Celastrol activates caspase-3/GSDME-dependent pyroptosis in tumor cells by inducing endoplasmic reticulum stress

Obj.ctive: To investigate the pyroptosis-inducing effects of celastrol on tumor cells and to explore the potential mechanisms involved, specifically focusing on the role of the caspase-3/gasdermin E (GSDME) signaling pathway and the impact of endoplasmic reticulum (ER) stress and autophagy. Methods: Necrostatin-1 (Nec-1), lactate dehydrogenase release (LDH) assay, and Hoechst/propidium iodide (PI) double staining were employed to validate the mode of cell death. Western blot was used to detect the cleavage of GSDME and the expression of light chain 3 (LC3) and BIP. Results: Celastrol induced cell swelling with large bubbles, which is consistent with the pyroptotic phenotype. Moreover, treatment with celastrol induced GSDME cleavage, indicating the activation of GSDME-mediated pyroptosis. GSDME knockout via CRISPR/Cas9 blocked the pyroptotic morphology of celastrol in HeLa cells. In addition, cleavage of GSDME was attenuated by a specific caspase-3 inhibitor in celastrol-treated cells, suggesting that GSDME activation was induced by caspase-3. Mechanistically, celastrol induced endoplasmic reticulum (ER) stress and autophagy in HeLa cells, and other ER stress inducers produced effects consistent with those of celastrol. Conclusion: These findings suggest that celastrol triggers caspase-3/GSDME-dependent pyroptosis via activation of ER stress, which may shed light on the potential antitumor clinical applications of celastrol. © 2024 Beij.ng University of Chinese Medicine

Electroacupuncture with different waveforms for primary dysmenorrhea: A randomized controlled trial

Obj.ctive: To observe and compare the clinical effects of different electroacupuncture waveforms on primary dysmenorrhea. Methods: This was a prospective, randomized, three-group, parallel-controlled trial. Participants with primary dysmenorrhea were randomly divided into dense-sparse wave, continuous wave, and discontinuous wave groups in a 1:1:1 ratio. Two lateral Ciliao (BL 32) points were used. All three groups started treatment 3–5 days before menstruation, once a day for six sessions per course of treatment, one course of treatment per menstrual cycle, and three menstrual cycles. The primary outcome measure was the proportion with an average visual analog scale (VAS) score reduction of ≥50% from baseline for dysmenorrhea in the third menstrual cycle during treatment. The secondary outcome measures included changes in dysmenorrhea VAS scores, Cox Menstrual Symptom Scale scores and the proportion of patients taking analgesic drugs. Results: The proportion of cases where the average VAS score for dysmenorrhea decreased by ≥50% from baseline in the third menstrual cycle was not statistically significant (P > .05). Precisely 30 min after acupuncture and regarding immediate analgesia on the most severe day of dysmenorrhea, there was a statistically significant difference in the dense-sparse wave group compared with the other two groups during the third menstrual cycle (P mary dysmenorrhea and its related symptoms in patients. The three groups showed similar results in terms of short- and long-term analgesic efficacy and a reduction in the number of patients taking analgesic drugs. Regarding achieving immediate analgesia, the dense-sparse wave group was slightly better than the other two groups. © 2024 Beij.ng University of Chinese Medicine

Universal insertion of molecules in ionic compounds under pressure

Using first-principles calculations and crystal structure search methods, we found that many covalently bonded molecules such as H2, N2, CO2, NH3, H2O and CH4may react with NaC l, a prototype ionic solid,and form stable compounds under pressure while retaining their molecular structure. These molecules,despite whether they are homonuclear or heteronuclear, polar or non-polar, small or large, do not show strong chemical interactions with surrounding Na and Cl ions. In contrast, the most stable molecule among all examples, N2, is found to transform into cyclo-N5-anions while reacting with NaC l under high pressures. It provides a new route to synthesize pentazolates, which are promising green energy materials with high energy density. Our work demonstrates a unique and universal hybridization propensity of covalently bonded molecules and solid compounds under pressure. This surprising miscibility suggests possible mixing regions between the molecular and rock layers in the interiors of large planets.

Generalized Hopf Bifurcation in a Delay Model of Neutrophil Cells Model

The DDE-Biftool software is applied to solve the dynamical stability and bifurcation problem of the neutrophil cells model. Based on Hopf point finding with the stability property of the equilibrium solution loss, the continuation of the bifurcating periodical solution starting from Hopf point is exploited. The generalized Hopf point is tracked by seeking for the critical value of free parameter of the switching phenomena of the open loop, which describes the lineup of bifurcating periodical solutions from Hopf point. The normal form near the generalized Hopf point is computed by Lyapunov-Schimdt reduction scheme combined with the center manifold analytical technique. The near dynamics is classified by geometrically different topological phase portraits.

