Adult Stress Promotes Purinergic Signaling to Induce Visceral Pain in Rats with Neonatal Maternal Deprivation

Chronic visceral pain is one of the primary symptoms of patients with irritable bowel syndrome(IBS),which affects up to 15%of the population *** detailed mechanisms of visceral pain remain largely *** previous studies have shown that neonatal maternal deprivation(NMD)followed by adult multiple stress(AMS)advances the occurrence of visceral pain,likely due to enhanced norepinephrine(NE)-β2 adrenergic *** study was designed to explore the roles of P2X3 receptors(P2X3Rs)in the chronic visceral pain induced by combined ***,we showed that P2X3 Rs were co-expressed inβ2 adrenergic receptor(β2-AR)-positive dorsal root ganglion neurons and that NE significantly enhanced ATP-induced Ca2+*** and AMS not only significantly increased the protein expression of P2X3Rs,but also greatly enhanced the ATP-evoked current density,number of action potentials,and intracellular Ca2+concentration of colon-related DRG *** injection of the P2X3R inhibitor A317491 greatly attenuated the visceral pain and the ATP-induced Ca2+signals in NMD and AMS ***,theβ2-AR antagonist butoxamine significantly reversed the expression of P2X3Rs,the ATP-induced current density,and the number of action potentials of DRG ***,our data demonstrate that NMD followed by AMS leads to P2X3R activation,which is most likely mediated by upregulation ofβ2 adrenergic signaling in primary sensory neurons,thus contributing to visceral hypersensitivity.

Radio Frequency Interference(RFI)mitigation is essential for supporting the science output of Five-hundred-meter Aperture Spherical radio Telescope(FAST)due to its high *** order to protect FAST from RFI,an Electromagnetic Compatibility(EMC)study has been carried out and the operation of a Radio Quiet Zone(RQZ)is *** measurements of the telescope instruments and monitoring of the active radio services outside the site have revealed the radiation properties of the RFI *** on the measurement results and theoretical analysis,various EMC methods have been implemented for the telescope to decrease the ***,the main RFI sources in the FAST RQZ,such as mobile stations,broadcast stations and navigation instruments,have been identified,and the technical measures have been adopted to protect the quiet radio environment around the *** early science outputs of FAST have demonstrated the efficiency of RFI mitigation methods.

By dynamic method under UV irradiation, commercial melt-blown polypropylene （PPMB） filter element was modified with acrylamide （AAm） using benzophenone （BP） as initiator. Attenuated total reflection-Fourier transform infrared spectroscopy and scanning electron microscope verified that polyacrylamide chain was grafted on the fiber surface of PPMB filter element. Elemental content analysis with energy dispersive X-ray of fibers revealed that the polymerization content in the inner part of filter element was relatively higher than that in the outer. Degree of grafting changed with initiator concentration, monomer concentration, reaction temperature and reached 2.6% at the reaction condition： CBp=0.06 mol/L, CAAm=2.0 mol/L, irradiation time： 80 min, temperature： 60℃. Relative water flux altered with the hydrophilicity and pore size of filter element. In the antifouling test, the modified filter gave greater flux recovery （approximately 70%） after filtration of the water extract of Liuweidihuang, suggesting that the fouling layer was more easily reversible due to the hydrophilic nature of the modified filter.

Aging is accompanied by the decline of organismal functions and a series of prominent hallmarks,including genetic and epigenetic *** aging-associated epigenetic changes include DNA methylation,histone modification,chromatin remodeling,non-coding RNA(ncRNA)regulation,and RNA modification,all of which participate in the regulation of the aging process,and hence contribute to aging-related ***,understanding the epigenetic mechanisms in aging will provide new avenues to develop strategies to delay ***,aging interventions based on manipulating epigenetic mechanisms have led to the alleviation of aging or the extension of the lifespan in animal *** molecule-based therapies and reprogramming strategies that enable epigenetic rejuvenation have been developed for ameliorating or reversing aging-related *** addition,adopting health-promoting activities,such as caloric restriction,exercise,and calibrating circadian rhythm,has been demonstrated to delay ***,various clinical trials for aging intervention are ongoing,providing more evidence of the safety and efficacy of these ***,we review recent work on the epigenetic regulation of aging and outline the advances in intervention strategies for aging and age-associated diseases.A better understanding of the critical roles of epigenetics in the aging process will lead to more clinical advances in the prevention of human aging and therapy of aging-related diseases.

A correlation of computed tomography perfusion and histopathology in tumor edges of hepatocellular carcinoma

