Mesenchymal stem cells(MSCs)and their byproducts have been widely validated as potential therapeutic products for regenerative *** therapeutic effects result mainly from the paracrine activity of MSCs,which consists of the secretion of bioactive molecules,whether dispersed in medium conditioned by cell culture or encapsulated in extracellular *** composition of the MSC secretome,which represents the set of these secreted cellular products,is crucial for the performance of the desired therapeutic *** cell culture strategies have been employed to adjust the secretome composition of MSCs to obtain the best therapeutic responses for different clinical ***,the manipulation of culture conditions has focused mainly on the use of different biochemical elements for the preconditioning of MSCs and less on the physical conditions of the cell culture ***,we offer our point of view regarding the importance of the physical properties of cell culture substrates and their mechanotransduction responses in preconditioning the MSCs *** highlight the relevance of studying mechanotransduction events associating cell morphology and the modulation of gene expression to customize and expand the use of MSCs secretomes.

Extracellular vesicles(EVs)are nanometric particles that enclose cell-derived bioactive molecules in a lipid bilayer and serve as intercellular communication ***,in various biological contexts,EVs are reported to engage in immune modulation,senescence,and cell proliferation and ***,EVs could be key elements for potential off-the-shelf cell-free *** has been studied regarding EVs derived from human pluripotent stem cells(hPSC-EVs),even though hPSCs offer good opportunities for induction of tissue regeneration and unlimited proliferative *** this review article,we provide an overview of studies using hPSC-EVs,focusing on identifying the conditions in which the cells are cultivated for the isolation of EVs,how they are characterized,and applications already *** topics reported in this article highlight the incipient status of the studies in the field and the significance of hPSC-EVs’prospective applications as PSC-derived cell-free therapy products.

Alkaline phosphatase-positive cells isolated from human hearts have mesenchymal stem cell characteristics

Tissue-specific resident cells have been identified as a promising population of progenitor cells for cell-based therapies. We describe here the isolation from adult human hearts of tissue nonspecific alkaline phosphatase-positive cells (ALPL+ cells) with mesenchymal stem cell (MSC) characteristics. Samples from 24 adult cadaveric donors were obtained from a valve bank. Mean total ischemia time was 21.5 ± 9.1 hours. The success rate for the isolation of human heart-derived cells by the explant culture technique was 70% for the right auricle (14 of 20 trials) and 33% for the right ventricle (7 of 21 trials). The total auricle-derived cell population (TAD) was used for the purification of ALPL+ cells. TAD and ALPL+ cells expressed markers for MSC and pericytes. TAD cells and ALPL+ cells differentiated into adipocytes, osteoblasts and chondroblasts, and ALPL+ cells expressed markers of these three lineages more strongly than TAD cells, as shown by RT-PCR. This population therefore has a greater potential for differentiation into mesechymal lineages than TAD cells. Both cell populations express some markers of cardiac progenitors, such as GATA4, CD117 and VEGF. ALPL+ cells expressed troponin T and ABCG2, which are also markers of the cardiac lineage. Heart samples from tissue banks could be considered as sources of MSC with putative commitment towards cardiac lineages, even after prolonged ischemia times.
