Common and small molecules as the ultimate regulatory and effector mediators of antigen-specific transplantation reactions

In spite of intensive research, the molecular basis of allograft and xenograft rejection still remains not fully understood. The acute rejection of an allograft is associated with the intragraft Th1 cytokine response, while tolerance of an allograft or xenograft rejection is accompanied by a higher production of the Th2 cytokines interleukin(IL)-4 and IL-10. Nevertheless, these cytokines are not the final regulatory and effector molecules mediating transplantation reactions. Data indicate that the functioning of common molecules with enzymatic activities, such are inducible nitric oxide synthase(i NOS), arginase, heme oxygenase-1(HO-1) or indoleamine-2,3-dioxygenase(IDO), the bioavailability of their substrates(L-arginine, tryptophan, heme) and the cytotoxic and regulatory actions of their small gaseous products(NO, CO) can be the ultimate mechanisms responsible for effector or regulatory reactions. Using models of transplantation immunity and tolerance we show that T cell receptor-mediated recognition of allogeneic or xenogeneic antigens as well as the balance between immunity/tolerance induces distinct cytokine production profiles. The ratio between Th1 and Th2 cytokines efficiently regulates the expression of genes for common enzymes, such as i NOS, arginase, HO-1 and IDO. These enzymes may compete for substrates, such as L-arginine or tryptophan, and the final product of their activity are small molecules(NO, CO) displaying effector or regulatory functions of the immune system. Thus, it is suggested that in spite of the high immunological specificity of transplatation reaction, the ultimate players in regulatory and effector functions could be small and common molecules.

Nature itself uses materials like, cellulose to provide the structure of plants, chitin as the exoskeleton of several insects and molluscs, collagen for mechanical support in connective tissues and so on. At present, the socioeconomic situation of the modern world has raised the interest in renewable materials being used in regenerative medicine. The composition of MCCh/?-TCP complex in sponge shape is derived from the junction of two or more different materials, containing organic and inorganic materials, including bioactivity and biodegradability as a characteristic. The chemical characteristics of MCCh/?-TCP complex composites showed that both of the components organic and inorganic exist in the material. All sponge preparations, with MCCh/?-TCP have a well-shaped 3-dimentional structure, a highly porous and interconnected and homogenous pore structure to ensure a biological environment conducive to cell attachment and proliferation as well as tissue growth, providing the passage of nutrient flow. These materials can be used in future for medical applications as a base for scaffolds production and as implants in regenerative medicine.

Red cabbage washing with acidic electrolysed water:effects on microbial quality and physicochemical properties

The effects of acidic electrolysed water(AEW)as‘green’technology on the microbiological and physicochemical properties of fresh-cut red cabbages were ***-cut red cabbages and artificially inoculated red cabbages with Salmonella typhimurium DT104 were washed with distilled water(DW)and different available chlorine concentrations(ACC)of AEW for different *** treatments significantly reduced the populations of native aerobic bacteria,molds,and yeasts,and artificially inoculated *** DT104 compared with the DW-treated and untreated red cabbage *** effectiveness of AEW treatments was greatly enhanced with increasing ACC and treatment *** DT104 were not detected in the washing water that were collected after the red cabbages treated by *** surface colour,pH,and total phenolic contents did not significantly change when the red cabbages were washed with DW and AEW containing 100 mg/L available chlorine for 3 *** anthocyanin contents and antioxidant activities of red cabbage were significantly reduced by 18.5 per cent for cyanidin,22.1 per cent for pelargonidin,and 11.2 per cent for 2,2-diphenyl-1-picrylhydrazyl(DPPH)radical scavenging activity,however,the impacts on the nutritional benefits of red cabbage were considered as limited and *** optimal process condition of AEW for washing red cabbage was 100 mg/L ACC for 3 *** these conditions,most of the native microflora were inactivated,and artificially inoculated *** DT104 on the red cabbage were reduced by 40.2 per cent[3.67 log CFU/g(log10 colony-forming units per gram)]and with minimal losses of nutrients and antioxidant activity,as well as no requirement of decontamination treatment on the washing water after AEW treatment.

