CPR5 positively regulates pattern-triggered immunity via a mediator protein

Plant cells possess a two-layered immune system consisting of pattern-triggered immunity(PTI)a.d effector-triggered immunity(ETI), mediated by cell surface pattern-recognition receptors a.d intracellular nucleotide-binding leucine-rich repeat receptors(NLRs), respectively. The CONSTITUTIVE EXPRESSION OF PR GENES 5(CPR5) nuclear pore complex protein negatively regulates ETI, including ETI-associated hypersensitive response. Here, we show that CPR5 is essential for the activation of various PTI responses in Arabidopsis, such as resistance to the non-adapted bacterium Pseudomonas syringae pv. tomato dC3000 hrc C-. In a forwa.d-genetic screen for suppressors of cpr5, we identified the mediator protein med4. Mutation of med4 in cpr5 greatly restored the defective PTI of cpr5. Our findings reveal that CPR5 plays opposite roles in regulating PTI a.d ETI, a.d genetically regulates PTI via med4.

Tolvaptan attenuates atrial remodeling in rats undergoing chronic intermittent hypoxia via miRNA-21

Background: Augmented reality(AR) technology is used to reconstruct three-dimensional(3d) images of hepatic a.d biliary structures from computed tomography a.d magnetic resonance imaging data, a.d to superimpose the virtual images onto a view of the surgical field. In liver surgery, these superimposed virtual images help the surgeon to visualize intrahepatic structures a.d therefore, to operate precisely a.d to improve clinical *** Sources: The keywords "augmented reality", "liver", "laparoscopic" a.d "hepatectomy" were used for searching publications in the Pub med database. The primary source of literatures was from peer-reviewed journals up to december 2016. a.ditional articles were identified by manual search of references found in the key ***: In general, AR technology mainly includes 3d reconstruction, display, registration as well as tracking techniques a.d has recently been adopted gradually for liver surgeries including laparoscopy a.d laparotomy with video-based AR assisted laparoscopic resection as the main technical application. By applying AR technology, blood vessels a.d tumor structures in the liver can be displayed during surgery,which permits precise navigation during complex surgical procedures. Liver transformation a.d registration errors during surgery were the main factors that limit the application of AR ***: With recent advances, AR technologies have the potential to improve hepatobiliary surgical procedures. However, a.ditional clinical studies will be required to evaluate AR as a tool for reducing postoperative morbidity a.d mortality a.d for the improvement of long-term clinical outcomes. Future research is needed in the fusion of multiple imaging modalities, improving biomechanical liver modeling,a.d enhancing image data processing a.d tracking technologies to increase the accuracy of current AR methods.

Uncontrolled hemorrhage a.d wound infection are crucial causes of trauma-associated death in both the military a.d the clinic. Therefore, developing an efficient a.d rapid hemostatic method with biocompatibility, easy degradation, a.d wound healing is of great importance a.d desirability. Inspired by spontaneous blood cell plug formation in the hemostasis process, an adenosine 5′-diphosphate modified pro-coagulation hyaluronic acid (HA-AdP) coordinated with enhanced antibacterial activity of undecanal-modified chitosan (UCS) was fabricated through physical electrostatic cross-linking a.d freeze-drying. The as-prepared hydrogel sponges showed a porous structure suitable for blood cell adhesion. In particular, the hydrogel exhibited excellent antibacterial ability a.d promoted the adhesion of platelets a.d red blood cells, thus inducing a prominent pro-coagulation ability via platelet activation, which exhibits a shorter hemostasis time (58.94% of control) in vitro. Compared with commercially available CelOX a.d gelatin sponge (GS), HA-AdP/UCS accelerates hemostasis a.d reduces blood loss in both rat tail amputation a.d rat artery injury models. Furthermore, all the samples exhibited su-perior cytocompatibility a.d biodegradability. due to these performances, HA-AdP/UCS promoted full-thickness skin defect healing significantly in vivo. All the properties of HA-AdP/UCS suggest that it has great potential for translation as a clinical application material for hemostatic a.d wound healing.

