Mutual antagonism of mouse-adaptation mutations in HA and pa proteins on H9N2 virus replication

Avian H9N2 viruses have wide host range among the influenza A ***,knowledge of H9N2 mammalian adaptation is *** explore the molecular basis of the adaptation to mammals,we performed serial lung passaging of the H9N2 strain A/chicken/Hunan/8.27 YYGK3W3-OC/2018(3W3)in mice and identified six mutations in the hemagglutinin(HA)and polymerase acidic(pa)*** L226Q,T511I,and A528V of HA were responsible for enhanced pathogenicity and viral replication in mice;notably,HA-L226Q was the key *** T97I,I545V,and S594G of pa contributed to enhanced polymerase activity in mammalian cells and increased viral replication levels in vitro and in ***-T97I increased viral polymerase activity by accelerating the viral polymerase complex *** findings revealed that the viral replication was affected by the presence of pa-97I and/or pa-545V in combination with a triple-point HA ***,the double-and triple-point pa mutations demonstrated antagonistic effect on viral replication when combined with ***,any combination of pa mutations,along with double-point HA mutations,resulted in antagonistic effect on viral *** also observed antagonism in viral replication between pa-545V and pa-97I,as well as between HA-528V and *** findings demonstrated that several antagonistic mutations in HA and pa proteins affect viral replication,which may contribute to the H9N2 virus adaptation to mice and mammalian *** findings can potentially contribute to the monitoring of H9N2 field strains for assessing their potential risk in mammals.

Metal–organic framework wafer enabled fast response radiation detection with ultra-low dark current

Semiconductive metal–organic frameworks(MOFs)have attracted great interest for the electronic ***,dark currents of present hybrid organic–inorganic materials are 1000–10,000 times higher than those of commercial inorganic detectors,leading to poor charge ***,we demonstrate a ZIF-8(Zn(mim)_(2),mim=2-methylimidazolate)wafer with ultra-low dark current of 1.27 pa·mm^(-2) under high electric fields of 322 V·mm^(-1).The isostatic pressing preparation process provides ZIF-8 wafers with good ***,the presence of redox-active metals and small spatial separation between components promotes the charge *** ZIF-8-based semiconductor detector shows promising X-ray detection sensitivity of 70.82μC·Gy^(-1)·cm^(-2) with low doses exposures,contributing to superior X-ray imaging capability with a relatively high spatial resolution of 1.2 lp·mm^(-1).Simultaneously,good peak discrimination with the energy resolution of~43.78%is disclosed when the detector is illuminated by uncollimated 241Am@5.48 MeVα-*** results provide a broad prospect of MOFs for future radiation detection applications.

Application value of dexmedetomidine in anesthesia for elderly patients undergoing radical colon cancer surgery

BACKGROUND Colon cancer presents a substantial risk to the well-being of elderly people *** advancements in medical technology,surgical treatment has become the primary approach for managing colon cancer ***,due to age-related physiological changes,especially a decline in cognitive function,older patients are more susceptible to the effects of surgery and anesthesia,increasing the relative risk of postoperative cognitive dysfunction(POCD).There-fore,in the surgical treatment of elderly patients with colon cancer,it is of pa-ramount importance to select an appropriate anesthetic approach to reduce the occurrence of POCD,protect brain function,and improve surgical success *** One hundred and seventeen patients with colon cancer who underwent elective surgery under general anesthesia were selected and divided into two groups:A and *** A received Dex before anesthesia induction,and B group received an equivalent amount of normal *** in the mini-mental state exami-nation,regional cerebral oxygen saturation(rSO2),bispectral index,glucose uptake rate(GluER),lactate production rate(LacPR),serum S100βand neuron-specific enolase(NSE),POCD,and adverse anesthesia reactions were compared between the two *** Surgical duration,duration of anesthesia,and intraoperative blood loss were comparable between the two groups(P>0.05).The overall dosage of anesthetic drugs used in group A,including propofol and remifentanil,was significantly lower than that used in group B(Piately after extubation,higher GluER values and lower LacPR values at the time of endotra-cheal intubation,30 min after the start of surgery,immediately after extubation,and 5 min after extubation(P<0.05).Group A exhibited lower levels of serum S100βand NSE 24 h postoperatively and a lower incidence of cognitive dysfunction on the 1st and

Swine are regarded as“intermediate hosts”or“mixing vessels”of influenza viruses,capable of generating strains with pandemic *** 2020 to 2021,we conducted surveillance on swine H1N2 influenza(swH1N2)viruses in swine farms located in Guangdong,Yunnan,and Guizhou provinces in southern China,as well as Henan and Shandong provinces in northern *** systematically analyzed the evolution and pathogenicity of swH1N2 isolates,and characterized their replication and transmission *** isolated viruses are quadruple reassortant H1N2 viruses containing genes from pdm/09 H1N1(PB2,PB1,pa and NP genes),triple-reassortant swine(NS gene),Eurasian Avian-like(HA and M genes),and recent human H3N2(NA gene)*** NA,PB2,and NP of SW/188/20 and SW/198/20 show high gene similarities to A/Guangdong/Yue Fang277/2017(H3N2).The HA gene of swH1N2 exhibits a high evolutionary *** five swH1N2 isolates replicate efficiently in human,canine,and swine cells,as well as in the turbinate,trachea,and lungs of mice.A/swine/Shandong/198/2020 strain efficiently replicates in the respiratory tract of pigs and effectively transmitted among ***,these current swH1N2 viruses possess zoonotic potential,highlighting the need for strengthened surveillance of swH1N2 viruses.

Angiosperm-wide analysis of fruit and ovary evolution aided by a new nuclear phylogeny supports association of the same ovary type with both dry and fleshy fruits

Sulfation of chondroitin and bile acids converges to antagonize Wnt/β-catenin signaling and inhibit APC deficiency-induced gut tumorigenesis

基于可靠度研究超高性能混凝土(Ultra‐High Performance Concrete,UHPC)增强钢筋短搭接RC构件的失效模式与承载力特征。首先阐述了UHPC加固机理,建立了包含活恒荷载效应比、搭接系数m、UHPC强度、混凝土强度、钢筋型号、考虑水平地震作用的弯矩效应比和轴力效应比等关键参数的设计空间;确立分析RC构件承载力可靠度的功能函数,运用Monte Carlo法对设计空间中的关键参数组进行可靠指标计算,然后进行关键参数敏感性分析和计算结果拟合分区。结果表明:设计空间关键参数中,搭接系数、效应比、配筋率、钢筋型号和UHPC强度对可靠指标的影响均显著;UHPC不仅具有良好的承载力增强能力,还能改变失效模式,梁和柱屈服时可靠指标分别提高109.6%和12.9%,屈服时搭接系数分别为0.58和0.57;对于受弯RC梁(压弯柱),m>0.67(0.65)时,钢筋基本能屈服,建议采用UHPC加固;0.50(0.49)
