Single-cell RNA-seq reveals the transcriptional program underlying tumor progression and metastasis in neuroblastoma

Neuroblastoma (NB) is one of the most common childhood malignancies. Sixty percent of patients present with widely disseminated clinical signs at diagnosis and exhibit poor outcomes. However, the molecular mechanisms triggering NB metastasis remain largely uncharacterized. In this study, we generated a transcriptomic atlas of 15 447 NB cells from eight NB samples, including paired samples of primary tumors and bone marrow metastases. We used time-resolved analysis to chart the evolutionary trajectory of NB cells from the primary tumor to the metastases in the same patient and identified a common 'starter' subpopulation that initiates tumor development and metastasis. The 'starter' population exhibited high expression levels of multiple cell cycle-related genes, indicating the important role of cell cycle upregulation in NB tumor progression. In addition, our evolutionary trajectory analysis demonstrated the involvement of partial epithelial-to-mesenchymal transition (p-EMT) along the metastatic route from the primary site to the bone marrow. Our study provides insights into the program driving NB metastasis and presents a signature of metastasis-initiating cells as an independent prognostic indicator and potential therapeutic target to inhibit the initiation of NB metastasis.

Functions of the adaptor protein p66shc in solid tumors

p66she is a 66 kDa Src homology 2 domain containing （Shc） adaptor protein homolog. Previous studies have demonstrated that p66she is a proapoptotic protein involved in the cellular response to oxidative stress and in regulating mammalian lifespan. However, accumulating evidence also indicates its critical role in solid tumor progression. The expression of p66she varies in different types of solid tumors, and it can paradoxically promote as well as suppress tumor progression, survival, and metastasis, depending on the cellular context. In this review, we discuss its functions in various solid tumors, the mechanisms by which it mediates the process of anoikis （detachment-induced cell death）, and the epigenetic mechanisms that regulate its expression. These studies indicate the potential of p66she as a novel prognostic marker and therapeutic target for the prevention of tumor progression and metastasis.

Androgen receptor(AR)mutation is closely associated with prostate cancer(PCa)and is one of the mechanisms of resistance to PCa therapies such as AR *** sequencing technologies like next-generation sequencing(NGS)contributes to the high-throughput and precise detection of AR mutations carried by PCa patients,the lack of interpretations of these clinical genetic variants has still been a roadblock for PCa-targeted precision ***,we established a designer yeast reporter assay to simulate natural androgen receptor(AR)selection using AR *** HIS3 gene transactivation was associated with the ligand-induced recruitment of steroid receptor coactivator-1(SRC-1)by AR mutants,where yeast growth in histidine-free medium was determined as the *** assay is applicable to determine a wide range of clinical AR mutants including those with loss of function relating to androgen insensitivity syndrome(AIS),and those associated with PCa conferring resistance to AR antagonists such as enzalutamide(ENZ),bicalutamide(BIC),and cyproterone acetate(CPA).One clinical AR mutant previously reported to confer ENZ-resistance,F877L,was found to confer partial resistance to CPA as well using designer *** simple and efficient assay can enable precise one-pot screening of AR mutants,providing a reference for tailored medicine.

Reinforcement learning behavioral control(RLBC)is limited to an individual agent without any swarm mission,because it models the behavior priority learning as a markov decision *** this paper,a novel multi-agent reinforcement learning behavioral control(maRLBC)method is proposed to overcome such limitations by implementing joint ***,a multi-agent reinforcement learning mission supervisor(maRLMS)is designed for a group of nonlinear second-order systems to assign the behavior priorities at the decision *** modeling behavior priority switching as a cooperative markov game,the maRLMS learns an optimal joint behavior priority to reduce dependence on human intelligence and high-performance computing *** the control layer,a group of second-order reinforcement learning controllers are designed to learn the optimal control policies to track position and velocity signals *** particular,input saturation constraints are strictly implemented via designing a group of adaptive *** simulation results show that the proposed maRLBC has a lower switching frequency and control cost than finite-time and fixed-time behavioral control and RLBC methods.

Breaking the optical efficiency limit of virtual reality with a nonreciprocal polarization rotator

A catadioptric lens structure,also known as pancake lens,has been widely used in virtual reality(VR)displays to reduce the ***,the utilization of a half mirror(HM)to fold the optical path thrice leads to a significant optical *** theoretical maximum optical efficiency is merely 25%.To transcend this optical efficiency constraint while retaining the foldable characteristic inherent to traditional pancake optics,in this paper,we propose a theoretically lossless folded optical system to replace the HM with a nonreciprocal polarization *** our feasibility demonstration experiment,we used a commercial Faraday rotator(FR)and reflective polarizers to replace the lossy *** theoretically predicted 100%efficiency can be achieved approximately by using two high-extinction-ratio reflective *** addition,we evaluated the ghost images using a micro-OLED panel in our imaging ***,the ghost images can be suppressed to undetectable level if the optics are with antireflection *** novel pancake optical system holds great potential for revolutionizing next-generation VR displays with lightweight,compact formfactor,and low power consumption.

Confusing Object Detection: A Survey

Confusing object detection (COD), such as glass, mirrors, and camouflaged objects, represents a burgeoning visual detection task centered on pinpointing and distinguishing concealed targets within intricate backgrounds, leveraging deep learning methodologies. Despite garnering increasing attention in computer vision, the focus of most existing works leans toward formulating task-specific solutions rather than delving into in-depth analyses of methodological structures. As of now, there is a notable absence of a comprehensive systematic review that focuses on recently proposed deep learning-based models for these specific tasks. To fill this gap, our study presents a pioneering review that covers both the models and the publicly available benchmark datasets, while also identifying potential directions for future research in this field. The current dataset primarily focuses on single confusing object detection at the image level, with some studies extending to video-level data. We conduct an in-depth analysis of deep learning architectures, revealing that the current state-of-the-art (SOTA) COD methods demonstrate promising performance in single object detection. We also compile and provide detailed descriptions of widely used datasets relevant to these detection tasks. Our endeavor extends to discussing the limitations observed in current methodologies, alongside proposed solutions aimed at enhancing detection accuracy. Additionally, we deliberate on relevant applications and outline future research trajectories, aiming to catalyze advancements in the field of glass, mirror, and camouflaged object detection. © 2024 The Authors.