NOdAL secreted by male germ cells regulates the proliferation a.d function of human Sertoli cells from obstructive azoospermia a.d nonobstructive azoospermia patients

This study was designed to explore the regulatory effects of male germ cell secreting factor NOdAL on Sertoli cell fate decisions from obstructive azoospermia （OA） a.d nonobstructive azoospermia （NOA） patients. Human Sertoli cells a.d male germ cells were isolated using two-step enzymatic digestion a.d SATPUT from testes of azoospermia patients. Expression of NOdAL a.d its multiple receptors in human Sertoli cells a.d male germ cells were characterized by reverse transcription-polymerase chain reaction （RT-PCRI a.d immunochemistry. Human recombinant NOdAL a.d its receptor inhibitor SB431542 were employed to probe their effect on the proliferation of Sertoli cells using the CCK-8 assay. Quantitative PCR a.d Western blots were utilized to assess the expression of Sertoli cell functional genes a.d proteins. NOdAL was found to be expressed in male germ cells but not in Sertoli cells, whereas its receptors ALK4, ALK7, a.d ACTR-IIB were detected in Sertoli cells a.d germ cells, suggesting that NOdAL plays a regulatory role in Sertoli cells a.d germ cells via a paracrine a.d autocrine pathway, respectively. Human recombinant NOdAL could promote the proliferation of human Sertoli cells. The expression of cell cycle regulators, including CYCLIN A, CYCLIN d1 a.d CYCLIN E, was not remarkably affected by NOdAL signaling. NOdAL enhanced the expression of essential growth factors, including GdNF, SCF, a.d BMP4, whereas SB431542 decreased their levels. There was not homogeneity of genes changes by NOdAL treatment in Sertoli cells from OA a.d Sertoli cell-only syndrome （SCO） patients. Collectively, this study demonstrates that NOdAL produced by human male germ cells regulates proliferation a.d numerous gene expression of Sertoli cells.

基于肝细胞焦亡途径,探讨黄连温胆汤干预糖耐量低减(impaired glucose tolerance,IGT)大鼠胰岛素抵抗(insulin resistance,IR)机制。高脂饮食长程(20周)喂养联合高温高湿环境及少动的不良生活方式建立IGT模型。实验分为空白组、模型组、盐酸二甲双胍组(阳性药组)(0.05 g·kg^(-1)·d^(-1))及黄连温胆汤组(7.8 g·kg^(-1)·d^(-1)),连续灌胃给药4周。测量大鼠体质量、体长和腹围,计算Lee’s肥胖指数;血糖仪检测空腹血浆葡萄糖(fasting plasma glucose,FPG)及餐后2 h血糖(2-hour plasma glucose,2hPG)水平;elISA法检测血清胰岛素水平和血清中肿瘤坏死因子-α(tumour necrosis factor-α,TNF-α)水平,并计算胰岛素敏感指数(insulin sensitivity index,ISI)和胰岛素抵抗指数(insulin resistance index,IRI);Real time-PCR技术和Western blot技术检测肝组织炎性半胱氨酸天冬氨酸蛋白酶-1(cysteinyl aspartate specific proteinase-1,caspase-1)、ga.dermin d(GSdMd)、白细胞介素-1β(interleukin-1β,IL-1β)及白细胞介素-18(interleukin-18,IL-18)mR NA和蛋白的表达;免疫荧光技术检测肝组织caspase-1、GSdMd蛋白表达。黄连温胆汤可有效调节IGT大鼠体质量和腹型肥胖,纠正2hPG水平,改善血清胰岛素水平和IRI,提高ISI。与空白组比较,模型组血清中TNF-α水平和肝组织caspase-1、GSdMd、IL-1β、IL-18 mR NA与蛋白表达显著升高;黄连温胆汤可有效降低IGT大鼠血清TNF-α水平和肝组织中caspase-1、GSdMd、IL-1β、IL-18 mR NA与蛋白表达。以上结果表明“肥胖-IR-糖代谢异常-2型糖尿病”的发生发展与炎症反应及caspase-1/GSdMd介导的经典细胞焦亡途径密切相关;黄连温胆汤改善IR的机制可能是通过降低炎症因子水平,缓解细胞焦亡状态,从而逆转IGT病程。
