Interleukin 17(IL-17)is an important cytokine that can induce tissue inflammation and is involved in the pathogenesis of numerous autoimmune ***,the regulation of its signaling transduction has not been well *** this study,we report that thousand and one kinase 1(TAOK1)functions as a negative regulator of IL-17-mediated signal transduction and ***1 knockdown promotes IL-17-induced cytokine and chemokine expression and the activation of mitogen-activated protein kinases and nuclear factor-κ*** further demonstrate that TAOK1 interacts with IL-17 receptor A(IL-17RA)independent of its kinase activity,and TAOK1 dose-dependently prevents the formation of the IL-17R-Act1(nuclear factor activator 1,also known as tumor necrosis factor receptor-associated factor 3 interacting protein 2)*** with this,TAOK1 deficiency exacerbates colitis in the 2,4,6-trinitrobenzenesulfonic acid-induced experimental model of inflammatory bowel disease,likely by its promotion of the IL-17-mediated signaling ***1 expression is decreased in the colons of ulcerative colitis *** conclusion,these findings suggest that TAOK1 is involved in the development of IL-17-related autoimmune disorders.

早古生代加里东造山运动使整个华南板块产生了广泛的岩浆活动,而关于华南早古生代的构造属性是俯冲碰撞还是陆内造山尚存争议。对钦杭结合带的I型花岗岩成因研究对揭示桂东南地区早古生代地球动力学背景及其构造演化具有重要的地质意义。本文对桂东/南莲垌和古龙岩体中的花岗岩样品进行了全岩地球化学分析、LA-ICP-MS锆石U-Pb定年和Lu-Hf同位素分析。莲垌和古龙岩体花岗岩的形成时代相近,分别为435~434和436~435 ma。莲垌和古龙岩体均为花岗闪长岩,并具有相似的地球化学特征:10000Ga/Al值为2.07,w(Zr+Nb+Ce+Y)值为183×10^(-6),A/CNK为0.87~1.1,平均值为0.97,εHf(t)值为-2.2~1.8,主要集中在0~2,对应的T DM2年龄主要分布在1.6~1.2 Ga,峰值在1.5~1.3 Ga,Nb/Ta值为11.47~14.66,平均值为12.80,Rb/Sr值为0.22~0.52。以上分析结果表明:莲垌和古龙岩体均为钙碱性I型花岗岩,来源于中元古代基底变质火成岩的部分熔融,并可能含一定的幔源物质。其主要形成过程为:华南板块南部与冈瓦纳北缘大规模碰撞所产生的应力,自南向北、自东向西传入华南板块,引发华南板块陆内造山作用。加厚的岩石圈经历了造山后伸展减薄,地幔物质上涌为下地壳部分熔融提供了热量,形成新生地壳熔融型(如古龙岩体)或新生地壳与古老地壳混合熔融型(如莲垌岩体)的母岩浆;在陆内造山作用期后的伸展环境下,母岩浆沿通道上涌,形成了具有俯冲碰撞特征的I型花岗岩。

SFRP2 affects prenatal muscle development and is regulated by micro RNA-1/206 in pigs

Secreted frizzled-related protein 2 （SFRP2）, a member of the SFRPs family, is associated with cell growth and differentiation in myogenesis. Our previous study suggested that SFRP2 was a potential target of microRNA （miRNA）-I/206, which was considered as myomiRs. To further explore the biological function and regulation mechanisms of the SFRP2 gene in porcine skeletal muscle development, we first analyzed the sequence structure of the porcine SFRP2 gene. Subsequently, we detected its tissue distribution in adult Tongcheng pigs （a Chinese indigenous breed） and investigated its dynamic expression in developmental skeletal muscle （13 prenatal and 7 postnatal time points）in Tongcheng pigs. An interaction analysis between SFRP2 and myomiRs was also performed. The results showed that the expression pattern of the SFRP2 varied greatly across diverse tissues. It exhibited abundant expression in prenatal skeletal muscle and peaked at 55 days post coitus （E55）, and had a lower expression in postnatal skeletal muscle, indicating that the SFRP2 gene might affect porcine embryonic skeletal muscle development. Co-expression analysis revealed that the expression levels of SFRP2 correlated negatively with miRNA-1 （t=-0.570, P-value=0.009） and miRNA-206 （r=-0.546, P-value=0.013）, but positively with SFRP1 （r=0.613, P-value=0.004）. The bioinformatics analysis and dual luciferase assay verified that the SFRP2 was a putative target of miRNA-1/206 in pigs. Therefore, this study is helpful for understanding the biological function and mo- lecular regulation of the SFRP2 gene during porcine skeletal muscle development.

Transcriptomic analysis elucidates the enhanced skeletal muscle mass, reduced fat accumulation, and metabolically benign liver in human follistatin-344 transgenic pigs

Follistatin(FST) is an important regulator of skeletal muscle growth and adipose deposition through its ability to bind to several members of the transforming growth factor-β(TGF-β) superfamily, and thus may be a good candidate for future animal breeding programs. However, the molecular mechanisms underlying the phenotypic changes have yet to be clarified in pig. We generated transgenic(TG) pigs that express human FST specifically in skeletal muscle tissues and characterized the phenotypic changes compared with the same tissues in wild-type pigs. The TG pigs showed increased skeletal muscle growth, decreased adipose deposition, and improved metabolism status(Pmajor muscle. Furthermore, the lipid metabolism-related process was changed in adipose tissue in the TG pigs. Gene set enrichment analysis revealed that genes related to lipid synthesis, lipid catabolism, and lipid storage were down-regulated(P1) in the TG pigs for subcutaneous fat, whereas genes related to lipid catabolism were significantly up-regulated(P<0.05) in the TG pigs for retroperitoneal fat compared with their expression levels in wild-type pigs. In liver, genes related to the TGF-β signaling pathway were over-represented in the TG pigs, which is consistent with the inhibitory role of FST in regulating TGF-β signaling. Together, these results provide new insights into the molecular mechanisms underlying the phenotypic changes in pig.

Microstructure characteristics of Ni-43Ti-4Al-2Nb-2Hf alloy prepared by conventional casting and directional solidification

To further investigate the microstructure characteristic and solidification mechanism, so as to provide knowledge for the microstructure control of a NiTi-AI based high-temperature structural material, the microstructure of Ni-43Ti-4AI-2Nb-2Hf （at.%） alloy ingots prepared by conventional casting （arc-melting） and directional solidification （DS） at various drawing velocities （2 ***-＇, 18 ***-1, 30 mm-min-＇ and 60 mmmin~, respectively） was investigated by means of electron probe microanalyses. Experimental results reveal that the microstructures are composed of NiTi matrix phase,/3-Nb phase and Ti2Ni phase for samples obtained by both conventional casting and DS. Conventional casting has an equiaxial structure, while DS has a slender and acicular cellular structure which grows along the [001] orientation preferentially. Small amounts of white/3-Nb phase and black Ti2Ni phase co-exist at the grain boundaries or intercellular regions. With an increase in drawing velocity, the NiTi matrix phase is inclined to grow along （100） and （200） crystallographic planes, and the cellular arm spacing reduce gradually, but the directionality of the solidified structure weakens significantly. The homogeneous dispersion of,8-Nb phase and the decrease of Ti2Ni phase in DS samples are beneficial to improving the mechanical properties. Solidification mechanism analysis indicates that the dark grey NiTi matrix phase initially precipitates from the liquid phase, and then the divorced eutectic reaction takes place, which produces the light gray matrix phase and/^-Nb phase. Finally, the peritectic reaction happens, which generates the black Ti2Ni phase.
