Polysaccharide of Alocasia cucullata Exerts Antitumor Effect by Regulating Bcl-2, Caspase-3 and ERK1/2 Expressions during Long-Time Administration

Objective: To study the in vitro and in vivo antitumor effects of the polysaccharide of Alocasia cucullata(PAC) and the underlying mechanism. Methods: B16F10 and 4T1 cells were cultured with PAC of 40 μg/m L, and PAC was withdrawn after 40 days of administration. The cell viability was detected by cell counting kit-8. The expression of Bcl-2 and Caspase-3 proteins were detected by Western blot and the expressions of ERK1/2 mRNA were detected by quantitative real-time polymerase chain reaction(qRT-PCR). A mouse melanoma model was established to study the effect of PAC during long-time administration. Mice were divided into 3 treatment groups: control group treated with normal saline, positive control group(LNT group) treated with lentinan at 100 mg/(kg·d), and PAC group treated with PAC at 120 mg/(kg·d). The pathological changes of tumor tissues were observed by hematoxylin-eosin staining. The cell apoptosis of tumor tissues was detected by TUNEL staining. Bcl-2 and Caspase-3 protein expressions were detected by immunohistochemistry, and the expressions of ERK1/2, JNK1 and p38 mRNA were detected by qRT-PCR. Results: In vitro, no strong inhibitory effects of PAC were found in various tumor cells after 48 or 72 h of administration. Interestingly however, after 40 days of cultivation under PAC, an inhibitory effect on B16F10 cells was found. Correspondingly, the long-time administration of PAC led to downregulation of Bcl-2 protein(P mote apoptosis of tumor cells, and had obvious antitumor effect in tumor-bearing mice.

目的探讨钩藤碱对甲基苯丙胺依赖人神经母细胞瘤细胞(SH-SY5Y细胞)模型及微小RNA-375-3p(miR-375-3p)/胚胎致死异常视觉-样4蛋白(Elavl4)表达的影响。方法用最大安全剂量诱导法建立甲基苯丙胺依赖细胞模型。将细胞分成正常组(完全培养液)、对照组(完全培养液+400μmol·L^(-1)钩藤碱孵育48 h)、模型组(完全培养液+100μmol·L^(-1)甲基苯丙胺孵育48 h)、实验组(完全培养液+400μmol·L^(-1)的钩藤碱孵育15 min后,再加入100μmol·L^(-1)甲基苯丙胺孵育48 h)。以酶联免疫吸附法检测细胞环腺苷酸(cAMP)、5-羟色胺(5-HT)水平;以实时荧光定量逆转录聚合酶链反应(qRT-PCR)检测细胞miR-375-3p表达情况;以蛋白质印迹法检测Elavl4蛋白表达情况。结果正常组、对照组、模型组和实验组cAMP表达水平分别为(6.33±0.93)、(6.57±1.12)、(10.89±1.03)和(7.81±1.32)pmol·mg^(-1);5-HT分别为(682.46±17.32)、(690.31±15.09)、(510.11±27.67)和(649.99±21.42)pg·mL^(-1);miR-375-3p表达水平分别为1.00±0.13、1.13±0.24、3.48±0.18和1.58±0.19;Elavl4表达水平分别为1.00±0.05、0.89±0.10、0.50±0.09和0.90±0.11。模型组上述指标与正常组比较,差异均有统计学意义(P<0.01,P<0.05);实验组上述指标与模型组比较,差异均有统计学意义(P<0.05,P<0.01)。结论本研究建立甲基苯丙胺依赖细胞模型,同时发现钩藤碱可能调控miR-375-3p/Elavl4表达拮抗甲基苯丙胺成瘾。

Placental site trophoblastic tumor(PSTT),a rare gestational trophoblastic neoplasm(GTN),exhibits slow growth,late-onset metastasis,and chemotherapy insensitivity[1,2].The invasive capacity of trophoblasts induces angiogenesis in PSTT,yet the specific mechanism remains unclear[3].PSTT is typically resistant to chemotherapy and hysterectomy stands as the primary therapeutic choice for patients with an overall cure rate of approximately 75%-80%[4,5].Therefore,investigating the precise emergence and progression of PSTT is crucial,aiming to identify novel targets for diagnosis and therapy.

Preparation and Structure-Property Regulation Mechanism of Reversible Thermochromic Polydiacetylene

The thermochromic mechanism and the structure-property regulation principle of reversible thermochromic polydiacetylene (PDA) materials have always been a challenging issue. In this work, a series of diacetylene monomers (m-PCDA) containing phenyl and amide or carboxyl groups were synthesized from 10,12-pentacosadiynoic acid (PCDA) through the esterification or amidation reactions. The effects of the number and the distribution of the functional groups in m-PCDA molecules on their solid-state polymerization capability, and the thermochromic mechanism of their corresponding polymers (m-PDA) were investigated and discussed in detail. The results show that the m-PCDA monomers containing both benzene ring and groups that can form hydrogen bonding interactions have strong intermolecular interaction, and are easy to carry out the solid phase polymerization under 254-nm UV irradiation to obtain the corresponding new thermochromic m-PDA materials. The thermochromic behavior of m-PDA depends on its melting process. The initial color-change temperature (blue to red) is determined by the onset melting temperature, and the temperature range in which reversible color recovery can be achieved by repeat heating-cooling treatment is determined by its melting range. According to the proposed thermochromic mechanism of PDA, various new PDA materials with precise thermochromic temperatures and reversible thermochromic temperature ranges can be designed and synthesized through the appropriate introduction of benzene ring and groups that can form hydrogen bonding interactions into the molecular structure of linear diacetylene monomer. This work provides a perspective to the precise molecular structure design and the property regulation of the reversible thermochromic PDA materials.

Present situation and prospect of immunotherapy for unresectable locally advanced esophageal cancer during peri-radiotherapy

Four major studies(Checkmate577,Keynote-590,Checkmate649 and Attraction-4)of locally advanced esophageal cancer published in 2020 have established the importance of immunotherapy,represented by anti-programmed death protein(PD)-1 in postoperative adjuvant treatment and advanced first-line treatment of locally advanced or advanced esophageal cancer and esophagogastric junction cancer,from the aspects of proof of concept,long-term survival,overall survival rate and progression-free *** unresectable or inoperable nonmetastatic esophageal cancer,concurrent radiotherapy and chemotherapy is the standard treatment recommended by various *** its curative effect is still not ideal,it is necessary to explore radical radiotherapy and chemotherapy in the future,and it is considered to be promising to combine them with immunotherapeutic drugs such as *** paper mainly discusses how to combine radical concurrent radiotherapy and chemotherapy with immunotherapy for unresectable local advanced esophageal cancer.
