Carotenoids and chlorophylls are among the most widely distributed pigments in nature that play essential roles in the photosynthetic apparatus and confer diverse colours in plants.Among all vegetables,cauliflower(Brassica oleracea L.ssp.***.botrytis)is rich in phytochemicals and is an important crop grown all over the world.This study investigates carotenoid and chlorophyll concentrations in differently pigmented cultivars and elucidates the role of transcriptional regulation of carotenoid accumulation including lutein andβ-carotene.Here,we characterised changes in pigments by UHPLC-DAD-ToF-MS and changes in transcript levels of carotenoid metabolic genes by qRT-PCR in florets and leaves of orange(‘Jaffa'and‘Sunset'),purple(‘Di Sicilia Violetto'and‘Graffiti'),green(‘Trevi')and white(‘Clapton')cultivars.Transcript levels of all carotenoid metabolic genes showed different transcript level patterns in the leaves and florets.Compared to the other cultivars,the orange cultivars had the highest levels ofβ-carotene in the florets and lutein in the leaves resulting in changes lutein/β-carotene ratios.In the green cultivar,higher transcript levels were also found,especially for phytoene synthase and phytoene desaturase genes of the core biosynthesis pathway.However,no increased carotenoid concentrations were observed,possibly due to a higher carotenoid turnover induced by the carotenoid cleavage dioxygenase 4 in the green cultivar.In the white(‘Clapton')and purple(‘Di Sicilia Violetto'and‘Graffiti')cultivars the phytoene desaturase transcript levels as well as carotenoid concentrations were low.Chlorophyll concentrations changed in trend comparable to the carotenoid concentrations and were only significantly lower in the leaves of the orange cultivar‘Jaffa'.Also,the chlorophyll a/b ratio changed in‘Jaffa'.In florets the highest chlorophylls concentrations were observed for the green cultivar(‘Trevi')and the purple cultivar(‘Di Sicilia Violetto').Taken together,the study demonstrates the complex source-sink relationship o

Radiopharmaceutical therapy(RPT)has shown great promise in the treatment of various cancers.However,the current approach is a"one size fits all"model,which results in varied patient responses,and there is a need for personalized RPT(Fig.S1 online).This personalized approach requires a better understanding of the physics,biology,radiobiology,pharmacology,immunology,chemistry,and imaging involved in RPT[1].Multidisciplinary research using experimental,numerical,and simulation platforms would be key to accelerating progress in this area.

A Deep Reinforcement Learning-Based Technique for Optimal Power Allocation in Multiple Access Communications

Formany years,researchers have explored power allocation(PA)algorithms driven bymodels in wireless networks where multiple-user communications with interference are present.Nowadays,data-driven machine learning methods have become quite popular in analyzing wireless communication systems,which among them deep reinforcement learning(DRL)has a significant role in solving optimization issues under certain constraints.To this purpose,in this paper,we investigate the PA problem in a k-user multiple access channels(MAC),where k transmitters(e.g.,mobile users)aim to send an independent message to a common receiver(e.g.,base station)through wireless channels.To this end,we first train the deep Q network(DQN)with a deep Q learning(DQL)algorithm over the simulation environment,utilizing offline learning.Then,the DQN will be used with the real data in the online training method for the PA issue by maximizing the sumrate subjected to the source power.Finally,the simulation results indicate that our proposedDQNmethod provides better performance in terms of the sumrate compared with the available DQL training approaches such as fractional programming(FP)and weighted minimum mean squared error(WMMSE).Additionally,by considering different user densities,we show that our proposed DQN outperforms benchmark algorithms,thereby,a good generalization ability is verified over wireless multi-user communication systems.

对比研究使用未热解粉末制备的厚热障涂层(UPTTBC)和传统TBC、厚TBC(TTBC)及密集垂直裂纹(DVC)TTBC 4种涂层的隔热能力和黏接强度。采用降温法测试涂层的隔热能力;根据ASTM C633标准,采用拉拔试验检测涂层的黏接强度。结果显示,UPTTBC和DVC TTBC的黏接强度分别比TTBC高35%和25%。这可能是由于UPTTBC结构中存在亚微米尺度的未熔化区域,起到了阻止裂纹扩展的作用。此外,隔热测试的结果表明,DVCTTBC涂层的隔热能力最差,为0.28℃/μm,而UPTTBC涂层的隔热能力最强,为0.40℃/μm。由于UPTTBCs显微组织中存在亚微米区域,使涂层展现出优越的隔热能力。相反,DVCTTBC因其致密的结构,具有最低的隔热能力。

The effect of celastrol in combination with 5-fluorouracil on proliferation and apoptosis of gastric cancer cell lines

Background:Despite the availability of chemotherapy drugs such as 5-ﬂuorouracil(5-FU),the treatment of some cancers such as gastric cancer remains challenging due to drug resistance and side effects.This study aimed to investigate the effect of celastrol in combination with the chemotherapy drug 5-FU on proliferation and induction of apoptosis in human gastric cancer cell lines(AGS and EPG85-257).Materials and Methods:In this in vitro study,AGS and EPG85-257 cells were treated with different concentrations of celastrol,5-FU,and their combination.Cell proliferation was assessed using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide(MTT)assay.The synergistic effect of 5-FU and celastrol was studied using Compusyn software.The DNA content at different phases of the cell cycle and apoptosis rate was measured usingﬂow cytometry.Results:Co-treatment with low concentrations(10%inhibitory concentration(IC10))of celastrol and 5-FU signiﬁcantly reduced IC50(p<0.05)so that 48 h after treatment,IC50 was calculated at 3.77 and 6.9μM for celastrol,20.7 and 11.6μM for 5-FU,and 5.03 and 4.57μM for their combination for AGS and EPG85-257 cells,respectively.The mean percentage of apoptosis for AGS cells treated with celastrol,5-FU,and their combination was obtained 23.9,41.2,and 61.9,and for EPG85-257 cells 5.65,46.9,and 55.7,respectively.In addition,the 5-FU and celastrol-5-FU combination induced cell cycle arrest in the synthesis phase.Conclusions:Although celastrol could decrease the concentration of 5-ﬂuorouracil that sufﬁced to suppress gastric cancer cells,additional studies are required to arrive at conclusive evidence on the anticancer effects of celastrol.

