Assessment of the safety and efficacy of combination chemotherapy and PD-1/PD-L1 inhibitor treatment of breast cancer:A meta-analysis

Background:As the efficacy of programmed cell death-1/programmed death-ligand 1(PD-1/PD-L1)inhibitors combined with chemotherapy in curing breast cancer is still controversial,this meta-analysis compares the efficacy and safety of PD-1/PD-L1 inhibitors combined with chemotherapy and chemotherapy alone in the treatment of breast cancer,which provides guidance for the clinical treatment.Methods:Relevant studies published as of April 2022 in the various databases including EMBASE,PubMed,and Cochrane Library were selected.Randomized controlled trials(RCTs)in which control patients underwent chemotherapy alone and experimental group patients underwent combination chemotherapy and PD-1/PD-L1 inhibitor treatment were included in this investigation.Investigations without complete information,researches from which information could not be extracted,duplicate articles,animal studies,review articles,and systematic reviews were excluded.STATA 15.1 was employed for all statistical analyses.Results:In total,eight eligible studies were identified,revealing that combination chemotherapy and PD-1/PD-L1 inhibitor treatment was linked to significant increases in progression-free survival(PFS)relative to chemotherapy alone(hazard ratio[HR]=0.83,95%confidence interval[CI]:0.70–0.99,P=0.032)but not overall survival(HR=0.92,95%CI:0.80–1.06,P=0.273).Pooled adverse event rates were also increased within the group of combination treatment relative to the chemotherapy group(risk ratio[RR]=1.08,95%CI:1.03–1.14,P=0.002).Specifically,nausea rates were lesser within the group of combination treatment relative to the group of chemotherapy(RR=0.48,95%CI:0.25–0.92,P=0.026).Subgroup analyses indicated that the PFS of patients who underwent combination atezolizumab or pembrolizumab and chemotherapy treatment were substantially longer than those of patients who underwent chemotherapy alone(HR=0.79,95%CI:0.69–0.89,P≤0.001;HR=0.79,95%CI:0.67–0.92,P=0.002).Conclusions:The pooled results suggest that combination chemotherapy and PD-1/PD-L1 inhibitor tre

Dynamic process analysis of the Niumiangou landslide triggered by the Wenchuan earthquake using the finite difference method and a modified discontinuous deformation analysis

The Niumiangou landslide was the largest landslide triggered by the 2008 Wenchuan earthquake,which was significantly affected by the amplification effect of seismic acceleration.The ringshear experiments indicated that the materials in the source area of the Niumiangou landslide were subjected to friction degradation under a big shear displacement,which may result in rapid movement of the landslide.In order to better understand the landslide movement and study the effect of the friction degradation on movement mechanisms,the dynamic process of Niumiangou landslide was simulated with a new numerical method,which combines the finite difference method(FDM)and the discontinuous deformation analysis(DDA).First,the FDM was used to study the initiation time,amplification effect and velocity of the landslide.Afterwards,these initiation velocities were applied to the blocks in the DDA model by corresponding coordination in the FDM model.A displacementdependent friction model of the sliding surface was incorporated into DDA code to further understand the kinetic behavior of the landslide.The results show that the displacement-dependent friction strongly decreases the friction coefficient of sliding surface under a big displacement,which can obviously promote the run-out and velocity of landslide.The model output well matches the topographic map formed by the landslide.This implies that the proposed model can be applied to the simulation of earthquake-induced landslides with amplification effect,and the friction degradation model is important to clarify the movement mechanism of high-speed and long-distance landslides.

Pivotal micro factors associated with endothelial cells

Objective:Recent studies have shown the important influence of various micro factors on the general biological activity and function of endothelial cells (ECs).Vascular endothelial growth factor (VEGF) and angiogenin (ANG) are classic micro factors that promote proliferation,differentiation,and migration of ECs.The underlying pathophysiological mechanisms and related pathways of these micro factors remain the focus of current research.Data sources:An extensive search was undertaken in the PubMed database by using keywords including "micro factors" and "endothelial cell." This search covered relevant research articles published between January 1,2007 and December 31,2018.Study selection:Original articles,reviews,and other articles were searched and reviewed for content on micro factors of ECs.Results:VEGF and ANG have critical functions in the occurrence,development,and status of the physiological pathology of ECs.Other EC-associated micro factors include interleukin 10,tumor protein P53,nuclear factor kappa B subunit,interleukin 6,and tumor necrosis factor.The results of Gene Ontology analysis revealed that variations were mainly enriched in positive regulation of transcription by the RNA polymerase Ⅱ promoter,cellular response to lipopolysaccharides,negative regulation of apoptotic processes,external side of the plasma membrane,cytoplasm,extracellular regions,cytokine activity,growth factor activity,and identical protein binding.The results of the Kyoto Encyclopedia of Genes and Genomes analysis revealed that micro factors were predominantly enriched in inflammatory diseases.Conclusions:In summary,the main mediators,factors,or genes associated with ECs include VEGF and ANG.The effect of micro factors on ECs is complex and multifaceted.This review summarizes the correlation between ECs and several micro factors.