Potential Secretory Transporters and Biosynthetic Precursors of Biological Nitrification Inhibitor 1,9-Decanediol in Rice as Revealed by Transcriptome and Metabolome Analyses

Biological nitrification inhibitors(BNIs)are released from plant roots and inhibit the nitrification activity of microorganisms in soils,reducing NO_(3)^(‒)leaching and N2O emissions,and increasing nitrogenuse efficiency(NUE).Several recent studies have focused on the identification of new BNIs,yet little is known about the genetic loci that govern their biosynthesis and *** applied a combined transcriptomic and metabolomic analysis to investigate possible biosynthetic pathways and transporters involved in the biosynthesis and release of BNI 1,9-decanediol(1,9-D),which was previously identified in rice root *** results linked four fatty acids,icosapentaenoic acid,linoleate,norlinolenic acid,and polyhydroxy-α,ω-divarboxylic acid,with 1,9-D biosynthesis and three transporter families,namely the ATP-binding cassette protein family,the multidrug and toxic compound extrusion family,and the maj.r facilitator superfamily,with 1,9-D release from roots into the soil *** finding provided candidates for further work on the genes implicated in the biosynthesis and secretion of 1,9-D and pinpoint genetic loci for crop breeding to improve NUE by enhancing 1,9-D secretion,with the potential to reduce NO_(3)^(‒)leaching and N2O emissions from agricultural soils.

many migratory birds exhibit interannual consistency in migration schedules, routes and stopover sites. Detecting the interannual consistency in spatiotemporal characteristics helps understand the maintenance of migration and enables the implementation of targeted conservation measures. We tracked the migration of Whimbrel (Numenius phaeopus) in the East Asian-Australasian Flyway and collected spatiotemporal data from individuals that were tracked for at least two years. Wilcoxon non-parametric tests were used to compare the interannual variations in the dates of departure from and arrival at breeding/nonbreeding sites, and the interannual variation in the longitudes when the same individual across the same latitudes. Whimbrels exhibited a high degree of consistency in the use of breeding, nonbreeding, and stopover sites between years. The variation of arrival dates at nonbreeding sites was significantly larger than that of the departure dates from nonbreeding and breeding sites. Repeatedly used stopover sites by the same individuals in multiple years were concentrated in the Yellow Sea coast during northward migration, but were more widespread during southward migration. The stopover duration at repeatedly used sites was significantly longer than that at sites used only once. When flying across the Yellow Sea, Whimbrels breeding in Sakha (Yakutia) exhibited the highest consistency in migration routes in both autumn and spring. Moreover, the consistency in migration routes of Yakutia breeding birds was generally higher than that of birds breeding in Chukotka. Our results suggest that the northward migration schedule of the Whimbrels is mainly controlled by endogenous factors, while the southward migration schedule is less affected by endogenous factors. The repeated use of stopover sites in the Yellow Sea coast suggests this region is important for the migration of Whimbrel, and thus has high conservation value.

Enhanced properties of stone coal-based composite phase change materials for thermal energy storage

Phase change materials (PCMs) can be incorporated with low-cost minerals to synthesize composites for thermal energy storage in building *** coal (SC) after vanadium extraction treatment shows potential for secondary utilization in composite *** prepared SC-based composite PCMs with SC as a matrix,stearic acid (SA) as a PCM,and expanded graphite (EG) as an *** combined roasting and acid leaching treatment of raw SC was conducted to understand the effect of vanadium extraction on promoting loading *** showed that the combined treatment of roasting at 900℃ and leaching increased the SC loading of the composite by 6.2%by improving the specific surface *** loading capacity and thermal conductivity of the composite obviously increased by 127%and 48.19%,respectively,due to the contribution of 3wt% *** data were supported by the high load of 66.69%and thermal conductivity of 0.59 W·m^(-1)·K-1of the designed *** obtained composite exhibited a phase change temperature of 52.17℃,melting latent heat of 121.5 j.g^(-1),and good chemical *** SC-based composite has prospects in building applications exploiting the secondary utilization of minerals.

In view of the problem that the sensing characteristics of the multi-mode interferometric fiber sensors cannot be accurately analyzed,an analysis method based on the fast Fourier transform(FFT)and inverse fast Fourier transform(IFFT)is proposed and demonstrated theoretically and *** suitabilities of the rectangular window function with the narrow main lobe(high spectrum resolution)and low side lobe(high main mode energy leakage)and the Hanning window function with the wide main lobe(low spectrum resolution)and high side lobe(high energy concentration)in this kind of sensor analysis are discussed,*** method can not only realize the sensing performance analysis of the various modes,but also overcome the inconsistency of the different interference wavelength(dip)sensing characteristics in the conventional analysis *** the same time,this method is also beneficial to solve the repetitive problem of such sensors.

Night warming increases wheat yield by improving pre-anthesis plant growth and post-anthesis grain starch biosynthesis

Global climate change is characterized by asymmetric warming,i.e.,greater temperature increases in winter,spring,and nighttime than in summer,autumn,and *** experiments were conducted using four wheat cultivars,namely‘Yangmai 18’(YM18),‘Sumai 188’(SM188),‘Yannong 19’(YN19),and‘Annong 0711’(AN0711),in the two growing seasons of 2019-2020 and 2020-2021,with passive night warming during different periods in the early growth *** treatments were night warming during the tillering-j.inting(NW_(T-j.),j.inting-booting(NWj.B),and booting-anthesis(NWB-A)stages,with ambient temperature(NN)as the *** effects of night warming during different stages on wheat yield formation were investigated by determining the characteristics of dry matter accumulation and translocation,as well as sucrose and starch accumulation in wheat *** wheat yields of all four cultivars were significantly higher in NW_(T-j.than in NN in the 2-year *** yield increases of semi-winter cultivars YN19 and AN0711 were greater than those of spring cultivars YM18 and *** NW_(T-j.increased wheat yield mainly by increasing the 1,000-grain weight and the number of fertile spikelets,and it increased dry matter accumulation in various organs of wheat at the anthesis and maturity stages by increasing the growth rate at the vegetative growth *** flag leaf and spike showed the largest increases in dry matter ***_(T-j.also increased the grain sucrose and starch contents in the early and middle grain-filling stages,promoting yield ***,night warming between the tillering and j.inting stages increased the pre-anthesis growth rate,and thus,wheat dry matter production,which contributed to an increase in wheat yield.
