A New Diterpene Extracted from Daphne Mucronata, Effects on Human K562 and CCRF-CEM Cell Lines

Background: Daphne Mucronata extract has a decreasing effect on the size of breast adenocarcinoma in rats. So in this study, the effect of Daphne Mucronata purified diterpene were investigated on co culture of human monocytes and two human leukemia cell lines (K562, CCRF-CEM). Materials and Methods: Each cell line mono-layer culture, in log phase growth, was treated with 10 to 160 μL of the extract (1 g/ml leave powder) and purified compound (0.94 nM). For a comparative study, Taxol (5 to 40 μM) was used in the presence and absence of LPS. Human monocytes were isolated by adhesion method. TNF-α in cultured media were measured by sensitive biotin-streptoavidin ELISA method. Results: Fifty percent of growth inhibition was shown by 160 μL (1:100 dilution, 0.5 g of the powdered leaves/ml) of the extract and 0.94 nM of the purified component, and there was more inhibition in K562 cells (P < 0.05). Four fold increases in growth inhibition was shown in co culture of isolated human monocytes and leukemia cell lines. There was a direct relationship between monocytes TNF-α secretion and growth inhibition degree. Conclusion: Daphne Mucronata extract and its purified diterpene through increasing monocytes TNF-α releasing, potentially inhibit Leukemia cell line.

马铃薯晚疫病广谱抗性基因Rpi-blb1(克隆于Solanum bulbocastanum)及其两个同源抗病基因Rpi-sto1(克隆于Solanum stoloniferum)和Rpi-pta1(克隆于Solanum papita)的抗性可能已被高度进化的晚疫病菌株所克服。为证实此观点,将从荷兰田间和墨西哥野外***和***病株上采集的晚疫病菌株重新接种于具有共同遗传背景(感病栽培品种Desiree)的Rpi-blb1及其同源基因转化体和3个野生种植株上,测试Rpi-blb1及其同源基因的抗性是否已被克服。结果表明,采自墨西哥野外***的两个菌株Pic99189和Pic99192已经克服了Rpi-blb1及其同源基因的抗性;在野生种植株的测试中,只有***的抗性明显地被菌株Pic99192所克服。同时发现在转基因植株离体叶片接种试验中,高抗晚疫病菌的转化体需要在多个转化体间进行选择。从采自中国4个不同地区晚疫病菌株接种***转化体的小规模测试中可推断,Rpi-blb1及其同源抗性基因在中国仍具有潜在的应用价值。

Metastatic melanoma to the common bile duct causing obstructive jaundice:A case report

Metastatic melanoma to the common bile duct is very rare with only 18 cases reported so far. We report a 46 year old women who, 18 mo after excision of a skin melanoma, developped a painless progressive obstructive jaundice. At operation a melanoma within the distal third of the common bile duct was found. There were no other secondaries within the abdomen. The common bile duct, including the tumor, was resected and anastomosed with Roux-en-Y jejunal limb. The patient survived 31 mo without any sign of local recurrence and was submitted to three other operations for axillar and brain secondaries, from which she finally died. Radical resection of metastatic melanoma to the common bile duct may result in lifelong relief of obstructive jaundice. It is safe and relatively easy to perform. In other cases, a less aggressive approach, stenting or bypass procedures, should be adopted.

Obstructive jaundice secondary to tuberculosis (TB) is extremely rare. It can be caused by TB enlargement of the head of the pancreas, TB lymphadenitis, TB stricture of the biliary tree, or a TB mass of the retroperitoneum. A 29-year-old man with no previous history of TB presented with abdominal pain, obstructive jaundice, malaise and weight loss. Ultrasonography (US), computer tomography (CT) scan and endoscopic retrograde cholangiopancreatography (ERCP) were suggestive of a stenosis of the distal common bile duct (CBD) caused by a mass in the posterior head of the pancreas. Tumor markers, CEA and CA19-9 were within normal limits. At operation, an enlarged, centrally caseous lymph node of the posterior head of the pancreas was found, causing inflammatory stenosis and a fistula with the distal CBD. The lymph node was removed and the bile duct resected and anastomosed with the Roux-en Y jejunal limb. Histology and PCR based-assay confirmed tuberculous lymphadenitis. After an uneventful postoperative recovery, the patient was treated with anti-tuberculous medication and remained well 2.5 years later. Though obstructive jaundice secondary to tuberculous lymphadenitis is rare, abdominal TB should be considered as a differential diagnosis in immunocompromised patients and in TB endemic areas. Any stenosis or fistulation into the CBD should also be taken into consideration, and biliary bypass surgery be performed to both relieve jaundice and prevent further stricture.