Exploration of potential biomarkers and therapeutic targets for trauma-related acute kidney injury

Purpose:Acute kidney injury(AKI)is one of the most common functional injuries observed in trauma patients.However,certain trauma medications may exacerbate renal injury.Therefore,the early detection of trauma-related AKI holds paramount importance in improving trauma prognosis.Methods:Qualified datasets were selected from public databases,and common differentially expressed genes related to trauma-induced AKI and hub genes were identified through enrichment analysis and the establishment of protein-protein interaction(PPI)networks.Additionally,the specificity of these hub genes was investigated using the sepsis dataset and conducted a comprehensive literature review to assess their plausibility.The raw data from both datasets were downloaded using R software(version 4.2.1)and processed with the"affy"package19 for correction and normalization.Results:Our analysis revealed 585 upregulated and 629 downregulated differentially expressed genes in the AKI dataset,along with 586 upregulated and 948 downregulated differentially expressed genes in the trauma dataset.Concurrently,the establishment of the PPI network and subsequent topological analysis highlighted key hub genes,including CD44,CD163,TIMP metallopeptidase inhibitor 1,cytochrome b-245 beta chain,versican,membrane spanning 4-domains A4A,mitogen-activated protein kinase 14,and early growth response 1.Notably,their receiver operating characteristic curves displayed areas exceeding 75%,indicating good diagnostic performance.Moreover,our findings postulated a unique molecular mechanism underlying trauma-related AKI.Conclusion:This study presents an alternative strategy for the early diagnosis and treatment of trauma-related AKI,based on the identification of potential biomarkers and therapeutic targets.Additionally,this study provides theoretical references for elucidating the mechanisms of trauma-related AKI.

To the Editor:Breast cancer(BC)is the most common malignancy in women.Statistics show that 1.2 million women worldwide have BC each year,and 500,000 women die from BC.It is the leading cause of death in women.[1]According to the different expressions of estrogen receptor(ER),progesterone receptor(PR),and epidermal growth factor receptor 2(HER2),three receptor markers,BC is divided into four subtypes in clinical diagnosis.[2]MicroRNAs(miRNAs)have a wide range of regulatory effects and can regulate a variety of normal or pathogenic cell processes.

A numerical method of combined SPF-MEM-LBM on the rockfall-induced surge and its application

Rockfalls in reservoirs are prone to induce surges, posing a severe threat to passing vessels and facilities. A scheme combined Single-phase freesurface method(SPF), momentum exchange method(MEM), and Lattice Boltzmann method(LBM) is proposed to predict the impact of rockfall-induced surges. First, the LBM-SPF model is used to simulate the motion of the free surface, and the MEM model is used to calculate the hydrodynamic force acting on rock mass. To address the incompatibility issue arising from the coupling of LBM-SPF model and MEM model, a correction scheme inside the solid is induced. The simulation results of the single particle and double particle sedimentation in cavity show the feasibility and accuracy of the method designed in this paper. Moreover, the validation experiments of Scott Russel’s wave generator show that the proposed scheme can simulate wave profile stably. The simulation results emphasize that the waves induced by rockfalls have a significant impact on the safe operation of the Laxiwa dam and the passing vessels in the reservoir.

