Effects of P301L-taU on post-translational modifications of microtubules in human iPSC-derived cortical neurons and taU transgenic mice

taU is a microtubule-associated protein that promotes microtubule assembly and stability in the *** is missorted and aggregated in an array of diseases known as *** are essential for neuronal function and regulated via a complex set of post-translational modifications,changes of which affect microtubule stability and dynamics,microtubule interaction with other proteins and cellular structures,and mediate recruitment of microtubule-severing *** impairment of microtubule dynamics causes neuronal dysfunction,we hypothesize cognitive impairment in human disease to be impacted by impairment of microtubule *** therefore aimed to study the effects of a disease-causing mutation of taU(P301L)on the levels and localization of microtubule post-translational modifications indicative of microtubule stability and dynamics,to assess whether P301L-taU causes stability-changing modifications to *** investigate taU localization,phosphorylation,and effects on tubulin post-translational modifications,we expressed wild-type or P301L-taU in human maPT-KO induced pluripotent stem cell-derived neurons(i Neurons)and studied taU in neurons in the hippocampus of mice transgenic for human P301L-taU(p R5 mice).Human neurons expressing the longest taU isoform(2N4R)with the P301L mutation showed increased taU phosphorylation at the AT8,but not the p-Ser-262 epitope,and increased polyglutamylation and acetylation of microtubules compared with endogenous taU-expressing neurons.P301L-taU showed pronounced somatodendritic presence,but also successful axonal enrichment and a similar axodendritic distribution comparable to exogenously expressed 2N4R-wildtype-taU.P301L-taU-expressing hippocampal neurons in transgenic mice showed prominent missorting and tauopathy-typical AT8-phosphorylation of taU and increased polyglutamylation,but reduced acetylation,of microtubules compared with non-transgenic *** sum,P301L-taU results in changes in micr

As the scale of the power system continues to expand,the environment for power operations becomes more and more *** risk management and control methods for power operations can only set the same risk detection standard and conduct the risk detection for any scenario ***,more reliable and accurate security control methods are urgently *** order to improve the accuracy and reliability of the operation risk management and control method,this paper proposes a method for identifying the key links in the whole process of electric power operation based on the spatiotemporal hybrid convolutional neural *** provide early warning and control of targeted risks,first,the video stream is framed adaptively according to the pixel changes in the video ***,the optimized MobileNet is used to extract the feature map of the video stream,which contains both time-series and static spatial scene *** feature maps are combined and non-linearly mapped to realize the identification of dynamic operating ***,training samples and test samples are produced by using the whole process image of a power company in Xinjiang as a case study,and the proposed algorithm is compared with the unimproved *** experimental results demonstrated that the method proposed in this paper can accurately identify the type and start and end time of each operation link in the whole process of electric power operation,and has good real-time *** average accuracy of the algorithm can reach 87.8%,and the frame rate is 61 frames/s,which is of great significance for improving the reliability and accuracy of security control methods.

We investigate and concentrate on new infinitesimal generators of Lie symmetries for an extended(2+1)-dimensional Calogero-Bogoyavlenskii-Schif(eCBS)equation using the commutator table which results in a system of nonlinear ordinary differential equations(ODEs)which can be manually *** two stages of Lie symmetry reductions,the eCBS equation is reduced to non-solvable nonlinear ODEs using different combinations of optimal Lie *** the integration method and the Riccati and Bernoulli equation methods,we investigate new analytical solutions to those *** substituting to the original variables generates new solutions to the eCBS *** results are simulated through three-and two-dimensional plots.

This work aims to present nonlinear models that arise in ocean *** are many models of ocean waves that are present in *** shallow water,the linearization of the equations requires critical conditions on wave capacity than it make in deep water,and the strong nonlinear belongings are *** use Lie symmetry analysis to obtain different types of soliton solutions like one,two,and three-soliton solutions in a(2+1)dimensional variable-coefficient Bogoyavlensky Konopelchenko(VCBK)equation that describes the interaction of a Riemann wave reproducing along the y-axis and a long wave reproducing along the x-axis in engineering and *** use the Lie symmetry analysis then the integrating factor method to obtain new solutions of the VCBK *** demonstrate the physical meaning of the solutions obtained by the presented techniques,the graphical performance has been demonstrated with some *** presented equation has fewer dimensions and is reduced to ordinary differential equations using the Lie symmetry technique.

Mixed sensitivity H_∞ control for LTI systems with varying time delays

Designing a robust controller for a system with timevarying delays poses a major challenge. In this paper, we propose a method based on mixed sensitivity H∞ for the control of linear time invariant（LTI） systems with varying time delays. The time delay is assumed bounded and the upper bound is known. In the technique we propose, the delay affecting the plant to be controlled is treated as an unmodeled uncertainty（in form of multiplicative uncertainty）. That uncertainty is approximated and then an H∞based controller, for the plant represented by the multiplicative uncertainty and the nominal model, is calculated. The obtained H∞controller is used to control the LTI systems with varying time delays. Simulation examples are given to illustrate the effectiveness of the proposed method.
