression and variation of Lewis determinants, arranged in O-specific chains of H. pylori LPS. By mimicking host components, this phenomenon leaves these bacteria “invisible” to immune cells. Together, these mechanisms allow H. pylori to survive and live for many years within the

Successful antiviral therapy is associated with a decrease of serum prohepcidin in chronic hepatitis C

AIM: To assess serum concentrations of prohepcidin in chronic hepatitis C individuals and evaluate their associations with disease activity and efficacy of pegylated interferon (PEG-IFN)/ribavirin (RBV) therapy. METHODS: Prohepcidin was measured in sera of 53 chronic hepatitis C patients. Concentrations of prohepcidin and other iron metabolism markers were analyzed at 9 time points before, during and after the end of antiviral therapy. RESULTS: In hepatitis C virus (HCV) genotype 1-infected individuals, a gradual decrease of prohepcidin during antiviral therapy was observed in responders (88.8 ± 14.7 ng/mL before therapy vs 60.6 ± 0.3 ng/mL in the 48th wk, P = 0.04). In contrast, no decrease was observed in non-responders. A similar association was observed in HCV genotype 3a individuals, with a statistically significant decline in serum prohepcidin only in the responder group (99.5 ± 5.2 ng/mL at baseline vs 72.7 ± 6.1 ng/mL in the 24th wk, P = 0.01). Moreover, HCVRNA at week 12 of therapy was positively correlated with baseline (R = 0.63, P 0.005) and week 12 (R = 0.60, P = 0.01) serum prohepcidin concentrations in HCV genotype 1 infection. CONCLUSION: Successful PEG-IFN/RBV therapy results in a decline of serum prohepcidin concentration in chronic hepatitis C, which may suggest a direct effect of HCV on iron metabolism at the prohormonal level of hepcidin.

A new genus Microelectrona gen. nov. and species Microelectrona cladara sp. nov. of the extinct tribe Protodikraneurini of the leafhoppers （Cicadellidae： Typhlocybinae） from the Eocene Baltic amber is described. The piece of amber containing leafhopper inclusion is broken, unveiling some of the morphological structures, and enabling their studies using scanning electron microscopy.

The distribution of selected critical elements in the sedimentary rocks of the Carboniferous coal-bearing series within the Polish Coal Basins is *** elements such as Be,Mg,Si,P,Sc,V,Co,Y,Nb,In,Sb,La,Ce,Hf,Ta,W,Bi were analysed using inductively-coupled plasma mass spectrometry(ICP/MS).Concentrations of elements such as Sb,Bi,In and,to a slightly lesser extent,Nb,as well as Sc,show average concentrations higher than those from the upper continental *** average concentrations of elements like Hf,Mg,P,Y,La,and Ce are slightly lower than in the upper continental *** elements,such as Be,Co,Si,Ta,W and V have average concentrations that are similar,but slightly enriched or slightly depleted,relative to the upper continental *** research showed enrichment of some critical elements in the analysed samples,but not high enough that extraction would be economically *** methods,which include correlation coefficients between elements and cluster analysis,reveal a strong positive correlation between elements like Be,Bi,Nb,Sc,Ta,W and *** high,almost total,positive correlation is also noted between La and Ce.

Comprehensive Approach for Porous Materials Analysis Using a Dedicated Preprocessing Tool for Mass and Heat Transfer Modeling

The paper presents a comprehensive, newly developed software – poROSE(poROus materials examination SoftwarE) for the qualitative and quantitative assessment of porous materials and analysis methodologies developed by the authors as a solution for emerging challenges. A low porosity rock sample was analyzed and thanks to the developed and implemented methodologies in poROSE software, the main geometrical properties were calculated. A tool was also used in preprocessing part of the computational analysis to prepare a geometrical representation of the porous material. The basic functions as elimination of blind pores in the geometrical model were completed and the geometrical model was exported for CFD software. As a result, it was possible to carry out calculations of the basic properties of the analyzed porous material sample. The developed tool allows to carry out quantitative and qualitative analysis to determine the most important properties characterized porous materials. In presented tool the input data can be images from X-ray computed tomography(CT), scanning electron microscope(SEM) or focused ion beam with scanning electron microscope(FIB-SEM) in grey level. A geometric model developed in the proper format can be used as an input to modeling mass, momentum and heat transfer, as well as, in strength or thermo-strength analysis of any porous materials. In this example, thermal analysis was carried out on the skeleton of rock sample. Moreover, thermal conductivity was estimated using empirical equations.