Effect of seepage-induced erosion on soil macropore structure

Internal erosion is one of the important factors causing geological disasters.The microstructure of soil can change with seepage erosion,resulting in changes in the hydraulic and mechanical properties of the soil.The evolution of seepage erosion is investigated with X-ray computed tomography(CT)in this study.The change in macropore structure characteristics during the seepage erosion test is quantified and the influence of seepage erosion on soil deformation is analyzed.Moreover,a pore network model(PNM)is established for the specimens and the evolution of the connected pore size characteristics is assessed.The results show that the macropore structure is significantly affected by seepage erosion,especially in terms of the porosity and pore geometry characteristics.The changes in macropore structure characteristics are most obvious in the lower part of the specimen.The influence of seepage erosion on the pore size distribution(PSD)and soil deformation is heterogeneous and closely dependent on the spatial location of the soil.Moreover,seepage erosion enhances macropore connectivity and has a directional impact on macropore orientation.These findings can provide a reference for the theoretical modeling and numerical simulation of the seepage erosion and improve the understanding of the seepage erosion evolution in engineering practice.

Hybrid data-driven framework for shale gas production performance analysis via game theory, machine learning, and optimization approaches

A comprehensive and precise analysis of shale gas production performance is crucial for evaluating resource potential,designing a field development plan,and making investment decisions.However,quantitative analysis can be challenging because production performance is dominated by the complex interaction among a series of geological and engineering factors.In fact,each factor can be viewed as a player who makes cooperative contributions to the production payoff within the constraints of physical laws and models.Inspired by the idea,we propose a hybrid data-driven analysis framework in this study,where the contributions of dominant factors are quantitatively evaluated,the productions are precisely forecasted,and the development optimization suggestions are comprehensively generated.More specifically,game theory and machine learning models are coupled to determine the dominating geological and engineering factors.The Shapley value with definite physical meaning is employed to quantitatively measure the effects of individual factors.A multi-model-fused stacked model is trained for production forecast,which provides the basis for derivative-free optimization algorithms to optimize the development plan.The complete workflow is validated with actual production data collected from the Fuling shale gas field,Sichuan Basin,China.The validation results show that the proposed procedure can draw rigorous conclusions with quantified evidence and thereby provide specific and reliable suggestions for development plan optimization.Comparing with traditional and experience-based approaches,the hybrid data-driven procedure is advanced in terms of both efficiency and accuracy.

Truxene-based Conjugated Microporous Polymers via Different Synthetic Methods

Four truxene-based conjugated microporous polymers(Tr-CMPs)were prepared via different synthetic methods and their structure-property relationships were studied.The polymer networks have high Brunauer-Emmett-Teller(BET)specific surface areas ranging from 554 m^2·g^–1to 1024 m^2·g^–1.Pore sizes of the CMPs with different linkers are mainly located between 0.60 and 1.96 nm.Among all the Tr-CMPs,TrCMP4 has the highest BET surface area of 1024 m^2·g^–1and exhibits the highest H2 uptake of 0.88 wt%.Tr-CMP2 prepared by Suzuki-Miyaura coupling reaction has the highest photoluminescence quantum yields(PLQYs)of 13.06% and CO2 uptake of 6.25 wt%.

Construction of novel TiO2/Bi4Ti3O(12)/MoS2 core/shell nanofibers for enhanced visible light photocatalysis

TiO2/Bi4 Ti3 O12 hybrids have been widely prepared as promising photocatalysts for decomposing organic contaminations.However,the insufficient visible light absorption and low charge separation efficiency lead to their poor photocatalytic activity.Herein,a robust methodology to construct novel TiO2/Bi4 Ti3 O12/MoS2 core/shell structures as visible light photocatalysts is presented.Homogeneous bismuth oxyiodide(BiOI) nanoplates were immobilized on electrospun TiO2 nanofiber surface by successive ionic layer adsorption and reaction(SILAR) method.TiO2/Bi4 Ti3 O12 core/shell nanofibers were conveniently prepared by partial conversion of TiO2 to high crystallized Bi4 Ti3 O12 shells through a solid-state reaction with BiOI nanoplates,which is accompanied with certain transition of TiO2 from anatase to rutile phase.Afterwards,MoS2 nanosheets with several layers thick were uniform decorated on the TiO2/Bi4 TiO3 O12 fiber surface resulting in TiO2/Bi4 Ti3 O12/MoS2 structures.Significant enhancement of visible light absorption and photo-generated charge separation of TiO2/Bi4 Ti3 O12 were achieved by introduction of MoS2.As a result,the optimized TiO2/Bi4 Ti3 O12/MoS2-2 presents 60% improvement for photodegrading RhB after 120 min irradiation under visible light and 3 times higher of apparent reaction rate constant in compared with the TiO2/Bi4 Ti3 O12.This synthetic method can also be used to establish other photocatalysts simply at low cost,therefore,is suitable for practical applications.
