Current Injection Provokes Rapid Expansion of the Guard Cell Cytosolic Volume and Triggers Ca^2＋ Signals

High-resolution microscopy opens the door for detailed single-cell studies with fluorescent reporter dyes and proteins. We used a confocal spinning disc microscope to monitor fluorescent dyes and the fluorescent protein Venus in tobacco and Arabidopsis guard cells. Multi-barreled microelectrodes were used to inject dyes and apply voltage pulses, which provoke transient rises in the cytosolic Ca^2＋ level. Voltage pulses also caused changes in the distribution of Lucifer Yellow and Venus, which pointed to a reversible increase of guard cell cytosolic volume. The dynamic cytosolic volume changes turned out to be provoked by current injection of ions. A reduction of the clamp current, by blocking K^＋ uptake channels with Cs^＋, strongly suppressed the cytosolic volume changes. Cs^＋ not only inhibited the expansion of the cytosol, but also inhibited hyperpolarization-induced elevations of the cytosolic Ca^2＋ concentration. A complete loss of voltage-induced Ca^2＋ signals occurred when Ca^2＋-permeable plasma membrane channels were simultaneously blocked with La^3＋. This shows that two mechanisms cause hyperpolarization-induced elevation of the cytosolic Ca^2＋-concentration：（i） activation of voltage-dependent Ca^2＋-permeable channels, （ii） osmotically induced expansion of the cytosol, which leads to a release of Ca^2＋ from intracellular stores.

Background： Rhythmical massage therapy（RMT） is a massage technique used in anthroposophic ***： The authors aimed to investigate the physiological action of RMT on the cardiovascular system by analysing heart rate variability（HRV）.Design, setting, participants and intervention： This study was a randomised, controlled and single-blinded trial, involving 44 healthy women（mean age：（26.20 ± 4.71） years）. The subjects were randomised to one of three arms： RMT with aromatic oil（RA）, RMT without aromatic oil（RM） or standardised sham massage（SM）. In the study the subjects were exposed to a standardised stress situation followed by one of the study techniques and Holter electrocardiograms（ECGs） were recorded for 24 *** outcome measures： HRV parameters were calculated from linear（time and frequency domain） and nonlinear dynamics（symbolic dynamics, Poincare plot analysis） of the 24-h Holter ECG ***： Short-and long-term effects of massage on autonomic regulation differed significantly among the three groups. Immediately after an RMT session, stimulation of HRV was found in the groups RA and RM. The use of an aromatic oil produced greater short-term measurable changes in HRV compared with rhythmic massage alone, but after 24 h the effect was no longer distinguishable from the RM *** lowest stimulation of HRV parameters was measured in the SM ***： RMT causes specific and marked stimulation of the autonomic nervous system. Use of a medicinal aromatic oil had only a temporary effect on HRV, indicating that the RM causes the most relevant long-term effect. The effect is relatively specific, as the physiological effects seen in the group of subjects who received only SM were considerably less pronounced.

AIM： To investigate the mechanisms underlying the reduction in gastric blood flow induced by a luminal water extract of Hellcobacter pylori （HPE）. METHODS： The stomachs of isoflurane-anesthetized mice were exteriorized, and the mucosal surface exposed. Blood flow was measured with the laserDoppler technique, and systemic arterial blood pressure monitored. C57BL/6 mice were exposed to water extract produced from Hpylori strain 88-23. To investigate the role of a nerveor iNOS-mediated pathway, we used intraluminal lidocaine and iNOS-/- mice. Blood flow response to the endogenous nitric oxide synthase inhibitor asymmetric dimethyl arginine （ADMA） was also assessed. RESULTS： In wild-type mice, HPE decreased mucosal blood flow by approximately 30%. This reduction was abolished in iNOS-deficient mice, and by pre-treatment with lidocaine. Luminally applied ADMA resulted in reduction in blood flow similar to that observed in wildtype mice exposed to HPE. CONCLUSION： A H py/ori water extract reduces gastric mucosal blood flow acutely through iNOS- and nerve-mediated pathways.

The area is a part of the Egyptian Eastern Desert in the northwestward to the Gulf of Suez. It covers an area of about 542 square kilometers. Wadi Bada’a is devoid of vegetation, because of the arid climate and water scarcity. However, the present study concerns the flash flood and its impact on the industrial zone and connected road at wadi Bada’a. In this work, the bivariate statistical method using frequency ratio was used to evaluate the areas of potential risk. Geographic Information System package (GIS) was used to analyze and calculate different data sets. The different data source has been used in the research to produce a flood hazard susceptibility map of the area, including the geologic maps, Landsat-8 imagery, land use, and soil type associated with field investigation and data collection. Spatial database with elements at risk, related features and attributes at wadi Bada’a, were constructed. Training data were created randomly in the study area to create an inventory map with testing data. The inventory location of 95 location points has been created. The inventory datasets were divided into 75% of training datasets and 25% testing data. The independent flood-related factors were evaluated by analyzing each independently and assessing their impact on flooding with inventory datasets. The flood susceptibility maps were constructed-using training and testing datasets have been used to evaluate using the success rate method. The results of the accuracy assessment showed a success rate of 76.6% of Area Under Curve. Therefore, the main road in the study area almost at high risk in many parts because of flash flood, additionally the industrial activities located in the moderate risk zone.

Few studies have attempted to investigate the effect of body composition and leptin with vitamin D in *** To investigate the association of serum leptin with body composition, lipids and25-hydroxyvitamin D(25 OHD) in *** This cross-sectional study was conducted on 167 apparently healthy *** were recorded, bioelectrical impedance analysis was performed and clinical history noted. Serum leptin was measured using DIA source kit on ELISA and total 25 OHD was measured on ADVIA-Centaur; Siemens. Total cholesterol and high density lipoprotein cholesterol were quantified using Enzymatic Endpoint Method and Cholesterol Oxidase-Phenol Aminophenazone method respectively. Biochemical analysis was done in the Departments of Pathology and Laboratory Medicine and Biological and Biomedical Sciences, Aga Khan University Hospital Karachi *** Median age of the group(n = 167) was 20 years(IQR 27-20); 55.7% were ***(89.2%, n = 149) of the study group was 25 OHD deficient, 6%(n = 10)had insufficient serum 25 OHD levels and 4.8%(n = 8) had sufficient D ***, had higher median leptin levels [2.71(IQR 4.76-1.66 ng/mL)] compared to their counterparts [1.3(3.60-0.54 ng/mL), P < 0.01].
