Mucuna pruriens (velvet bean) represents an interesting source of protein poorly studied. The effect of dietary inclusion of meal and protein isolate of Mucuna seeds on biochemical and oxidative stress parameter of broilers (135-one-day Cobb500 chickens) was investigated. Three isonitrogenous diets were formulated from soya bean meal (Control group: RTS), Mucuna meal (coded RFM) and Mucuna protein isolate (coded RIM). Each of the dietary treatments was triplicated with 15 birds per replicate in a completely randomized design. The birds were offered feed and water ad libitum. The results revealed significant (p 0.05) effect of N source on the organ total proteins with treatment RFM and RIM exhibiting lower but comparable levels in the Liver (2.01 and 1.98 g/dL), Heart (1.95 and 1.89 g/dL) and Kidney (1.92 and 1.91 g/dL). Synthetic control of topology connectivity is the crowning achievement for covalent organic framework(COF)*** a large number of one-,two-,and three-dimensional(1D,2D,and 3D)COFs have been reported,their topology structure constructions are restricted to the use of symmetric monomers with the purpose of increasing the crystallinity and/or ***,three imine-linked COFs with different topology nets(namely,sql-b and sql-c)were constructed by symmetric variation of monomers via the condensation of a tetra-amine monomer with a D2h-symmetry and lower C_(2v)-symmetric dialdehyde monomers,bearing a phenolic hydroxyl group at different *** results indicated that a reasonable introduction of the phenolic hydroxyl group could effectively tune the topological structure of COFs at the molecular level during the crystallization ***,the remarkable difference in the dye uptake ability between these COF materials indicated that the fabricated specific pore geometries,as well as different steric hindrance and H-bonding interactions,played a pivotal role in accessing molecules in the ***,this work might boost the explorations of COF materials with expected topologies and pore geometries from conventional monomers through bottom-up synthesis methodology by molecular designing and engineering.

In Central African Republic, edible caterpillars of Imbrasia gender are income of rural populations, present an opportunity to improve food security and belonging to the category of Non-wood forest products. The goal of this study is to document the ecology of caterpillars of genre Imbrasia in relation to its host plants in Boukoko. The questionnaire was addressed to the actors of the collection and consumption of caterpillars in this Commune, to their local knowledge concerning this food resource as well as the plants of their locality on which caterpillars are collected. The survey took place in September to October 2022 and as a general rule, the 120 respondents were elderly people whose age range varied from 35 to 70 years old. Plant samples are collected in their natural environment. Based on local names;herbaria are mounted, plants photographed and samples collected. The types of edible caterpillars have been inventoried in the field in collaboration with the local populations who consume them. Based on local names, the caterpillars are captured manually from morphological traits and then stored in bottles containing 70% ethanol. The specimens were identified. The data was entered into the Excel spreadsheet which also made it possible to produce histograms with the frequencies of the plant species most valued by the different caterpillars. A Principal Component Analysis (PCA) was then performed using R software to highlight the correlation between caterpillars and host plants. The main economic importance trees hosting edible caterpillars of Imbrasia gender in Boukoko region are: Entandrophragma cylindricum, Pentaclethra macrophylla, Maesopsis eminii, Albizia ferruginea, Macaranga spinosa, Amphimas ferrugineus, Pentaclethra macrophylla, Racosperma auriculiforme, Eribroma oblonga. PCR-HRM for Genomic Surveillance of SARS-CoV-2: A Variant Detection Tool in Côte d’Ivoire, West Africa