BACKGROUND： The peripheral morphologic characteristics of hepatocellular carcinoma （HCC） reflect tumor growth patterns. Computed tomography （CT） perfusion is a new method to analyze hemodynamic changes in tissues. We assessed the relationship between CT perfusion and histopathologic findings in the periphery of HCC lesions. METHODS： Non-contrast CT, enhanced dual-phase CT, and CT perfusion were performed on 77 subjects （47 patients and 30 controls）. Based on the imaging findings of enhanced dual- phase CT, the tumor edges were classified into three types： type Ⅰ （sharp）; type Ⅱ （blurry）; and type Ⅲ （mixed）. The CT perfusion parameters included hepatic blood flow, hepatic arterial fraction, hepatic arterial perfusion, and hepatic portal perfusion. The tissue sections from resected specimens were subjected to routine hematoxylin and eosin staining and immunohistochemical staining for CD34. The correlations between microvessel density （MVD） and the CT perfusion parameters were analyzed using Pearson＇s product-moment correlation coefficient. Changes in the perfusion parameters in tumor edges of different tumor types were evaluated. RESULTS： Type Ⅰ （sharp）： the pathologic findings showed fibrous connective tissue capsules in the tumor edges, and an MVD 〈30/ram2. Type Ⅱ （blurry）： the histology showed that the edges were clear with no capsules and an MVD 〉30/ram2. Type Ⅲ （mixed）： the pathology was similar to that of types I and II, and an MVD 〉30/mm~. hepatic blood flow, hepatic arterial fraction, hepatic arterial perfusion, and hepatic portal perfusion were significantly increased in the tumor edges of HCC patients compared to those of the controls （P〈0.05）. The correlation between CT perfusion parameters and MVD was higher in blurry tumor edges of type II than in those of types Ⅰ or Ⅲ. CONCLUSION： CT perfusion imaging of tumor edges may be helpful in revealing histopathological features, and indirectly reflect angiogenic changes of HCCs.

Malignant glioma is a highly heterogeneous and invasive primary brain tumor characterized by high recurrence rates,resistance to combined therapy,and dismal *** stem cells(GSCs)are likely responsible for tumor progression,resistance to therapy,recurrence,and poor prognosis owing to their high self-renewal and tumorigenic *** a family member of BMP signaling,bone morphogenetic protein4(BMP4)has been reported to induce the differentiation of GSCs and neural stem cells(NSCs).However,the molecular mechanisms underlying the BMP4-mediated effects in these two cell types are *** this study,we treated hGSCs and hNSCs with BMP4 and com-pared the phenotypic and transcriptional changes between these two cell ***,we found that the growth of hGSCs was greatly inhibited by BMP4,but the same treatment only increased the cell size of *** the RNA sequencing results showed that BMP4 treatment evoked significantly transcriptional changes in both hGSCs and hNSCs,the profiles of differentially expressed genes were distinct between the two groups.A gene set that specifically targeted the proliferation and differentiation of hGSCs but not hNSCs was enriched and then validated in hGSC *** results suggested that hGSCs and hNSCs responded differently to BMP4 *** and investigating different responses between hGSCs and hNSCs will benefit finding partner factors working together with BMP4 to further suppress GSCs proliferation and stemness without disturbing NSCs.

The neutron capture cross section of 197 Au was measured using the time-of-flight(TOF)technique at the Back-n facility of the China Spallation Neutron Source(CSNS)in the 1 eV to 100 keV *** c-rays originating from neutron-induced capture events were detected by four C_(6)D_(6) liquid scintillator *** height weighting technology(PHWT)was used to analyze the *** results are in good agreement with ENDF/B-VIII.0,CENDL-3.1,and other evaluated libraries in the resonance region,and in agreement with both n TOF and GELINA experimental data in the 5–100 keV ***,the resonance peaks in the energy range from 1eV to 1 keV were fitted by the SAMMY R-matrix code.

Zircon is a key accessary mineral for metamorphic geochronology and geochemical tracing,but it has been a challenge to interpret its complex chemical zoning and age record acquired during multiple episodes of anatectic metamorphism in collisional *** is illustrated by a combined study of petrography,phase equilibrium modeling and metamorphic P-T-t determination for granulites from the Bohemian Massif in the Variscan *** rocks record multiple episodes of zircon growth during anatectic *** started from the compressional heating for prograde metamorphism to high-pressure(HP)to ultrahigh-pressure(UHP)eclogite facies with low degrees of partial ***,they underwent a decompressional stage from UHP eclogite facies to HP granulite facies for dehydration *** were followed by a further decompressional stage either to kyanite granulite facies or to sillimanite granulite facies at ultrahigh-temperature(UHT)*** of zircon growth are linked to specific metamorphic conditions for peritectic reactions on the basis of zoning patterns,trace element signatures,index mineral inclusions in dated domains and textural relationships to coexisting *** results indicate that relict zircon domains are preserved even at UHT granulite facies conditions.A few zircon domains in the kyanite granulite grew during the prograde to peak UHP metamorphism,possibly corresponding to consumption of biotite and plagioclase but growth of *** the decompressional exhumation to the HP granulite-facies,relict or prograde zircon domains were mostly dissolved into anatectic melts produced by muscovite *** zircon grains grew during this transition to the HP granulite-facies in the kyanite granulite and are chemically related to continuous growth of garnet,whereas abundant zircon grains grew subsequently at the UHT granulite facies in the sillimanite granulite and are chemically related to garnet breakdown reactions.A

In this article,we present detailed seasonal,monthly and daily statistics of temperature difference in the surface layer at the Muztagh-ata site based on the temperature measurements at two heights of 2 m and6 *** find that temperature inversion occurs frequently at our site during nighttime,especially during the cold *** temperature inversion always represents stable atmospheric turbulence,which is crucial for an optical *** analyzing the behavior of temperature inversion and its correlation with wind and cloud amount,one conclusion can be made that radiation inversion is the main reason for the existence of temperature inversion in the surface-layer at the Muztagh-ata site.