植物根茎过渡区(root-stem transition zone,RSTZ)将根和茎相互连接,其发育形态决定了大豆的地上部株型和抗倒伏潜力。本研究通过EMS诱变获得一个RSTZ弯曲或旋转的大豆突变体M_(rstz),其形态特征能够稳定遗传,是探究大豆茎秆发育规律的特异材料。将该突变体和栽培大豆中黄13杂交构建重组自交系群体,对群体中直立和弯曲型后代的RSTZ进行解剖结构比较,发现弯曲型株系比直立型株系的维管形成层较宽、次生木质部细胞层数较多、细胞形状不规则,表明维管组织分化可能是导致RSTZ形态发生差异的重要因素之一。进一步对化学组分测定发现,木质素和粗纤维含量越高越不易弯曲。选取RIL群体中弯曲型和直立型2种极端株系进行BSA-Seq,采用SNP-index和InDel-index关联分析方法鉴定到调控RSTZ形态的关联区域Chr19:43030943~45849854,该区间共含有319个基因。结合生物信息学分析、基因注释信息和表达丰度分析筛选到7个候选基因,分别为Glyma.19G170200、Glyma.19G201500、Glyma.19G187800、Glyma.19G178200、Glyma.19G197000、Glyma.19G179100、Glyma.19G196900。其中,Glyma.19G187800、Glyma.19G178200和Glyma.19G196900在大豆驯化中潜在影响RSTZ形态建成。本研究为解析大豆RSTZ组织形成及其遗传基础提供了材料基础,并为挖掘调控大豆茎秆发育基因提供新的见解。

Tribological behavior of polydopamine/polytetrafluoroethylene coating on laser textured stainless steel with Hilbert curves

Shallow Hilbert curve patterns with easily programmable texture density were selected for laser texturing of stainless steel substrates.Two different texture path segment lengths(12 and 24 μm)and four different laser power percentages(5%,10%,15%,and 20%)were investigated.The textured and smooth substrates were coated with thin polydopamine/polytetrafluoroethylene(PDA/PTFE)coatings for tribological property assessment.The effects of texture density(texture area coverage)and laser power on the durability and friction of the coated surfaces were studied.Laser texturing the substrates improved the coating durability up to 25 times,reduced the friction coefficient,and prevented coating global delamination.The textures fabricated with a laser power of 15%and a texture path segment length of 12 μm yielded the best coating durability.The textures provided the interlocking for the PTFE coating and thus prevented its global delamination.Furthermore,the PTFE inside the texture grooves replenished the solid lubricant worn away in the wear track and prolonged the coating wear life.

Street network patterns for mitigating urban heat islands in arid climates

This study explores the impact of street pattern measurements on urban heat islands(UHI)in the arid climate of Mashhad,Iran.The Landsat-8 top-of-the-atmosphere(TOA)brightness images from 2013 to 2021,average values of normalized difference vegetation index(NDvI)and land surface temperature(LST)were calculated.Street pattern measurements,including closeness-centrality,straightness,and street orientation,were employed to analyse the patterns in each district.The results indicated that districts with higher straightness and lower closeness-centrality exhibit,cooler surface temperatures.Strong correlations were observed between LST and NDVl,straightness,and local closeness-centrality.The research highlighted the importance of considering street network measurements in long-term urban planning and design to mitigate the UHI effect in arid regions.A moderate grid street pattern with a reasonable distribution of green spaces throughout the region is suggested to reduce surface temperatures sustainably.Street pattern indexes,such as straightness and local closeness-centrality,are identified as significant factors in urban design to mitigate UHl.These findings have implications for urban planners,who can use this information to create street network patterns with lower UHI effects by reducing local closeness-centrality and increasing straightness.

Experimental investigation of shock-buffet criteria on a pitching airfoil

An experimental investigation of the shock-buffet phenomenon subject to unsteady pitching supercritical airfoil around its quarter chord has been conducted in a transonic wind tunnel.The model was equipped with pressure taps connected to the fast response pressuretransducers.Measurements were conducted at different free-stream Mach number from 0.61 to0.76.The principle goal of this investigation was to experimentally discuss the shock-buffet criterion over a SC(2)-0410 supercritical pitching related to the hysteresis loops of total drag and trailing edge pressure,the behaviour of the shock wave foot location,the pressure distribution over the upper surface,and by implementing the wavelet analysis of the normal force.To ensure capturing the buffet phenomenon by utilizing these criteria,a pressure port has been drilled exactly at the trailing edge of the airfoil where its output was used to detect the buffet phenomenon for different conditions.Visual representation of the flow using the shadow graph flow visualization technique for different test cases is further used to illustrate the unsteady shock wave motion.A comparative analysis of experimental measurements shows that the conducted criteria confirm each other when the buffet phenomenon occurs at the position of the oscillating cycle.