The rise of new viruses, like SARS-CoV-2 causing the COVID-19 outbreak, along with the return of antibiotic resistance in harmful bacteria, demands a swift and efficient reaction to safeguard the health and welfare of the global population. It is crucial to have effective measures for prevention, intervention, and monitoring in place to address these evolving and recurring risks, ensuring public health and international security. In countries with limited resources, utilizing recombinant mutation plasmid technology in conjunction with PCR-HRM could help differentiate the existence of novel variants. cDNA synthesis was carried out on 8 nasopharyngeal samples following viral RNA extraction. The P1 segment of the SARS-CoV-2 Spike S protein was amplified via conventional PCR. Subsequently, PCR products were ligated with the pGEM-T Easy vector to generate eight recombinant SARS-CoV-2 plasmids. Clones containing mutations were sequenced using Sanger sequencing and analyzed through PCR-HRM. The P1 segment of the S gene from SARS-CoV-2 was successfully amplified, resulting in 8 recombinant plasmids generated from the 231 bp fragment. PCR-HRM analysis of these recombinant plasmids differentiated three variations within the SARS-CoV-2 plasmid population, each displaying distinct melting temperatures. Sanger sequencing identified mutations A112C, G113T, A114G, G214T, and G216C on the P1 segment, validating the PCR-HRM findings of the variations. These mutations led to the detection of L452R or L452M and F486V protein mutations within the protein sequence of the Omicron variant of SARS-CoV-2. In summary, PCR-HRM is a vital and affordable tool for distinguishing SARS-CoV-2 variants utilizing recombinant plasmids as controls.

The improvement of microbial characterization has increased the comprehension of microbial population and their ability in the microbiological metal dissolution. Bioleaching processes have been expanded to use microorganisms for the recovery of metals from ores and wastes. This study introduces Palca mine tailings pond in Peru which turned into acid mine drainage (AMD). AMD is a source of microbial communities whose microorganisms may support the aqueous extractive metallurgy for metal recovery. Four samples of AMD were collected from different locations and the elemental characterization showed concentrations of metals, such as Cu, Zn, Al, Mn, and Fe. The pH of the samples showed variation between 1.9 - 6.8. Twenty-one microorganisms were isolated and selected according the cell morphology. 16s rRNA gene sequences identified five species of which three belong to the bacterial kingdom and two to the Fungi kingdom. Two of the bacterial species were ferrous ion oxidizing bacteria, identified as Acidithiobacillus ferriphilus and Acidithiobacillus ferridurans;and the other one a ferric ion reducing bacteria identified as Acidiphilium acidophilum. The fungi species identified were Rhodotorula sinensis and Exophiala xenobiotica, a filamentous fungus isolated for the first time from an AMD.

The emergence of antiretroviral resistance mutations represents a major threat to the achievement of national and global goals for the elimination of HIV-1 infection. The global strategy in 2019 in Cte d'Ivoire is a new national policy for the management of people living with HIV with the administration of dolutegravir (DTG)-based fixed-dose combination. The aim of our study was to evaluate HIV-1 resistance to antiretrovirals (ARVs) in infected adult subjects in Cte d’Ivoire in the context of a systematic switch to a DTG-based combination. Between February 2022 and October 2023, a cross-sectional survey with random sampling was conducted in 06 services caring for people living with HIV. A total of 139 participants were included in the study. Adults with a viral load ≥ 1000 copies/mL were tested for HIV-1 ARV resistance mutations. Molecular analyses were performed using protocol of ANRS-MIE (National Agency for Research on AIDS and emerging infectious diseases). The interpretation is performed by HIVGRAD (https://***/cms/grade/). The frequencies of HIV-1 resistance to non-nucleotide reverse transcriptase inhibitors (NNRTIs), nucleotide reverse transcriptase inhibitors (NRTIs), integrase inhibitors (IINTs) and protease inhibitors (PIs) were 82%, 73%, 19% and 11% respectively. The main mutations observed in the different classes were K103N (45%), M184V (64%), E157Q (19%) and L10V/M46I/A71V/I54V (6%) respectively. This study reveals the emergence of resistance to DTG-based fixed-dose combinations, favored by high rates of resistance to NRTIs and NNRTIs. This finding underlines the need for enhanced viral load monitoring and HIV-1 genotyping tests to guide the choice of NRTIs for combination therapy. In addition, monitoring for mutations to second-generation NRTIs is essential, given the scale-up of DTG-based regimens currently underway in Cte d’Ivoire.

