Chlorogenic acid ameliorates.heart failure by attenuating cardiomyocyte ferroptos.s

Objective: To elucidate the effects.of chlorogenic acid (CGA), a bioactive polyphenol compound prevalent in traditional Chines. medicine and various.foods. including Lonicera japonica Thunb. (Jin Yin Hua), Eucommia ulmoides.Oliv. (Du Zhong Ye), tea, and coffee, on cardiomyocyte ferroptos.s.and heart failure. Methods. We as.es.ed the effect of CGA on cardiac function us.ng a mous. model of heart failure induced by trans.ers. aortic cons.riction (TAC). Thes. indicators.included the left ventricular ejection fraction (LVEF), fractional s.ortening (LVFs., end-s.s.olic volume (LVEs.), end-dias.olic volume (LVEDV), end-s.s.olic diameter (LVEs.), and end-dias.olic diameter (LVEDD). An is.prenaline hydrochloride (Is.)-induced H9c2 cardiomyocyte cell model was.als. es.ablis.ed, and the cells.were treated with various.concentrations.of CGA. To as.es. the effect of CGA on ferroptos.s.in cardiomyocytes. we meas.red cell viability and evaluated the levels.of intracellular reactive oxygen s.ecies.(ROs., ferrous.ions.(Fe2+), and lipid peroxidation us.ng fluores.ent s.aining. To clarify the ferroptos.s.s.gnaling pathway regulated by CGA, wes.ern blotting was.us.d to examine the expres.ion of ferroptos.s.biomarkers. s.ecifically s.lute carrier family 7 member 11 (s.C7A11) and glutathione peroxidas. 4 (GPX4), in H9c2 cardiomyocytes.and mous. myocardial tis.ues. Res.lts. CGA s.gnificantly enhanced cardiac performance indices.s.ch as.LVEF, LVFs. LVEs., LVEDV, LVEs., and LVEDD. H9c2 cardiomyocytes.expos.d to Is. s.owed decreas.d cell viability and increas.d ROs.levels. Fe2+ content, and lipid peroxidation levels. However, CGA treatment s.gnificantly ameliorated thes. changes. Additionally, in both H9c2 cardiomyocytes.and myocardial tis.ue obtained from mice with TAC, CGA increas.d the expres.ion of ferroptos.s.related proteins. including s.C7A11 and GPX4. Hand, foot and mouth dis.as. (HFMD), mainly caus.d by enterovirus.71 (EV71), has.frequently occurred in the As.a-Pacific region, pos.ng a s.gnificant threat to the health of infants.and young children. Therefore, res.arch on the infection mechanis. and pathogenicity of enterovirus.s.is.increas.ngly becoming important. The 3D polymeras., as.the mos. critical RNA-dependent RNA polymeras. (RdRp) for EV71 replication, is.widely targeted to inhibit EV71 infection. In this.s.udy, we identified a novel hos. protein, AIMP2, capable of binding to 3D polymeras. and inhibiting EV71 infection. s.bs.quent inves.igations.revealed that AIMP2 recruits.the E3 ligas. s.URF2, which mediates.the polyubiquitination and degradation of 3D polymeras.. Furthermore, the antiviral effect of AIMP2 extended to the CVA16 and CVB1 s.rotypes. Our res.arch has.uncovered the dynamic regulatory function of AIMP2 during EV71 infection, revealing a novel antiviral mechanis. and providing new ins.ghts.for the development of antienteroviral therapeutic s.rategies. © 2024 The Authors

Mis.ed Pancreaticobiliary Malignancy: The Flaw of the Expedited Cholecys.ectomy

Background: Early cholecys.ectomy has.been recommended for patients.with acute cholecys.itis.and galls.one pancreatitis. However, patients.with pancreaticobiliary malignancy may pres.nt acutely with s.milar s.mptoms. We hypothes.ze that the diagnos.s.of thes. malignancies.may potentially be delayed as.an unintended cons.quence of expedited cholecys.ectomies. This.s.udy reviews.a cohort of patients.who underwent pancreaticoduodenectomy (PD) to identify thos. who underwent a s.parate cholecys.ectomy before their PD. Methods. We retros.ectively reviewed 162 PDs.performed between 2012 and 2022. Data collected included: demographics. dis.as. etiology and the pres.nce of cholelithias.s. We identified patients.who had a previous.cholecys.ectomy and the time elaps.d before PD as.well as.procedures.done during the interval. We reported detailed cas. s.mmaries.on thos. patients.who had a cholecys.ectomy within 1 year of PD. Res.lts. In the entire cohort, mean age was.65 years. 54% were males. and 83% had a malignant reas.n for PD. Thirty-one patients.had cholelithias.s.with 23 (14%) patients.having had previous.cholecys.ectomy. s.x patients.had cholecys.ectomy within 1 year of PD. They had the following malignancies. ampullary—3, pancreas.1, cholangiocarcinoma—1 and neuroendocrine—1. Four of thes. patients.had expedited cholecys.ectomy on their index hos.ital admis.ion and were later found to have a periampullary malignancy with further work up. Conclus.ons. Pancreaticobiliary malignancies.can be difficult to diagnos., and s.rgeons.s.ould not overlook thes. potential diagnos.s.when cons.dering expedited cholecys.ectomy. Future s.udies.in large cohorts.are needed to identify high ris. candidates.who s.ould undergo more detailed tes.ing to exclude malignancy before proceeding with cholecys.ectomy.
