TL1A Promotes Fibrogenesis in Colonic Fibroblasts via the TGF-β1/Smad3 Signaling Pathway

Objective Intestinal fibrosis is a refractory complication of inflammatory bowel disease(IBD).Tumor necrosis factor ligand-related molecule-1A(TL1A)is important for IBD-related intestinal fibrosis in a dextran sodium sulfate(DSS)-induced experimental colitis *** study aimed to explore the effects of TL1A on human colonic *** A trinitrobenzene sulfonic acid(TNBS)-induced experimental colitis model of LCK-CD2-TL1A-GFP transgenic(Tg)or wild-type(WT)mice was established to determine the effect and mechanism of TL1A on intestinal *** human colonic fibroblast CCD-18Co cell line was treated concurrently with TL1A and human peripheral blood mononuclear cell(PBMC)*** proliferation and activation of CCD-18Co cells were detected by BrdU assays,flow cytometry,immunocytochemistry and Western *** metabolism was tested by Western blotting and real-time quantitative polymerase chain reaction(RT-qPCR).Results The level of collagen metabolism in the TNBS+ethyl alcohol(EtOH)/Tg group was greater than that in the TNBS+EtOH/WT *** growth factor-β1(TGF-β1)and p-Smad3 in the TNBS+EtOH/Tg group were upregulated as compared with those in the TNBS+EtOH/WT *** proliferation of CCD-18Co cells was promoted by the addition of human PBMC supernatant supplemented with 20 ng/mL TL1A,and the addition of human PBMC supernatant and TL1A increased CCD-18Co proliferation by 24.4%at 24 ***1A promoted cell activation and increased the levels of COL1A2,COL3A1,and TIMP-1 in CCD-18Co *** of CCD-18Co cells with TL1A increased the expression of TGF-β1 and *** TL1A promotes TGF-β1-mediated intestinal fibroblast activation,proliferation,and collagen deposition and is likely related to an increase in the TGF-β1/Smad3 signaling pathway.

Natural product-induced oxidative stress-synergistic anti-tumor effects of chemotherapeutic agents

Reactive oxygen species are closely related to tumor *** recent years,reactive oxygen species has become a hot spot in tumor therapy,and many natural substances in nature contain compound components with anti-tumor ***,there is a lack of discussion on the synergistic anti-tumor effects of natural products in combination with chemotherapeutic drugs through reactive oxygen *** terms“natural products”,“reactive oxygen species”,“anti-tumor”,and“chemotherapy”were used to identify the synergistic effects of natural *** conducted a systematic literature search in PubMed and Web of Science databases for relevant research articles and reviews published in recent *** systematically summarized the studies related to anti-tumor active ingredients in natural compounds in the field of reactive oxygen species in recent years.A total of 77 relevant literatures were *** them,45 literatures containing various natural products such as terpenoids,flavonoids,alkaloids,*** anti-tumor effects by regulating reactive oxygen species levels,and 32 literatures regarding adjunctive role of natural products in anti-tumor *** this study,we found that natural products exert anti-tumor effects by elevating reactive oxygen species *** provides strong theoretical support for future clinical studies.

In this study,we conducted simulations to find the geometric aberrations expected for images taken by the Main Survey Camera of the Chinese Space Station Telescope(CSST)due to its *** anticipated by previous work,our findings indicate that the geometric distortion of light impacts the focal plane's apparent scale,with a more pronounced influence as the size of the focal plane *** models suggest that the effect consistently influences the pixel scale in both the vertical and parallel *** apparent scale variation follows a sinusoidal distribution throughout one orbital *** reveal that the effect is particularly pronounced in the center of the Galaxy and gradually diminishes along the direction of ecliptic *** low ecliptic latitudes,the total aberration leads to about a 0.94 pixel offset(a 20 minute exposure)and a 0.26 pixel offset(a 300 s exposure)at the edge of the field of *** processings for the geometric effect during the CSST pre-and post-observation phases are presented.