Introduction: The epidemiology of both hepatitis B virus (HBV) and hepatitis C virus (HCV) infections among drug users (DUs) is little known in West Africa. The study aimed to assess the prevalence of hepatitis B and C viruses among drug users in Burkina Faso. Methodology: This was a cross-sectional biological and behavioral survey conducted between June and August 2022, among drug users in Ouagadougou and Bobo Dioulasso, the two main cities of Burkina Faso. A respondent-driven sampling (RDS) was used to recruit drug users. Hepatitis B surface antigen was determined using lateral flow rapid test kits and antibodies to hepatitis C virus in serum determined using an Enzyme-Linked Immunosorbent Assay. Data were entered and analyzed using Stata 17 software. Weighted binary logistic regression was used to identify the associated factors of hepatitis B and C infections and a p-value Results: A total of 323 drug users were recruited with 97.5% males. The mean age was 32.7 years old. The inhaled or smoked mode was the most used by drug users. The adjusted hepatitis B and hepatitis C prevalence among study participants were 11.1% and 2.3% respectively. The marital status (p = 0.001), and the nationality (p = 0.011) were significantly associated with hepatitis B infection. The type of drug used was not significantly associated with hepatitis B infection or hepatitis C infection. Conclusion: The prevalence of HBsAg and anti-HCV antibodies among DUs are comparable to those reported in the general population in Burkina Faso. This result suggests that the main routes of contamination by HBV and HCV among DUs are similar to those in the population, and could be explained by the low use of the injectable route by DUs in Burkina Faso.

Molecular aspects of intestinal calcium absorption

Intestinal Ca2+ absorption is a crucial physiological process for maintaining bone mineralization and Ca2+ homeostasis. It occurs through the transcellular and paracellular pathways. The first route comprises 3steps: the entrance of Ca2+ across the brush border membranes(BBM) of enterocytes through epithelial Ca2+ channels TRPV6, TRPV5, and Cav1.3; Ca2+ movement from the BBM to the basolateral membranes by binding proteins with high Ca2+ affinity(such as CB9k); and Ca2+ extrusion into the blood. Plasma membrane Ca2+ ATPase(PMCA1b) and sodium calcium exchanger(NCX1) are mainly involved in the exit of Ca2+ from enterocytes. A novel molecule, the 4.1R protein, seems to be a partner of PMCA1 b, since both molecules colocalize and interact. The paracellular pathway consists of Ca2+ transport through transmembrane proteins of tight junction structures, such as claudins 2, 12, and 15. There is evidence of crosstalk between the transcellular and paracellular pathways in intestinal Ca2+ transport. When intestinal oxidative stress is triggered, there is a decrease in the expression of several molecules of both pathways that inhibit intestinal Ca2+ absorption. Normalization of redox status in the intestine with drugs such as quercetin, ursodeoxycholic acid, or melatonin return intestinal Ca2+ transport to control values. Calcitriol [1,25(OH)2D3] is the major controlling hormone of intestinal Ca2+ transport. It increases the gene and protein expression of most of the molecules involved in both pathways. PTH, thyroid hormones, estrogens, p ro l a c t i n, g ro w t h h o r m o n e, a n d g l u c o c o r t i c o i d s apparently also regulate Ca2+ transport by direct action, indirect mechanism mediated by the increase of renal 1,25(OH)2D3 production, or both. Different physiological conditions, such as growth, pregnancy, lactation, and aging, adjust intestinal Ca2+ absorption according to Ca2+ demands. Starch Digestibility and Functional Properties of Rice Starch Subjected to Gamma Radiation

This study investigated the effect of gamma radiation on the digestibility and functional properties of rice *** cultivars IRGA417 and IAC202 were used for isolation of starch by the alkaline *** samples were irradiated with 1,2 and 5 k Gy doses of ^(60)Co at a rate of 0.4 k Gy/h.A control sample,which was not irradiated,was used for *** and control starches were characterized by in vitro starch digestibility,total dietary fiber,color,water absorption index,water solubility index,syneresis,swelling factor,amylose leaching,pasting properties and gel *** changed starch digestibility differently in either *** radiation doses promoted increase in the color parameter b*(yellow),elevation in the capacity to absorb water,and solubility in water as well as the amylose leached from granules for both *** properties showed a decrease that was proportional to the dose applied,caused by the depolymerization of starch *** firmness of the starch from IAC202 was inversely proportional to the radiation dose applied,whereas for IRGA417,there was a reduction at 5 k Gy *** starches can be modified by irradiation to exhibit different functional characteristics and they can be used by the food industries in products such as soups,desserts,flans,puddings and others.