AIM:To assess differing patterns and levels of ascitic fluid cyctokine and growth factors exist between those with a high risk and low risk of spontaneous bacterial peritonitis(SBP). METHODS: A total of 57 consecutive patients with ascites requiring a large volume paracentesis were studied. Their age, gender, specific underlying disease conditions were recorded after a review of their clinical records. Each underwent a routine assessment prior to their paracentesis consisting of a complete blood count, complete metabolic profile and prothrombin time/international normalized ratio(INR) determination. The ascitic fluid was cultured and a complete cellcount and albumin determination was obtained on the fluid. In addition, blood and ascitic fluid was assessed for the levels of interleukin interleukin(IL)-1A, IL-1B, IL-2, IL-4, IL-8, IL-10, monocyte chemotactic protein(MCP)-1, tumor necrosis factor(TNF)-α, interferon(IFN)-γ, vascular endothelial growth factor(VEGF) and epidermal growth factor(EGF) utilizing the Randox Biochip platforms(Boston, MA). A serum-ascites gradient, for each cytokine and growth factor was calculated. The results are reported as mean ± SEM between disease groups with statistical analysis consisting of the student t-test(two tailed) with a P value of 0.05 defining significance. RESULTS: No clinically important demographic or biochemical differences between the 4 groups studied were evident. In contrast, marked difference in the cytokine and growth factors levels and pattern were evident between the 4 disease groups. Individuals with alcoholic cirrhosis had the highest levels of IL-1A, IL-1B, IL-4, IFNγ. Those with malignant disease had the highest levels of IL-2. Those with hepatitis C virus(HCV) associated cirrhosis had the highest value for IL-6, IL-8, IL-10, MCP-1 and VEGF. Those with cardiac disease had the highest level of TNF-α and EGF. Plant water use efficiency(WUE) is becoming a key issue in semiarid areas, where crop production relies on the use of large volumes of water. Improving WUE is necessary for securing environmental sustainability of food production in these areas. Given that climate change predictions include increases in temperature and drought in semiarid regions,improving crop WUE is mandatory for global food production. WUE is commonly measured at the leaf level, because portable equipment for measuring leaf gas exchange rates facilitates the simultaneous measurement of photosynthesis and transpiration. However,when those measurements are compared with daily integrals or whole-plant estimates of WUE, the two sometimes do not agree. Scaling up from single-leaf to whole-plant WUE was tested in grapevines in different experiments by comparison of daily integrals of instantaneous water use efficiency [ratio between CO2assimilation(AN) and transpiration(E); AN/E] with midday AN/E measurements, showing a low correlation, being worse with increasing water stress. We sought to evaluate the importance of spatial and temporal variation in carbon and water balances at the leaf and plant levels. The leaf position(governing average light interception) in the canopy showed a marked effect on instantaneous and daily integrals of leaf WUE. Night transpiration and respiration rates were also evaluated, as well as respiration contributions to total carbon balance. Two main components were identified as filling the gap between leaf and whole plant WUE: the large effect of leaf position on daily carbon gain and water loss and the large flux of carbon losses by dark respiration. These results show that WUE evaluation among genotypes or treatments needs to be revised.

Chronic kidney disease(CKD) typically evolves over many years, with a long latent period when the disease is clinically silent and therefore diagnosis, evaluation and treatment is based mainly on biomarkers that assess kidney function. Glomerular filtration rate(GFR) remains the ideal marker of kidney function. Unfortunately measuring GFR is time consuming and therefore GFR is usually estimated from equations that take into account endogenous filtration markers like serum creatinine(SCr) and cystatin C(Cys C). Other biomarkers such as albuminuria may precede kidney function decline and have demonstrated to have strong associationswith disease progression and outcomes. New potential biomarkers have arisen with the promise of detecting kidney damage prior to the currently used markers. The aim of this review is to discuss the utility of the GFR estimating equations and biomarkers in CKD and the different clinical settings where these should be applied. The CKD-Epidemiology Collaboration equation performs better than the modification of diet in renal disease equation, especially at GFR above 60 m L/min per 1.73 m2. Equations combining Cys C and SCr perform better than the equations using either Cys C or SCr alone and are recommended in situations where CKD needs to be confirmed. Combining creatinine, Cys C and urine albumin to creatinine ratio improves risk stratification for kidney disease progression and mortality. Kidney injury molecule and neutrophil gelatinase-associated lipocalin are considered reasonable biomarkers in urine and plasma to determine severity and prognosis of CKD.

Seasonal variations play an essential role in the metabolism, behavior and activity of the laboratory animals. This study was aimed to examine whether mitochondrial function can be influenced by the seasonal changes and how large is the impact of these fluctuations on experiments with using animal models and further results interpretation. Liver mitochondria were isolated from male Wistar rats and exposed to calcium ions, PAMAM dendrimers G2.5 or their combination: (Ca2+) and dendrimer. The scientific hypothesis assumed that dendrimer G2.5 is able to limit the detrimental effect of Ca2+ ions on mitochondria function, possibly through affecting the following parameters: calcium transport, mitochondrial potential and membrane fluidity. The activity of mitochondria was monitored using fluorescent labels. The changes in calcium transport were detected using Calcium Green 5-N, the mitochondrial membrane potential and membrane fluidity were elucidated using JC-1 and DPH, respectively. The experiments were carried out during the autumn (October/November) or during the spring (May/June). The obtained data emphasize the effect of seasonal differences on liver mitochondria originating from laboratory animals and outline the importance of planning the experiments during the same seasonal period in order to receive objective and reliable results in the future. Finally, it was revealed the neutral effect of G2.5 dendrimer on mitochondria and its inability to protect mitochondria against overload of calcium ions regardless of seasonality. It was also evidenced that liver mitochondria isolated from autumn-derived animals were more sensitive to calcium and/or dendrimer exposure in comparison with mitochondria isolated from animals investigated during the spring.