Hydroxysafflor Yellow A Inhibits Pyroptosis and Protecting HUVECs from OGD/R via NLRP3/Caspase-1/GSDMD Pathway

Objective: To observe the protective effect and mechanism of hydroxyl safflower yellow A(HSYA)from myocardial ischemia-reperfusion injury on human umbilical vein endothelial cells(HUVECs). Methods:HUVECs were treated with oxygen-glucose deprivation reperfusion(OGD/R) to simulate the ischemia reperfusion model, and cell counting kit-8 was used to detect the protective effect of different concentrations(1.25–160 μmol/L)of HSYA on HUVECs after OGD/R. HSYA 80 μmol/L was used for follow-up experiments. The contents of inflammatory cytokines interleukin(IL)-18, IL-1β, monocyte chemotactic protein 1(MCP-1), tumor necrosis factor α(TNF-α) and IL-6 before and after administration were measured by enzyme-linked immunosorbent assay. The protein expressions of toll-like receptor, NOD-like receptor containing pyrin domain 3(NLRP3), gasdermin D(GSDMD) and GSDMD-N-terminal domain(GSDMD-N) before and after administration were detected by Western blot. NLRP3 inflammasome inhibitor cytokine release inhibitory drug 3 sodium salt(CRID3 sodium salt, also known as MCC950) and agonist were added, and the changes of NLRP3, cysteine-aspartic acid protease 1(Caspase-1), GSDMD and GSDMD-N protein expressions were detected by Western blot. Results: HSYA inhibited OGD/R-induced inflammation and significantly decreased the contents of inflammatory cytokines IL-18,IL-1β, MCP-1, TNF-α and IL-6(P<0.01 or P<0.05). At the same time, by inhibiting NLRP3/Caspase-1/GSDMD pathway, HSYA can reduce the occurrence of pyroptosis after OGD/R and reduce the expression of NLRP3,Caspase-1, GSDMD and GSDMD-N proteins(P<0.01). Conclusions: The protective effect of HSYA on HUVECs after OGD/R is related to down-regulating the expression of NLRP3 inflammasome and inhibiting pyroptosis.

Sea-level change in coastal areas of China:Status in 2021

The sea level in coastal areas of China reached the second highest in 2021,just after that recorded in *** force and dynamic analyses based on tide gauges,satellite observations,reanalysis data and regional numerical outputs were conducted to understand these abnormally high sea levels and determine their possible *** show that the coastal sea level of China had increased at an annual rate of 3.4±0.3 mm during 1980-2021,with an acceleration of 0.06±0.02 mm per *** superposition of significant oscillations of quasi-2,3-7,quasi-9,quasi-11,quasi-19 and 20-30 years contributed to the anomalously high sea *** negative-phased El Nino/Southern Oscillation was correlated with the anomalously high sea level and the north-south anti-phase pattern of the coastal sea level in ***,phase lags of 1-4 months occurred with the sea-level *** a decadal timescale,the Pacific Decadal Oscillation(PDO)was negatively correlated with the anomalous mean sea level(MSL),and the negative-phased PDO contributed to the anomalous sea-level change in ***,the monthly MSL peaked in April and July,and the contribution of wind stress to the anomalously high sea level was 38.5%in the south of the Taiwan Strait in April and 30%along the coast of China in *** results were consistent with the tide gauge and satellite *** agreement was also observed between the coastal sea-level fingerprint and the air and sea surface temperatures.

Multichromophoric architectures with wellorganized chromophoric arrangements are of vital importance to gain insights into interchromophoric ***,we report a novel heteroleptic self-assembly strategy and construct three achiral and one chiral sandwich-like heterochromophore complexes(Zn_(4)L^(G)L^(A-C)_(2)and Zn_(4)L_(G)L^(D)_(2)).Possible dynamic products,including homoleptic oligomers and isomers,are eliminated,producing achiral complexes with exceptional *** are observed in chiral complexes due to the flexible geometry of *** aligned chromophore arrangements with precise spacing and orientations are evidenced by single-crystal X-ray *** distinct segments in the ligands lead to different interchromophoric ***,energy transfer and tunable emission,including white-light emission,are achieved for Zn_(4)L^(G)L^(A)_(2)and Zn_(4)L^(G)L^(B)_(2).In addition,chirality transfer is observed for Zn_(4)L_(G)L^(D)_(2),due to conformational restriction from chiral LD,leading to chiral rotational conformations of tetraphenylethylene moiety in LG.