Effects of a sex ratio gradient on female mate-copying and choosiness in Drosophila melanogaster

In many sexually reproducing species, individuals can gather information about potential mates by observing their mating success. This behavioral pattern, that we call mate-copying, was reported in the fruit fly Drosophila rnelanogaster where females choosing between 2 males of contrasting phenotypes can build a preference for males of the phenotype they previously saw being chosen by a demonstrator female. As sex ratio is known to affect mate choice, our goal was to test whether mate-copying is also affected by encountered sex ratios. Thus, we created a gradient of sex ratio during demonstrations of mate-copying experiments by changing the number of females observ- ing from a central arena 6 simultaneous demonstrations unfolding in 6 peripheral compartments of a hexagonal device. We also tested whether the sex ratio experienced by females during demon- strations affected their choosiness （male courtship duration and double courtship rate） in subse- quent mate-choice tests. Experimental male：female sex ratio during demonstrations did not affect mate-copying indices, but positively affected the proportion of both males courting the female during mate-choice tests, as well as male courtship duration, the latter potentially explaining the for- mer relationship. As expected, the sex ratio affected female choosiness positively, and Drosophila females seem to have evolved a mate-copying ability independently of sex ratio, and a capacity to adapt their choosiness to male availability. This suggests that, as in many animal species, individuals, especially females, can adapt their mate choice depending on the current sex ratio.

Comparison of SCS-CN Determination Methodologies in a Heterogeneous Catchment

The aim of this study was to assess the runoff amount from a catchment characterized by diverse land uses by using the Soil Conservation Service Curve Number(SCS-CN) method based on Curve Number(CN) defined for dominant homogeneous elementary *** calculations employed the SCS-CN method,involving the division of the catchment in two homogeneous parts and determining the runoff *** obtained results were compared with the results provided by three other CN determination methods,*** Hawkins function,the kinetics equation,and a complementary error function *** catchment is located in a mountain dominated by forest land *** CN-Precipitation(CN_(emp)-P) data pairs were analyzed using the mentioned methods,and the highest quality score was achieved from model *** results suggest that dividing a catchment into two homogeneous areas and determining their separate CN parameters,used later on to calculate the runoff by means of the presented approach,could be an alternative to the standard *** described method is relatively easy,and as it does not require an adoption of numerous parameters,and it can be employed for designing hydraulic facilities.

Random checkerboard based homogenization for estimating effective thermal conductivity of fully saturated soils

This paper proposes homogenization scheme for estimating the effective thermal conductivity of fully saturated soils. This approach is based on the random checkerboard-like microstructure. Two modeling scales and two modeling approaches are distinguished and used, i.e. microscale and mesoscale and 1-step and 2-step homogenizations, respectively. The 2-step homogenization involves sequential averaging procedure, i.e. first, at microscale, a mineralogical composition of soil skeleton is considered and averaging process results in estimation of the skeleton effective thermal conductivity, and then, at mesoscale, a random spatial packing of solid skeleton and pores via random checkerboard microstructure is modeled and leads to evaluation of the soil overall thermal conductivity. The 1-step homogenization starts directly at the mesoscale and homogenization procedure yields evaluation of the overall soil thermal conductivity. At the mesoscale, the distinct nature of soil skeleton, as composed of soil separates,is considered and random variability of soil is modeled via enriched random checkerboard-like *** approaches, i.e. 1-step and 2-step homogenizations, interrelate mineralogical composition with the soil texture characterized by the volume fractions of soil separates, i.e. sand, silt and clay. The probability density functions(PDFs) of thermal conductivity are assumed for each of the separates. The soil texture PDF of thermal conductivity is derived taking into consideration the aforementioned functions. Whenever the random checkerboard-like structure is used in averaging process, the Monte Carlo procedure is applied for estimation of homogenized thermal conductivity. Finally, the proposed methodology is tested against the laboratory data from our measurements as well as those available from literature.