基孔肯雅热已传播到一些印度洋岛屿和印度,包括著名的旅游胜地。为了比较诊断试验的有效性并了解这次暴发的程度,我们应用3种诊断方法检测了2006年从印度洋地区回到欧洲的680个病人的720份标本。上半年证实有24.4%的基孔肯雅病毒感染,下半年有9.9%。症状发作后4天内采集的所有标本逆转录PCR结果阳性。免疫荧光检测法检测到有的病人在发病(下转封三)第1天用IgM抗体阳性,发病第2天检测到IgG抗体,所有病人在发病第5天后都可以检测到IgM和IgG。在症状发作后不久,病人出现IgG和IgM并有很高的病毒载量(某些大于109拷贝/mL血浆)。这些数据将有助于医疗机构为归来旅行者选择诊断性检验。2006年从印度洋地区回到欧洲的旅行者的基孔肯雅热诊断检测@Marcus Panning@Klaus Grywna@marjan van Esbroeck@修文琼$福建省疾病预防控制中心@张拥军$福建省疾病预防控制中心

Is Any Correlation between Platelet Indices with Extent of Coronary Artery Involvement in Ischemic Heart Diseases?

Ischemic heart disease (IHD) is the most common cause of death around the world. Nowadays Platelet counts (PC) and volumetric platelet indices are available routinely in most laboratories and reflect the level of mobility and production of platelets. It seems that the excessive flexibility and size of the platelets and their local activation have correlation with extent of ischemic heart disease. So our objective is the study of platelet indices in ischemic heart disease. This non-randomized prospective study was performed on 245 patients with ischemic heart disease, who underwent the coronary angiography. The patients were divided into four groups: stable angina, unstable angina, acute myocardial infarction and control group;and then platelet indices, including the platelet counts (PC), the average platelet volume (MPV), the Platelet Distribution Width (PDW) and plateletcrit (PCT) in each group with the extent of coronary disease were compared based on an Syntax Score system and observational methods. The average ages of the patients were 57 years and 65% of them were male and the rest were female. A significant difference is exists between indices in all three groups compared to the control that this difference was related to gender and the type of the coronary artery involvement. However, only in infarction group, PDW in different disease intensities was significantly different. In this study, unlike many of the previous studies no relationship was found between the MPV with the extent of coronary disease.

Multivariate Chemometric Analysis of a Polluted River of a Megalopolis

A chemometrical study regarding a 10-years water quality monitoring plan at 15 sampling points along a section of the Reconquista River and its stream channels, which embraces 21 campaigns, is presented. The original data were pre-treated in order to eliminate missing data and outliers, obtaining a final data matrix of 270 samples containing 26 physical-chemistry variables each. Multivariate statistical methods like multi curve resolution, canonical correlation analysis and factor analysis methods, as well as current univariate statistics were applied. The interpretation was simplified when variables were separated in groups containing environmentally and chemically related variables instead of analyzing them all together. These methods have shown that the presence of metals likely come from at least 3 different type of sources. Although the stream channels arriving to the main river course are highly polluted, their flow rates are so low that do not significantly decrease its water quality. They mainly contribute to the high levels of biochemical-oxygen demand and chemical-oxygen demand as well as nitrogen-content species. Furthermore, regarding metals, the pollutants coming from the upstream of the river is higher than those introduced by all channels.

Cystic lymphangioma of the pancreas

Lymphangioma of the pancreas is an extremely rare benign tumour of lymphatic origin,with fewer than 60 published cases. Histologically,it is polycystic,with the cysts separated by thin septa and lined with endothelial cells. Though congenital,it can affect all age groups,and occurs more frequently in females. Patients usually present with epigastric pain and an associated palpable mass. Complete excision is curative,even though,depending on the tumour location,surgery may be simple or involve extensive pancreatic resection and anastomoses. The authors present a 49-year-old woman in whom a polycystic septated mass,35 mm x 35 mm in size,was discovered by ultrasonography (US) in the body of the pancreas during investigations for epigastric pain and nausea. At surgery,a well circumscribed polycystic tumor was completely excised,with preservation of the pancreatic duct. The postoperative recovery was uneventful. Histology confirmed a microcystic lymphangioma of the pancreas. Immunohistochemistry showed cystic endothelial cells reactivity to factor Ⅷ -RA (++),CD31 (+++) and CD34 (-). Postoperatively,abdominal pain disappeared and the patient remained symptomfree for 12 mo until now. Although extremely rare,lymphangioma of the pancreas should be taken into consideration as a differential diagnosis of a pancreatic cystic lesion,especially in women.
